Tobacco SABP2 has been shown to display high affinity for salicylic acid (SA) and methylsalicylate (MeSA) and plays an important role in SAR signal development. Using biochemical approach, SABP2 has been shown to demonstrate strong esterase activity in converting MeSA to SA. Recent study shows that tetra fluoroacetophenone, a synthetic analog of SA, competitively inhibits SABP2 esterase activity as well as suppresses SAR signal development in tobacco mosaic virus (TMV)-infected tobacco plants. The objective of this research was the evaluation and forensic validation of Denaturing High-Performance Liquid Chromatography (DHPLC) as a sequencingindependent means of detecting the presence of sequence differences in pair-wise mixtures of non-concordant amplicons of human mitochondrial DNA (mtDNA). The reproducibility and efficacy of DHPLC results, including amplification reproducibility, injection reproducibility, and column-to-column reproducibility were measured, showing negligible assay-to-assay variability. In addition, cross-contamination on the DHPLC columns demonstrated very low level DNA carryover between a high-abundance sample and subsequent zero-volume injections.

Salicylic acid binding protein 2 (SABP2) is one of the key enzymes in salicylic acid-dependent plant defense pathway. SABP2 is a 29 kDa protein present in extremely low abundance in plants and it catalyzes the conversion of signaling molecule methyl salicylate into salicylic acid. Although it has been shown that 6x His-tagged SABP2 over expressed in E. coli is a homodimer, its exact conformation in planta is still unknown. Inhibitors of cAMP phosphodiestereses (PDEs) promoted an increase in intracellular [cAMP]c and an influx of Ca2+, in both MMQ pituitary cells and α-Pancreatic cells. The specific targets for PDEs and the domains of PDEs are still unknown. Pituitary MMQ cells were used as a model to test modulation of calcium influx through the L-type calcium channel with PDE inhibitors. cAMP analogs were used to determine cAMP molecular targets. 8-cpt-cAMP initiated an Epac mediated pathway and had strong effects on calcium influx in the cell; suggesting, a relatively strong cAMP target. This study used indirect methods to estimate patterns of gene flow in a rare salamander species, the striped newt (Notophthalmus perstriatus). First, we used combined genetic and ecological methods to determine whether populations that appear to exist in two regions separated by 125 km, exhibited genetic and ecological distinctness such that the regions demarcate separate conservation units. Using mtDNA (cyt-b), we found that haplotypes were shared between localities within each region but none were shared between regions. Individuals sensitive to domestic cat allergen Fel d 1 experience a variety of symptoms including eye irritation, respiratory irritation, asthma, and severe respiratory distress. Fel d 1 is a protein produced in the saliva and on the skin of domestic cats. Previous studies have demonstrated that Fel d 1 adheres to clothing, upholstery, and human hair and has been found in non-cat environments in levels high enough to cause allergic reactions in sensitive individuals. Predicting the locations of new biological invasions has become a high priority for biologists as well as trying to predict if newly introduced species will become damaging to native ecosystems. Reefs of the eastern oyster Crassostrea virginica in Mosquito Lagoon, Florida have been highly disturbed in recent years resulting in dead reefs (piles of dead, disarticulated shells) some of which have been restored. Understanding of mechanisms that limit the abundance and distribution of species is central to ecology. The failure of mechanisms to regulate populations can result in population outbreaks. There have been two outbreaks of house mice in the past decade in central Florida. In my study, I examine the efficacy of landscape management in the form of mowing and plowed soil barriers to limit or prevent outbreaks of house mice in a former agricultural area. Many tropical aquatic environments worldwide are characterized by intermittent or prolonged hypoxia (low dissolved oxygen). Nevertheless, many tropical freshwater fishes are able to inhabit these challenging environments via a range of morphological, physiological and behavioral adaptations. These RyRY522S/+ knock-in mutant mice provide an excellent tool to investigate calcium dysregulation, its pathological consequences, and potential therapeutic approaches. Fragile X syndrome (FXS) is the most common form of inherited mental retardation in humans. FXS is caused by loss of the Fragile X Mental Retardation Protein (FMRP), an important regulator of neuronal mRNA translation. Patients with FXS