Contamination of oysters with a variety of viruses is one key pathway to trigger outbreaks of massive oyster mortality as well as human illnesses, including gastroenteritis and hepatitis. Much effort has gone into examining the fate of viruses in contaminated oysters, yet the current state of knowledge of nonlinear virus-oyster interactions is not comprehensive because most studies have focused on a limited number of processes under a narrow range of experimental conditions. Since 1991, the number of penaeid shrimp occurring in Virginia waters of Chesapeake Bay has steadily increased, prompting an interest in developing a fishery. Although development of a shrimp fishery in the Chesapeake Bay region could bring economic benefits, the fishery may be hampered by the presence of a disease syndrome known as shrimp black gill (sBG). The population structure of the blue crab Callinectes sapidus was examined in the Maryland Coastal Bays (MCB) from 2014 to 2016. Crabs were sampled from April to December of each year. Size–frequency distributions showed a strong seasonal cycle, with small crabs being abundant in April, increasing in size through September, with adult crabs observed in the highest proportions from August through October of each year. A subsample of crabs was assayed for hemolymph ecdysone concentrations to examine molting patterns in field-collected blue crabs. Climate change has resulted in increasing temperature and acidification in marine systems. Rising temperature and acidification act as stressors that negatively affect host barriers to infection, thus enhancing disease processes and influencing the emergence of pathogens in ecologically and commercially important species. Given that crustaceans are ectotherms, changes in temperature dominate their physiological and immunological responses to microbial pathogens and parasites. Because of this, the thermal ranges of several crustacean hosts and their pathogens can be used to project the outcomes of infections. Since 2012, aquacultured eastern oysters Crassostrea virginica have been reported by oyster farmers to display mortality approaching 30%, and in some cases 85%, in areas of the lower Chesapeake Bay, VA. Based on accounts from industry, this mortality has typically affected 1-y-old oysters between May and early July, and has tended to occur in triploid oysters, which represent the vast bulk of production in the area. Phenological mismatchmaladaptive changes in phenology resulting from altered timing of environmental cuesis an increasing concern in many ecological systems, yet its effects on disease are poorly characterized. American lobster (Homarus americanus) is declining at its southern geographic limit. Rising seawater temperatures are associated with seasonal outbreaks of epizootic shell disease (ESD), which peaks in prevalence in the fall. We used a 34-year mark-recapture data set to investigate relationships between temperature, molting phenology, and ESD in Long Island Sound, where temperatures are increasing at 0.4 degrees C per decade. Attempts to decipher the life cycle of Haplosporidium nelsoni began almost immediately after it was identified as the pathogen causing MSX disease in eastern oysters, Crassostrea virginica. But transmission experiments failed and the spore stage, characteristic of haplosporidans, was extremely rare. Researchers concluded that another host was involved: an intermediate host in which part of the life cycle was produced, or-if the oyster was an accidental host-an alternate host that produces infective elements. A later finding that spores were found more often in spat (< 1 y old) than in adults revived the idea of direct transmission between oysters. The new findings and the availability of molecular diagnostics led us to revive life cycle investigations. Over several years, oyster spat were examined for spores and searched for H. nelsoni in potential non-oyster hosts using both histological and polymerase chain reaction (PCR) methodologies. …

Infectious diseases are economically detrimental to aquaculture, and with continued expansion and intensification of aquaculture, the importance of managing infectious diseases will likely increase in the future. Here, we use evolution of virulence theory, along with examples, to identify aquaculture practices that might lead to the evolution of increased pathogen virulence. We identify eight practices common in aquaculture that theory predicts may favor evolution toward higher pathogen virulence. Four are related to intensive aquaculture operations, and four others are related specifically to infectious disease control. Male Gambusia holbrooki (Eastern Mosquitofish) express a heritable pigmentation polymorphism: ≈99% of males are silver, and only ≈1% have a melanic, black-spotted pattern. Sex-linkage, an autosomal modifier, and temperature control the expression of this heritable melanism. In many teleosts, melanin also accumulates around the site of parasitic invasion. We have identified black-spot disease in wild mosquitofish from their native habitat. Here, we demonstrate convergence upon the black-pigmented phenotype through two means: 1) heritable melanism, and 2) melanic spotting on the silver genotype that results from infection with immature encysted trematodes. Bivalve species, especially mussels, are biomass dominants in many deep-sea chemosynthetic ecosystems. As in shallow-water environments, parasites are likely to be important factors in the population dynamics of bivalve communities in chemosynthetic ecosystems, but there has been little study of parasitism in deep-sea seep or vent molluscs. In this study, Parasite types, diversity, prevalence, infection density and non-infectious indicators of stress or disease as related to host age, reproductive condition, and endosymbiont density were assessed in mussels (Bathmodiolus heckerae) from 2 seep sites and mussels (B. puteoserpentis) from 2 vent sites. A pathogenic virus was diagnosed from juvenile Caribbean spiny lobsters Panulirus argus from the Florida Keys. Moribund lobsters had characteristically milky hemolymph that did not clot. Altered hyalinocytes and semigranulocytes, but not granulocytes, were observed with light microscopy. Infected hemocytes had emarginated, condensed chromatin, hypertrophied nuclei and faint eosinophilic Cowdry-type-A inclusions. In some cases, infected cells were observed in soft connective tissues. With electron microscopy, unenveloped, nonoccluded, icosahedral virions (182 +/- 9 nm SD) were diffusely spread around the inner periphery of the nuclear envelope. Virions also occurred in loose aggregates in the cytoplasm or were free in the hemolymph. …

