Effects Of Melatonin On Heartbeat And Possible Identification Of A Melatonin Receptor In Drosophila Melanogaster, Tricia L. Vankirk

Chapter 1 of this manuscript is a literature review that serves as an introduction to the entire dissertation. Chapter 2 examines the effects of the melatonin injection on heart rate and rhythmicity in Drosophila melanogaster Canton-S (wild-type) pupae and pupae bearing a variety of heart mutations. Chapter 3 investigates further the possible mechanisms of melatonin’s ability to increase heart rhythmicity without significantly affecting heart rate. A melatonin antagonist, luzindole; a high-affinity melatonin agonist, 2-iodomelatonin and RNAi techniques are used to identify a possible melatonin receptor in Drosophila melanogaster.

The primary types of Onciderini Thomson, 1860 of the Muséum national d’Histoire naturelle (MNHN), Paris, are catalogued and illustrated. Data on the original combination, current name, gender, and type locality are verifi ed and presented. There are 139 primary types of Onciderini including 38 in Oncideres Lacordaire, 1830; 17 in Hypsioma Audinet-Serville, 1835; 10 in Hesycha Fairmaire and Germain, 1859; nine in Hypselomus Perty, 1832; and eight each in Eudesmus Audinet-Serville, 1835 and Trestonia Buquet, 1859. The toxic effects of older classes of pesticides on natural enemies are typically acute and exposure usually occurs through direct contact with foliar residues. However, older chemistries are being replaced by newer classes of pesticides that can cause sublethal effects in addition to direct mortality. We developed a set of life table response protocols to quantify the effects of multiple routes of exposure to pesticides on individual-level life history parameters of predators and parasitoids. Abstract The genus Chalcasthenes Arrow (Dynastinae: Oryctoderini), a scarab beetle genus endemic to the Solomon Islands, is reviewed. Based on examination of type specimens, the genus Strehlia Frey (Rutelinae: Rutelini: Parastasiina) is a new junior synonym of Chalcasthenes. The historical classification of these genera (either in the subfamily Dynastinae or Rutelinae) and character-based criteria for assigning the taxa to the Dynastinae are provided. Reverse transcriptase-quantitative polymerase chain reaction (RT-qPCR) is a reliable technique for quantifying gene expression across various biological processes, of which requires a set of suited reference genes to normalize the expression data. Coleomegilla maculata (Coleoptera: Coccinellidae), is one of the most extensively used biological control agents in the field to manage arthropod pest species. In this study, expression profiles of 16 housekeeping genes selected from C. maculata were cloned and investigated. The performance of these candidates as endogenous controls under specific experimental conditions was evaluated by dedicated algorithms, including geNorm, Normfinder, BestKeeper, and ΔCt method. Female crickets exhibit positive phonotaxis in response to calling songs with specific carrier frequencies (4-5 kHz), syllable periods (SP; 50-70ms), and other parameters. Among a number of ascending prothoracic auditory neurons identified, only L3 has been proposed to be involved in SP selective phonotaxis. L3 responds with selective decrement to 5 kHz stimulus with attractive SPs. Furthermore, L3 is highly tuned to 16 kHz auditory stimulus, which has been correlated with aversive phonotactic behaviors. L3’s cellular responses to simultaneous 16 kHz and 5 kHz stimuli were examined through extracellular recording, thereby further establishing the connection between L3’s selective decrement and …

Understanding the factors that limit the distribution of species is at the core of ecological and biogeographical research, and is critical if we are to predict the responses of key ecosystem components to ongoing climatic changes. My doctoral research seeks to provide an understanding of how thermal physiology influences species’ distributions and better define the mechanisms underlying geographic variation in biodiversity. By using natural temperature gradients (both elevational and latitudinal) and coupling controlled laboratory experiments with field observations and null modeling approaches, I was able to document the role of inter-specific variation in thermal physiology and, more interesting, inter-population variation …