display cognitive deficits including memory problems. Protein synthesis-dependent long-term changes in synaptic plasticity are involved in the establishment and maintenance of long-term memory. The primary goal of this thesis was to determine if spaced synaptic stimulation induced the differential expression of microRNAs (miRNAs) in the Drosophila melanogaster central nervous system (CNS). Prior to attaining this goal, we needed to identify and validate a spaced stimulation paradigm that could induce the formation of new synaptic growth at a model synapse, the larval neuromuscular junction (NMJ). Convergent extension is a highly conserved process among mammals, in which the tissue narrows in one axis, and extends across another. Tissue elongation is directed by the regulation of cell interface behaviors, which guides cell intercalation and rosette formation. Rosette formation occurs through the contraction of vertically oriented cell interfaces, and the subsequent elongation of new horizontal interfaces. Factors that affect swimming performance in fish can also affect fitness. I tested the effects of time of day, season and turbidity on swimming performance in black crappie Pomoxis nigromaculatus and white crappie Pomoxis annularis. These closely related fish co-occur and their relative abundance is thought to be governed by the levels of turbidity in the form of suspended sediments. Black crappie predominate in clear bodies of water and exhibited significantly greater critical swimming speeds in clear water than did white crappie during spring days and winter nights. Invasive species are detrimental to native biotas worldwide. Recently, Florida was invaded by a freshwater snail (Pomacea canaliculata) native to Argentina. This snail is a serious pest of rice crops in Asia, but little is known about its interactions within Florida ecosystems. Possible competitive exclusion of the native congener (P. paludosa) is a concern because it is the almost exclusive prey of the federally endangered Everglades snail kite (Rostrhamus sociabilis). CXCL16 is an interferon-inducible chemokine of the CXC-subfamily and functions as an adhesion molecule, when membrane bound, and a chemoattractant when soluble. Upregulation of cell associated CXCL16 (cCXCL16) in colorectal cancer is associated with increased tumor infiltrating lymphocytes and good prognosis. ADAM10 and ADAM17 are metalloproteinases responsible for cleaving CXCL16, releasing soluble CXCL16 (sCXCL16) and contributing to proliferation and migration of mesangial cells, in kidney inflammatory disease. We hypothesize that cCXCL16 is a substrate for ADAM10 and ADAM17 cleavage in colorectal cancer, releasing sCXCL16, which mediates cell proliferation. Allocation of assimilated carbon amongst plant metabolic processes and tissues is important to understanding ecosystem carbon cycles. Due to the range of spatio-temporal scales and complex process interactions involved, direct measurements of allocation in natural environments are logistically difficult. Modeling approaches provide tools to examine these patterns by integrating finer scale process measurements. The marine microalgae Nannochloropsis spp. are of both ecological and economic importance as they contribute to the base of the food chain of marine systems, and can accumulate a considerable amount of lipids. Their lipids can be grouped into two categories, storage lipids, which are mainly triglycerides (TGs), and structural lipids, which are composed of polyunsaturated fatty acids (PUFAs). PUFAs are essential nutrients for both aquatic animals and humans, while TGs can be transesterified to produce biodiesel. Constructed wetlands are used as a best management practice for sequestration of nutrients from agricultural runoff. This dissertation focuses on the influence of vegetation on bacterial community structure, microbial enzyme activities, and phosphorus retention capacity of constructed wetland systems. A greenhouse experiment was conducted using unvegetated or vegetated mesocosms with Juncus effusus, Carex lurida or Dichanthelium acuminatum var. acuminatum in monoculture or mixed culture.

Understanding the factors influencing mate choice is a major focus of sexual selection. Many factors are potentially involved, including age of the individual. The good genes model and the youth advantage model both make predictions about the effect of age on mate choice. Under the good genes model older mates would be the more preferable due to their proven high survivability. The “youth advantage” model, predicts that young to intermediate age males would be more advantageous as mates because of a decrease in sperm quality and the possibility of increased germ-line mutations in older animals.
