Bisphenol A (BPA) is one the most widely used plasticizing compounds. As an endocrine disruptor, BPA could affect ovarian function and embryonic development. Bisphenol S (BPS) has been used in many plastics and resins in lieu of BPA. However, the effects of BPS are not widely known. Apoptosis is a type of genetically programmed cell death that is important in ovarian homeostasis. One of the genes that regulates apoptosis, p53, has been shown to respond to estrogen in maintaining ovarian homeostasis. Phosphatase of Regenerating Liver (PRL) proteins regulate a number of important cellular processes, including cell growth and division. Humans have three PRL proteins: PRL-1, PRL-2, and PRL-3. An accumulation of evidence has shown that elevated levels of PRLs are strongly correlated with uncontrollable growth and metastasis of tumors. However, contradictory findings have arisen indicating that PRLs instead function to halt cell division thereby preventing uncontrollable tumor growth. In light of these results, the underlying mechanisms regarding how PRLs function within cellular processes remains unclear. Aberrant expression of members of the phosphatase of regenerating liver (PRL) family has been implicated as a key factor in the progression of several forms of human cancers. However, despite a wide range of studies supporting the role of the enzyme PRL as an oncogene, it has also been identified as a growth suppressor when tested under different conditions. One proposed explanation for this change in function is that redox regulation controls the accessibility of the active site of PRLs, which is necessary for oncogenic output. The genes bub1 and cdc20 are involved in regulation of meiotic arrest during maturation of the oocyte (immature egg). Meiotic arrest is important in oogenesis, the development of fertilizable eggs, and allows for identification of any DNA mutations or chromosome assembly errors, thus preventing damaged eggs from being ovulated. This study has examined the expression of spindle assembly checkpoint genes bub1 and cdc20. Oocytes were treated with DHP (hormone to induce maturation) or by nocodazole, a spindle destabilizing agent, and relative amounts of mRNA were measured using RT-PCR.
