ABSTRACT

RNA interference (RNAi) is a pervasive gene regulatory mechanism in eukaryotes based on the action of multiple classes of small RNA (sRNA). Exploiting RNAi pathways in non-model systems have great potential for creating potent RNAi technologies. Here, we accessed RNAi-mediated control of gene expression in the two-spotted spider mite, Tetranychus urticae (T. urticae) using engineered dsRNA designed to modulate the host RNAi pathway and increase RNAi efficacy. Analysis of Dicer (Dcr) generated fragments revealed how exogenous RNAs access the host RNAi pathway in this animal, opening avenues for designing RNAi technology for their control. Arctica islandica (ocean quahog) is the longest-lived bivalve on Earth. Individuals on the deep continental shelf of the Mid-Atlantic (US) can survive for centuries, and when found in the colder, boreal waters of Iceland, ages over 500 years can be reached. The ocean quahog is important in the US, yet very little is known about the resiliency of the ocean quahog stock to fishing activity, and ocean quahog recruitment patterns over time. As human populations rise, the level of man-made noise increases globally. Naval sonar and boat traffic are underwater sound sources of particular concern to marine mammal welfare. To better understand the impact of these noise increases on cetaceans, studies can explore animals’ behavioral changes in response to noise. Studies have investigated the ‘dose-response’ relationship between the received sound pressure level of sonar signals and the behavior of cetaceans in the wild, but exposure studies in controlled environments are limited. Natural and anthropogenic changes resulting from altered hydrology, hurricanes, variable precipitation, and the BP oil spill have all taken their toll on oyster reefs in Mississippi. In response, oyster reef restoration efforts are currently underway within the Northern Gulf of Mexico. In order to understand why these efforts succeed or fail, it is crucial to consider predator-prey relationships within the context of the trophic dynamics of oyster reefs. Leatherback turtles (Dermochelys coriacea) are a wide-ranging, oceanic species that feed exclusively on gelatinous zooplankton. Leatherback have been spotted in the Gulf of Mexico (GoM) for several decades and consistently had a high level of interactions with longline fisheries. However, no quantitative studies have been performed to address the spatiotemporal distribution of these turtles in the GoM. Seagrasses are important coastal resources facing numerous stressors, and losses have been documented from local to global assessments. Under the broad theme of habitat loss and fragmentation, a study of historical change in total area and landscape configuration of seagrasses in the Mississippi and Chandeleur Sounds was conducted. Mapping data was collated from a multitude of previous projects from 1940 to 2011.

Humpback whales (Megaptera novaeangliae) belonging to the “breeding G-stock” annually migrate from the Antarctic Peninsula and southern Chile to the southeastern Pacific to reproduce. Associations between mother-calf pairs and escorts were examined in the Gulf of Tribugá, northern Colombian Pacific, using photo-identification and behavioral/spatial sampling. Research hypotheses included: 1. The association between cows and escorts is short-lived, consistent with a male reproductive strategy, 2. While the scientific community is in consensus that coastal systems are threatened by climate change, few climate change studies test the effects of more than one variable directly related to climate change. The dissolved oxygen (DO) levels of the ocean are currently subject to both global warming and eutrophication; 94% of all hypoxia zones are expected to experience >2°C increase by 2035. This dissertation aims to examine how a model organism responds to simultaneous thermal and DO stress involving four levels of DO (100%, 70%, 50%, and 20%) saturation and three temperatures (15°C, 20°C, and 25°C).

The Pearson and Rosenberg (P-R) conceptual model of macrobenthic succession was used to assess the impact of hypoxia (dissolved oxygen [DO] ≤ 2 mg/L) on the macrobenthic community on the continental shelf of northern Gulf of Mexico for the first time. The model uses a stress-response relationship between environmental parameters and the macrobenthic community to determine the ecological condition of the benthic habitat. The ecological significance of dissolved oxygen in a benthic habitat is well understood. In addition, the annual recurrence of bottom-water hypoxia on the Louisiana/Texas shelf during summer months is well documented.

