This project had two primary goals: (1) to explore opportunities to further a deep-ocean vehicle’s reach using alternative pressure spheres, and (2) to implement an existing deep-ocean vehicle (lander) in active scientific research.
I gained a greater understanding of the limitations and design choices made for existing pressure spheres using Finite Element Analysis (FEA). My simplified FEA model predicted sphere failure for the existing 30% Fiber Glass 70% Nylon injection molded spheres at an external pressure of 3,954psi or 2,690m ocean-depth (only a 7.38% error compared to the tested minimum failure depth), so I determined it a valid model. In investigations, locating missing persons and clandestine remains are imperative. One way that first responder and police agencies can search for the remains is by using cadaver dogs as biological detectors. Cadaver dogs are typically used due to their olfactory sensitivity and ability to detect low concentrations of volatile organic compounds produced by biological remains. Cadaver dogs are typically chosen for their stamina, agility, and olfactory sensitivity. This study aspires to find a new screening approach to trace DNA recovery techniques to yield a higher quantity of trace DNA from larger items of evidence. It takes the path of visualizing trace DNA on items of evidence with potential DNA so analysts can swab a more localized area rather than attempting to recover trace DNA through the general swabbing technique currently used for trace DNA recovery. The first and second parts consisted of observing trace DNA interaction with Diamond Dye on porous and non-porous surfaces.

Naturally occurring reefs are some of the world’s most biologically diverse ecosystems formed by jagged rocks tucked slightly below sea level. In recent years global warming began to pose a major threat to many reef habitats. Most relevant is the increase in surface seawater temperatures that cause coral to bleach, taking away major food sources for larger marine species. Researchers have combated this by deploying artificial reefs in substitution for naturally formed limestone rock formations in order to promote the expansion of coastal habitats. This project examines the development of a smart boat which could serve as a possible marine research apparatus. The smart boat consists of a miniature vessel containing a low-cost microcontroller to live stream a camera feed, GPS telemetry, and compass data through its own WiFi access point. The smart boat also has the potential for autonomous navigation. My project captivated the interest of several members of California Polytechnic State University, San Luis Obispo’s (Cal Poly SLO) Marine Science Department faculty, who proposed a variety of fascinating and valuable smart boat applications.