The biotic potential and reproductive parameters of Spodoptera dolichos (Fabricius, 1794) were evaluated under controlled conditions (25 ± 1°C, 70 ± 10% RH and 14 hour photophase). The longevity, pre-, post- and oviposition periods, fecundity, and fertility of 25 couples were evaluated. The longevity of females (12.9 days) was not significantly different than that of males (12.4 days). The mean durations of the pre-, post- and oviposition periods were 3.0, 0.4 and 10.4 days, respectively. The mean fecundity was 4,086.0 eggs per female and mean fertility was 3,557.8 larvae per female. On average, a female copulated 1.4 times. The biotic …

research assessed the performance of the beet armyworm, Spodoptera exigua, following ad
libero feeding on Medicago truncatula accessions differing in specialized metabolite and
saponin profiles. Insect performance did not directly relate to the foliar saponin levels in the
accessions, which was measured using HPLC mass spectrometry. Accession ESP105 had the lowest foliar saponins overall, yet had similar levels of soyasaponins compared to the other accessions. Conversely, accessions A17, PRT178, and GRC43 had high foliar saponins with particularly high …

The early history of the beetle family Histeridae is still very obscure. In part this results from difficulty resolving phylogenetic relationships at deeper levels (Caterino & Vogler, 2002; McKenna et al., 2015a). But it is also partly a result of a sparse and poorly documented fossil record (Chatzimanolis et al., 2006). Here we describe a new genus and species of fossil histerid from Burmese amber (~99 mya), which helps to address both of these problems. Through global trade, spotted-wing drosophila, Drosophila suzukii (Matsumura) (Diptera: Drosophilidae), a native of Southeast Asia, has spread to at least 3 other continents since 2008. Initial reports of the fly in new regions often are associated with damage in agriculture fields, but the fly may be present in the landscape for years before populations reach a level at which they become a pest in fruit production. In 2012, spotted wing drosophila was reported in blueberry fields in eastern Tennessee, USA, for the first time. An apparent reduction in the performance of neonicotinoid seed treatments in controlling thrips, especially in cotton, has been observed, which has coincided with the increased use of both pre- and post-emergence herbicides to control glyphosate-resistant weeds. The objective of this research was to evaluate the potential interactions of both pre- and post-emergence herbicides on the efficacy of insecticide and fungicide seed treatments in cotton
Aldicarb along with imidacloprid and thiamethoxam seed treatments reduced thrips numbers compared with non-treated seed. However, thiamethoxam did not provide effective control. Some pre-emergence herbicide treatments reduced plant health. While there was evidence that thrips injury …

Insects in the genus Monochamus are medium to large-sized, wood-boring beetles whose primary hosts in the Northern Hemisphere are pine trees. These beetles interact with both conifer hosts and associated insects throughout their life history. Past research has demonstrated that Monochamus are saprophagic, but recent findings show that they may colonize healthy pine trees. To determine if southeastern Monochamus could colonize healthy pines, adult Monochamus were attracted to healthy shortleaf pine trees from May to September, 2014, using host volatiles, Ips bark beetle kairomones, and congeneric pheromones. Endemic arthropods of Arkansas were sampled and their nomenclature and distributions were updated. The Arkansas endemic species list is updated to 121 species, including 16 species of millipedes. A study of the millipedes of Arkansas was undertaken, and resulted in the first checklist and key to all millipede species in the state. 68 species are known from Arkansas, including the genera Cylindroiulus and Polydesmus. The first state records for Ophyiulus pilosus, Cylindroiulus sp., and Ptyoiulus coveanus are reported, and new county records are reported for 16 species. One method being utilized for detection of the invasive emerald ash borer, Agrilus planipennis (EAB) involves monitoring aggregations of the wasp Cerceris fumipennis for the presence of EAB in their collected prey. Currently there is little information published on its nest structure, phenology, and prey fidelity. The management of the soybean aphid Aphis glycines Matsumara is a major challenge to soybean production in the north-central United States. The identification and characterization of the insect predator community has informed integrated pest management strategies by providing insight on predators that can suppress soybean aphid populations. Hoverflies (Diptera: Syrphidae) are known predators of A. glycines, but more information is needed on their diversity, abundance, and performance to evaluate their importance as biological control agents of A. glycines. An effective management program for stink bugs (SBs) in pecan groves requires knowledge of: stages of the pecans susceptible to SB damage; strata of the tree with SB damaged nuts; a practical SB monitoring method; and, effects of landscapes contributing SBs into pecan groves.
Stink bugs produced feeding punctures in pecan shucks at all phenological stages. Pecans punctured before the dough stage drop from the tree. Kernel damage occurs in the pecans’ dough stage, whereas mature pecans are not damaged.
The pecans collected from the whole tree using the tree shaker had significantly less punctures than the pecans collected from …

