The purpose of this project was to study the edaphic and topographic characteristics of the 13 vernal pools on the Santa Rosa Plateau Preserve, thus documenting the role soil texture plays in vernal pool formation.

Several field and laboratory procedures were used to demonstrate that there is a higher percentage of clay present within vernal pools than in soils outside the margin of the pools, for 11 out of the 13 pools. Statistical tests showed that the percentage of clay was highly significant within the center soil of the pool as compared to the soil outside the margin of the …

The Effect Of Cold Acclimation On The Temperature Preference Of The Goldfish, Carassius Auratus, And The Brown Bullhead, Ictalurus Nebulosus, Alfred Lord

Two species of fish, Carassius auratus and Ictalurus nebulosus, were subjected to cold acclimation regimes. Acclimation temperatures were slowly lowered to 3°C, then held for a period of time. At various times during this regime, fish were taken out and allowed to spend time in a temperature gradient to determine their preferred temperatures. Carassius were left in the gradient just long enough to determine a measure of the acute temperature preference, while Ictalurus were left in the temperature gradient for longer periods of time to observe any changes that might occur as the fish adjusted to selected temperatures. A study of the age and growth of black crappie, Pomoxis nigromaculatus, was conducted in Lake Anna, Virginia, a cooling impoundment for Virginia Power's North Anna Power Station. Growth of the population in the reservoir was compared to that of the population in North Anna's Waste Heat Treatment Facility. Analysis of covariance (ANCOVA) using age as the covariate indicated the growth rate of the two populations did not differ significantly at the .05 level. The populations were combined and tests were conducted to determine differences in growth rates between the sexes. ANCOVA indicated the growth rates of males and females …

Morphological And Biochemical-Based Phylogenetic Analyses Of Opuntiae Prickly Pear Cacti Of Southern California, Lyla S. Richards

Phylogenetic analyses are used to suggest phylogenetic relationships for series Opuntiae (genus Opuntia, subgenus Opuntia) present in Southern California. The bases of the analyses were electrophoretic characters utilizing allelic band data obtained from electrophoretic assays conducted in this study and morphological and chromatographical characters compiled from data in existing literature. Cladograms produced from the different data bases yielded similar relationships. The electrophoretic-based cladogram provided the strongest support for Benson's (1982) classification with the o. littoralis complex represented as a monophyletic group, unlike the morphological and chromatographic-based cladograms. Placements of hybrids "Occidentalis C" and "Occidentalis D" were also considered.

Goldfish (Carassius auratus) (40-80g) were injected with dopamine into the forebrain to study the possible involvement of this amine in central temperature regulation in these fish. Dopamine caused a decrease in selected temperature after injection into the rostral nucleus preopticus periventricularis (NPP). This effect was dependent on the dose of dopamine administered. Doses of 25, 50, 100 and 250 ng were used, injected in a volume of 0.2μ1. Injections in regions adjacent to the NPP elicited hypothermic effects only at the higher dosages. These effects were not consistent. Injections in caudal regions of the NPP elicited no effect. …

The existence of a sodium-ion-for-hydrogen-ion exchange mechanism in the membranes of certain epithelial cells has been demonstrated by many investigators (Murer et al., 1976; Tsai et al., 1984; Seiler et al., 1985 ) • This mechanism has been shown to exchange one Na+ for one H+ (Aronson & Igarashi, 1986) and is thought to be more prominent in the brush-border membranes (Murer et al., 197 6; Reenstra et al., 1981 ; Ives et al., 1983) of mammalian kidney tubules and intestinal tissues than in basolateral membranes of these tissues. The study was designed to investigate the hypothesis that various environmental stimuli would elicit specific individual responses, as reflected in autonomic patterns. It then became the aim of the study to see whether the stimuli which controlled the physiological pattern would also control the verbal response. Three female and two male students volunteered for the single-subject experiment, where they were exposed to a 15-slide sequence of environment stimuli. Results indicated that when allowed to verbally respond without rating restrictions, responses varied greatly, limiting the possibilities of comparison. The relative contribution of arteriovenous oxygen (A-V O2) difference to thermally-induced and activity-induced changes in metabolic rate (VO2) were delineated in the toad, B. marinus. In conjunction, the influence of rest and activity, temperature, and removal of the pericardial sac on separation efficiency (Es) in the anuran ventricle was determined by direct measurement of arterial and venous percent oxygen saturation (% saturation), using a microsampling technique.

There has been heightened interest in the biology of striped bass (Morone saxatilis) because of increased pollution in their native spawning grounds and because of their extensive use in landlocked sport fisheries. Their euryhalinity makes them an excellent species for osmoregulation studies. The objective of this research was to study the rate of adaptation of striped bass gills to sea water (3% salt) after direct transfer from freshwater using biochemical (ion transport enzyme levels), physiological (chloride efflux), and ultrastructural methods. Populations of the freshwater mussel Anodonta imbecillis Say 1829, were sampled from six ponds at the Clifton F. McClintic Public Hunting and Fishing Area in Mason County West Virginia from April 1986 to May 1987. One of the six ponds, pond 12, is contaminated with byproducts from the manufacture of TNT which occurred at the site during World War II. Population density, length frequency distribution, growth rates and condition indices were examined for each population to establish baseline data for the species and to determine what effects the contaminants might be having on these organisms.