The integument plays essential roles in the structural support, protection, and hydrodynamic capability among fishes. Most shark skin research has been done on the external epidermal layer containing the dermal denticles, while the larger dermis layer has been mostly ignored. The dermis layer in sharks is composed of two layers, the upper stratum laxum and the lower stratum compactum, holding supportive collagen and elastin fibers. There may be morphological and compositional differences in the dermis layers across various species of sharks, although the extent of such is unknown. Satellite telemetry tags are used to track the migration patterns of large cetaceans. These tags penetrate the dermis and remain embedded in the underlying blubber tissue. As the dermis of cetaceans is host to a diverse microbiome, and it is impossible to clean the skin before implanting the devices, the potential for infection is increased when the tags penetrate through the skin. H2O2 is a potential antimicrobial agent that, in addition to showing broad-spectrum efficacy against gram-negative and gram-positive bacteria, can promote wound healing outcomes by promoting proliferative factors and peptides that protect against oxidative stress. Throughout the course of this project, our team helped the Cal Poly Biological Sciences department refine an intertidal zone simulator. The aim of this device is to allow any marine biologist to easily simulate a vast range of tidal zones in order to test animal behaviors within these zones. Another goal of this project is to make each simulation tank independent from the others by using a single microcontroller to handle all inputs and outputs of the system as well as logging all relevant data. Freshwater availability is a growing global concern, and desalination is often presented as the solution, but from this important technology comes issues of toxic waste. Ecosystems are delicate areas that contain species adapted to that specific location, and any chemical or physical changes can disrupt the fitness of species. Cladophora glomerata is a filamentous green alga native to the Great Lakes. However, its nuisance growth in phosphorus rich waters negatively affects the lakes’ aesthetic and water quality. The Great Lakes Cladophora Model (GLCM) v1, developed in 1982, was the first mechanistic model to simulate Cladophora growth basing phosphorus availability and environmental conditions followed by Cladophora Growth Model and GLCM v2. In this study, the light and temperature mediation factors for Cladophora net growth are revised as a necessary step prior to the development of a self-shading algorithm. The Marine Gastrobot sponsored by Dr. Christopher Kitts of the Cal Poly Center for Applications in Biotechnology was a research and development effort intended to explore the use of microbial fuel cell technology as a power source for underwater robots. Our team Ocean Locomotion succeeded in developing a first iteration of an underwater robotic platform suitable for microbial fuel cell integration. The primary feature of the design is its sinusoidal fin propulsion intended for benthic exploration with limited risk of entanglement. This report documents the design, fabrication, and testing for a marine biomass analyzer. The goal of the project and constructed system was to determine the validity and efficacy of a process that could flatten benthic macrofauna to a consistent thickness such that a biomass for the collected sample could be accurately determined. The name of this project is called “exPIERience” and the technological field of focus is virtual reality. We will be working on creating a virtual reality experience using the Oculus Rift for the Center of Coastal Marine Sciences (CCMS). Electromagnetic fields (EMF) produced by high voltage (HV), submarine transmission cables leading from offshore wind energy generation facilities could affect foraging or migratory behaviors of electro-receptive fishes, including endangered Atlantic Sturgeon. However, no published studies have quantitatively evaluated the possible behavioral effects of EMF exposure on sturgeon during residence in coastal waters. This study evaluated behavioral responses by sub-adult Atlantic Sturgeon to electromagnetic and magnetic fields under controlled laboratory conditions. The objective of this project was to determine if the electromagnetic field (EMF) emissions from undersea power cables impacted the local and transient marine life, with an emphasis on reef fishes. The work was done at South Florida Ocean Measurement Facility of Naval Surface Warfare Center, Carderock Division, Broward County, Florida. This facility functions as the hub for a range of active undersea detection and data transmission cables. Underwater explosions pose a significant threat to waterborne infrastructure though destructive pressure waves that can travel significant distances through the water. However, the use of bubble screens can attenuate the peak pressure and energy flux created by explosions to safe levels. This study investigates the prediction of pressure wave characteristics based on accumulated data, the damage potential of underwater explosions based on applied loads and effective material strength, and the bubble screen parameters required to prevent damage. The results were compiled to form a procedure for the design and implementation of a bubble screen the protection of waterborne infrastructure.

The goal is to determine if low-cost UV LEDs can mitigate marine biofouling on small glass or acrylic camera lenses. A microprocessor-controlled experimental setup to control the illumination of low-cost UV LEDs of various wavelength and packaging was fashioned. The system consists of a programmed microcontroller, a manufactured LED breakout interface, and a submergible UV LED array enclosed in a borosilicate glass tube. Anthropogenic forcing, such as ocean acidification caused by rising carbon dioxide emissions, and eutrophication due to increased nutrient loadings in run-off, are causing major changes to the biogeochemistry of the oceans. As a consequence, coastal phytoplankton are susceptible to altered biogeochemical environments. This study examined the effect of a lower pH and increased levels of nutrients on the common coastal harmful alga, Heterosigma akashiwo. Growth rates, maximal cell yields, neutral lipid accumulation and toxicity of cells grown under various pH and nutrients regimes were measured. H. akashiwo growth was near maximal when grown at lower pH levels. There was …

An ontogenetic series of nine species; Centrophorus granulosus, Centroscymnus coelolepis, Dalatias licha, Deania calcea, Echinorhinus cookei, Isistius brasiliensis, Oxynotus centrina and Squalus acanthias were studied to determine swimming capabilities, boundary-layer flow conditions and placoid scale functional morphologies. Body morphometric variables included the girth and the distance from snout to the orbitals, origin of the median and paired fins, and the caudal peduncle, body and fin surface area, fin aspect ratios and caudal fin angles.