The Interior Highlands is a biodiversity hotspot, with at least 200 known endemic species, but is understudied compared to hotspots, such as the Southern Appalachians. In order to begin to rectify this issue, a nine month study was conducted from mid-March through early December at a 4 ha site at Steel Creek, Buffalo National River, in Newton County, Arkansas. Thirteen collecting methods were employed, including three colors of Lindgren funnel trap, five colors of pan trap, Malaise traps, canopy traps with upper and lower collectors, pitfall traps, and Berlese-Tullgren extraction of leaf litter, which resulted in the collection of 1311 …

Thirteen new species of Testudacarus (Torrenticolidae: Testudacarinae) are described, four species are redescribed, and the status of previously problematic species are addressed. For Testudacarinae this represents the first published: 1) descriptions from multiple specimens (therefore providing ranges); 2) colored photographs; 3) explicit illustrations and discussion of sexual dimorphism within the subfamily; 4) genetic data. A comprehensive literature review is also included.

A challenge faced by organisms with complex life cycles is how environmental factors experienced at an earlier life stage affect the fitness at that stage or are carried over to subsequent life stages. I used container-inhabiting mosquitoes: Aedes albopictus, Aedes aegypti, Aedes triseriatus and Culex quinquefasciatus to study the interactions and performance of life history stages under specific environmental factors. I investigated the effects of egg-desiccation tolerance on egg viability and larval performance in the Aedes mosquitoes. The American chestnut, Castanea dentata, was once one of the most useful and abundant canopy trees in eastern North American forests. Over the last 200 years, the species has been decimated by two exotic pathogens, Phytophthora cinnamomi and Cryphonectria parasitica, killing millions of trees and reducing surviving Castanea dentata to short-lived sprouts. Cryphonectria parasitica-resistance breeding programs are currently producing advanced backcross generations, which are being compared with pure American chestnut in field tests of growth performance and Cryphonectria parasitica resistance. The family Thaumaleidae, also known as seepage midges, is an uncommonly encountered, understudied family of aquatic Diptera compared to its sister group, the Simuliidae. The goal of this project was to assess species diversity of the Nearctic Thaumaleidae and determine relationships among them. The western Nearctic fauna of Androprosopa Mik is revised to include twenty-eight species, six of which are described as new to science. 1. The success of invasive species is often thought to be due to release from natural enemies. This hypothesis relies on the assumption that species are regulated by top-down forces in their native range and implies that species are likely to be regulated by bottom-up forces in the invasive range. Pyrethroid insecticides have been used to control larvae or adults of the western corn rootworm (WCR), Diabrotica virgifera virgifera LeConte, a key pest of field corn in the United States. In response to reports of reduced efficacy of pyrethroids in WCR management programs in southwestern areas of Nebraska and Kansas the present research was designed to establish a baseline of susceptibility to the pyrethroid insecticide, bifenthrin, using susceptible laboratory populations and to compare this baseline with susceptibility of field populations. As revealed in a previous microarray study to identify genes regulated by 20-hydroxyecdysone (20E) and juvenile hormone (JH) in the silkworm, Bombyx mori, E93 expression in the fat body was markedly low prior to the wandering stage but abundant during larval-pupal metamorphosis. Induced by 20E and suppressed by JH, E93 expression follows this developmental profile in multiple silkworm alleles. The reduction of E93 expression by RNAi disrupted 20E signaling and the 20E-induced autophagy, caspase activity, and cell dissociation in the fat body. Reducing E93 expression also decreased the expression of the 20E-induced pupal-specific cuticle protein genes and prevented growth …

RNA interference, or RNAi, refers to a set of biological processes that make use of conserved cellular machinery to silence genes. Although there are several variations in the source and mechanism, they are all triggered by double stranded RNA (dsRNA) which is processed by a protein complex into small, single stranded RNA, referred to as small interfering RNAs (siRNA) with complementarity to sequences in genes targeted for silencing.