A yearlong hormonal study of immature populations of loggerhead turtles (Caretta caretta) and green turtles (Chelonia mydas) inhabiting the Indian River in Central Florida, USA, provided information about the relationship between serum testosterone levels and life history habits. By use of a specific radioimmunoassay, testosterone levels of both species were found to remain constant throughout the year with only a slight increase in the warmer months. Adrenal production of steroids in response to stress does not contribute significantly to overall testosterone levels may be reduced for turtles held in captivity. The influence of iron nutrition on patterns of photosynthetic behavior, nitrogen metabolism, and fixed-carbon allocation is reported for a common freshwater green alga. Cultures of Scenedesmus quadricauda were grown in Fraquil medium in which iron concentrations ranged from 1.0 μM to 0.01 μM (log 10-6 to 10-8 M, respectively). Carbon 14 and nitrogen uptake experiments were conducted at photosynthetically saturating and subsaturating photon flux densities.

Cannulae were implanted into forebrain loci of goldfish (Carassius auratus; 45-90 g) to determine (i) the effects and site of action of intracranial norepinephrine (NE) injections on behavioral thermoregulation and (ii) the mechanism and the types of adrenoreceptors involved in the thermoregulatory effect of NE. After 30 min in a thermal gradient, implanted fish were injected with norepinephrinebitartrate salt (2.5-500 ng NE) in a total volume of 0.2 ul (carrier was 0.7% NaCl). Microsurgical fertilization is the technique in which a spermatozoon or sperm nucleus is injected into the cytoplasm of an egg. To establish a foundation for the use of microsurgical fertilization as a means of treating infertility, this study evaluated the procedure in hamster and human eggs. Hamster sperm nuclei were microinjected into hamster eggs to determine the rate of abnormal fertilization and to ultrastructurally assess cellular damage by transmission electron microscopy. Arachnids were collected in three plant communities on the University of Central Florida campus, beginning in May of 1983 and ending in March 1984. These sites were undisturbed communities: pond pine, sand pine scrub, and flatwoods. Pitfall traps and sweep nets were used to catch arachnids every other month.

A total of 2,657 arachnids were collected in pitfall traps and 4,022 in sweep nets. Spiders were the most numerous group, consisting of 87.5% of the individuals in pitfall traps and 100.0% in sweep nets. Forty-two scorpions, 41 pseudoscorpions, and 248 opilionids were captured on the ground surface.

A Systematic Revision Of Atlantic Tonguefishes (Symphurus: Cynoglossidae: Pleuronectiformes) With A Preliminary Hypothesis Of Species Group Relationships, Thomas A. Munroe

Four of 29 previously described nominal species of Symphurus are junior synonyms; one sub-species (S. p. plagusia) is accorded full species status (S. tessellatus); two undescribed dwarf species are present in the eastern Atlantic; three new species were found among western Atlantic material; and several Atlantic forms are represented in the eastern Pacific by cognate species. Detailed and expanded descriptions, differential diagnoses, artificial keys and updated ecological information are provided for 29 species occurring in the Atlantic Ocean. Various stages of spermatogenesis in pupal testes of the mosquito Eretmapodites quinquevittatus (Theobald) were determined by 2% 1acto-aceto-orcein and silver nitrate staining techniques. Temporal relationships of stages were demonstrated and behavior and appearance of meiotic chromosomes were described.

Pupal testes were most active meiotically for the first 12-14 hours after onset of pupation. The prepachytene stage of leptotene and a nucleolus were observed with the use of silver nitrate staining. Experimental observations demonstrated the first spermatids and visible mature sperm at 6-8 and 18 hours, respectively, after onset on pupation. Isolation And Characterization Of Vinclozolin-Resistant Mutants Of Neurospora Crassa, Kimberly Booth Thurmon

Six vinclozolin-resistant mutants of N. crassa obtained following ultraviolet irradiation and selection on vinclozolin media, showed 1:1 segregation of the vinclozolin-resistant trait. In addition to being vinclozolin resistant, the six mutants had altered, non-wild-type morphology and were osmotic sensitive, with the different mutants exhibiting varying degrees of sensitivity. Genetic analysis indicated linkage of the three traits. Reappearance of osmotic sensitivity/altered morphology in crosses of phenotypically recombinant F3 progeny supported the presence of modifier(s). Rates of growth on vinclozolin media placed the six mutants into three groups. In most cases the rates were comparable to rates of growth on W-M …