Estuaries are dynamic regions in which there can be significant modification of the riverine flux of trace elements to the open ocean due to various geochemical, physical, and biological processes. Additionally, estuaries are often subject to anthropogenic inputs of trace elements. The first portion of this study investigated the source, behavior, and sediment interaction of anthropogenic stable cesium (Cs) in St. Louis Bay (SLB), MS. A consistent increase in stable Cs concentration was noticed in sediment cores starting from a period when a titanium dioxide refinery on SLB started operations. Selected trace elements (TEs), dissolved organic carbon, and nutrients were studied in Louisiana Shelf waters including the Mississippi (MR) and Atchafalaya (AR) River plumes during periods of high, intermediate, and low river discharges. Seasonal variations in TEs were observed at low salinity, reflecting seasonal changes in the river water endmembers. Shelf surface water dissolved Mo, Cs, U, Ni, and Cu showed conservative behavior with minor scattering in some high salinity waters. Marine fungi are an important but often overlooked component of marine ecosystems. Primarily saprotrophic, they are vital to coastal nutrient cycling processes and food webs. However, basic marine fungal distribution data are lacking in many parts of the world, as is knowledge of the sampling intensity required to characterize the biodiversity of these communities. Vibrio parahaemolyticus (Vp) is a gram-negative bacterium found naturally in marine and estuarine environments. Vp is found in oysters including those which are later consumed by the public. Sub-populations of potentially virulent Vp contain specific virulence factors and are relevant human pathogens capable of causing gastroenteritis, wound infection, and death. The tdh and trh genes, both encoding hemolysins, have been correlated with the majority of clinical Vp isolates but have not been shown to be the definitive virulence factors.

Effects Of Hypoxia And 4-Tert-Octylphenol On Gene Expression Profiles Of The Sheepshead Minnow (Cyprinodon Variegatus), Arthur Alan Karels

Hypoxia occurs in estuaries of northern Gulf of Mexico and world-wide, with increasing frequency/severity via eutrophication and anthropogenic influences. Hypoxia inducible factors (HIFs) form transcriptional complex and bind DNA at hypoxia responsive elements (HREs) in promoter regions of genes needed for systemic and cellular adaptation of fish to low dissolved oxygen (hypoxia, DO <2.0 mg/ml). Hypoxia-induced activation of HIF-αs can lead to a cascade of downstream activation, such as erythropoietin (EPO). Return to normal DO levels (normoxia), prolyl hydroxylases (PHDs) are activated to degrade HIF-αs back to baseline. Fish are affected by environmental estrogen mimics, like 4-tert-octylphenol (4tOP), binding estrogen receptor alpha (ERα) at estrogen responsive elements (EREs) and activating genes vitellogenin (VTG). Previous research showed overlap or crosstalk between these two mechanistic pathways. Hypoxia triggers unknown factors regulating ERE-mediated ERα signaling pathway, and stressor combinations could increase/decrease hypoxic or endocrine pathway. Research examined molecular/physiological effects of hypoxia (acute and chronic, moderate and severe) and 4tOP (~60μg/L)on adult male and/or female sheepshead minnow (Cyprinodon variegatus). Three genes identified, cloned, and sequenced (HIF-1α, HIF-2α, and PHD3), plus previously identified genes EPO and VTG, were examined in liver/testes exposed to hypoxia and/or 4tOP for cellular/physiological changes. Endpoints examined included mRNA expression from real-time PCR of HIF-1α, HIF-2α, PHD3, EPO, and VTG using cDNA from total RNA extracts, and microarray analyses of genes expressed during the transition from hypoxia back to normoxia. Phylogenetic analyses confirmed isolation of two HIF-α isofoms (HIF-1α and HIF-2α) and the PHD3 isoform. Significant up-regulation of PHD3 occurred within 10 hrs of chronic hypoxia, and persisted when severe (1.5 mg/L) and declined when moderate (~2.5mg/L). Significant up-regulation of HIF-1α and EPO occurred within 30 minutes to 2 hours of onset of acute severe and very severe (~1.08mg/L) hypoxia. Hypoxia acted similar to an estrogen mimic, with huge up-regulation of VTG gene expression in males, and increased VTG levels (additive effect) when hypoxia was combined with 4tOP. Microarray analyses showed 125 genes with significant transcriptional change, with up- or down-regulation from transitions of: (1) hypoxia (72 hrs) to normoxia (74 hrs) and (2) hypoxia+4tOP (72 hrs) to normoxia+4tOP (74 hrs).

The course of the ancestral Tennessee River has been debated in both the geological and biological literature for over 100 years. Several of the proposed courses for the ancestral Tennessee place its course across the state of Mississippi. Geochemical analysis of sedimentary rocks in the Pascagoula River basin supports these hypotheses suggesting that the rocks in the Pascagoula basin were derived from the Highland Rim of Tennessee and northern Alabama, while geochemical analysis of rocks from the Pearl River basin point to deposition from a mixture of sources including the ancestral Mississippi River and perhaps the ancestral Tennessee.