The diseases of blue crabs have received relatively little attention compared to those of the American oyster, Crassostrea virginica, or the penaeid shrimps, Penaeus spp. This is primarily due to differences in resource management (fishery vs. aquaculture), and the magnitude of financial losses suffered by the industries from protozoal diseases in oysters and viral diseases in shrimp, respectively. Nonetheless, several agents including Vibrio spp., Hematodinium perezi, Paramoeba perniciosa, Ameson michaelis and Loxothylacus texanus are highly pathogenic in blue crabs, and have the capacity to severely damage certain segments of the crab population. Blue crabs, Callinectes sapidus, infected with Hematodinium perezi frequently show signs of weakness and lethargy and die when stressed by handling or capture. Radical changes to the hemolymph of heavily infected crabs are obvious by reduced clotting ability, discoloration. and a 50% to 70% decline in total hemocyte density. Few other signs of infection are associated with infections and the resulting mortalities of blue crabs. To assay physiological changes in infected crabs, we measured serum proteins, hemocyanin, serum acid phosphatase, various hemolymph enzymes, hernagglutination activity, and tissue glycogen levels in relation to intensity of infection with H. perezi. Slowly growing, non-pigmented mycobacteria were isolated from striped bass (Morone saxatilis) during an epizootic of mycobacteriosis in the Chesapeake Bay. Growth characteristics, acid-fastness and results of 16S rRNA gene sequencing were consistent with those of the genus Mycobacterium. A unique profile of biochemical reactions was observed among the 21 isolates. A single cluster of eight peaks identified by analysis of mycolic acids (HPLC) resembled those of reference patterns but differed in peak elution times from profiles of reference species of the Mycobacterium tuberculosis complex. A gill-associated Perkinsus sp. isolated from the softshell clam (Myo arenaria) is described as a new species, P. chesapeaki sp. nov. Examination of the parasite in seawater cultures revealed life cycle stages and zoosporulation processes similar to those described for other species of the genus Perkinsus. Prezoosporangia developed thickened cell walls upon contraction of the cytoplasm and development of a distinctive clear area between the cell wall and the protoplast. Successive bipartition of the protoplast led to the formation of hundred's of zoospores within mature sporangia. Teleosts possess mechanism(s) by which they can generate considerable structural diversity within their tetrameric antibody molecules. In salmonids, this diversity is generated through a process of random polymerisation of the constituent monomeric subunits rather than dependency upon isotypic gene diversity. Thus, one gene product can give rise to as many as six different structural forms of immunoglobulin. In contrast to mammals, evidence suggests that this polymerisation process occurs late in the secretory process and not within the endoplasmic reticulum. This assembly process is likely to be important in the generation of teleost antibody functional diversity, thereby potentially simulating isotypy.

Temperature and salinity are two important factors limiting the distribution and abundance of Perkinsus marinus, a protozoan parasite of Eastern oysters (Crassostrea virginica). Results of laboratory studies ale consistent with field observations and clearly demonstrate that P. marinus susceptibility and disease advancement are positively correlated with temperature, salinity and in situ number of infective cells. Laboratory findings also suggest that environmental degradation may enhance the epizootic, although disease caused by P. marinus in oysters is known to be predominantly exacerbated by elevated temperature and salinity. A selective breeding program was implemented to attempt to decrease the disease susceptibility of the eastern oyster, Crassostrea virginica, to Perkinsus marinus. Six oyster strains were spawned and the progeny exposed to Haplosporidium nelsoni (MSX) and P. marinus in the lower Chesapeake Bay. The Chesapeake Bay oyster fishery for Crassostrea virginica (Gmelin) is in a state of continuing decline. Two diseases, Haplosporidium nelsoni and Perkinsus marinus have effectively eliminated oysters from many sections of the Bay. Despite over 30 years of disease activity the native oysters have developed neither tolerance nor absolute resistance to these diseases, and do not exhibit any recovery in disease endemic areas in Virginia. Repletion programs have completely failed to recover to permanent production areas lost to disease. The susceptibility of Pacific oysters, Crassostrea gigas, to the oyster parasite Perkinsus marinus was compared with that of eastern oysters, Crassostrea virginica, in two separate experiments. Experiments were conducted in flow-through seawater systems with quarantined effluent. Oysters were challenged by addition of infective P. marinus. In the first experiment, which used only diploid oysters, 40% of C. gigas became infected with P. marinus after 83 days compared to 100% of C. virginica.
