Assessment Of Genetic Diversity Of The Invasive Callery Pear, Pyrus Calleryana Decne. Using Mitochondrial Microsatellites And Its Molecular Detection Using Lamp, Alina Pokhrel

Masters Theses

Pyrus calleryana Decne. (Callery pear; [PC]) is a popular ornamental tree in the urbanized areas of the United States of America (US), owing to its aesthetical value, with showy white blossoms in early spring and vibrant fall foliage. The tree native to Asia is now becoming one of the most problematic invasive tree species in the eastern US. From its introduction in the early 20th century, PC has been commercially used as rootstocks for propagating fruiting pears, alongside other closely related pear species. Several states are restricting the sale of rootstocks to limit the ongoing spread of PC. As …

A Modular Cloning Toolbox For Chloroplast Metabolic Engineering, Alexander C. Pfotenhauer Aug 2021

A Modular Cloning Toolbox For Chloroplast Metabolic Engineering, Alexander C. Pfotenhauer

Masters Theses

Plant synthetic biology is a rapidly evolving field with new tools constantly emerging to drive innovation. Of particular interest is the application of synthetic biology to chloroplast biotechnology to generate plants capable of producing new metabolites, vaccines, biofuels, and high-value chemicals. Progress made in the assembly of large DNA molecules, composing multiple transcriptional units, has significantly aided in the ability to rapidly construct novel vectors for genetic engineering. In particular, Golden Gate assembly has provided a facile molecular tool for standardized assembly of synthetic genetic elements into larger DNA constructs. In this work a complete modular chloroplast cloning system (MoChlo) …

Quantitative Trait Loci Analysis Of A Ril Soybean Population To Determine Chromosomal Regions Governing Seed Protein, Oil, And Linolenic Acid Content, Ronald E. Moore Aug 2020

Quantitative Trait Loci Analysis Of A Ril Soybean Population To Determine Chromosomal Regions Governing Seed Protein, Oil, And Linolenic Acid Content, Ronald E. Moore

180 RILs (recombinant inbred lines) segregating for protein, oil, and fatty acids were produced from a cross between TN12-4098 and TN13-4303. These lines were grown across three locations spread horizontally across Tennessee: Research Education Center at Milan (RECM), Highland Rim Research and Education Center (HRREC), and East Tennessee Research and Education Center (ETREC) in 2018 and 2019. 21 quantitative trait loci (QTLs) spanning 7 chromosomes were found using WinQTLCart2.5.

Population Structure Of Late Blight (Phytophthora Infestans) In Colombia And Ecuador And Downy Mildew (Peronospora Farinosa F. Sp. Spinaciae) On Spinach In Arizona And California, Rebecca Marie Lyon

Masters Theses

In this study, Phytophthora infestans and Peronospora farinosa f. sp. spinaciae populations were analyzed using single nucleotide polymorphisms (SNPs). In Ecuador and Colombia, Phytophthora infestans causes significant damage to potato and tomato and the epidemiology is known to be highly clonal. Our objective was to measure population structure within the context of this clonal epidemiology using both synonymous and nonsynonymous markers. Candidate SNP sites were selected by comparing the draft genomes of the Ecuadorian isolates EC1-3527 and EC1-3626. Genotypes were assessed directly from infected tissue using a targeted sequencing approach. A total of 54 polymorphic sites were assessed in 93 …
