Billfishes (marlins, spearfishes, sailfishes and swordfish) are one of the fastest and largest marine apex predators, and perhaps their most recognizable attribute is their bill or rostrum. The proposed function for this novel structure has ranged from hydrodynamic enhancement to defensive weaponry. However, the most supported hypothesis for its function has been linked to feeding. Billfishes have been observed to subdue their prey with their rostrum, either stunning or cutting them into pieces before ingestion. Due to their large body sizes and pelagic lifestyles a thorough investigation of the function of this structure has been logistically challenging. The goal of …

Background: Sperm contain highly compact nuclei, inhibiting DNA extraction using traditional techniques. Current methods extracting sperm DNA involve lengthy lysis and no means of stabilizing DNA, hindering clinical research.
Objective: We sought to optimize an efficient method of extracting high quality human sperm DNA.
Methods: Sperm from three volunteers were isolated using PureCeption. We tested 1) proteinase K with DNA/RNA Shield, 2) DTT and TCEP as reducing agents, 3) QIAshredder homogenization, and 4) stability of sperm DNA fresh (baseline) or after 4 weeks of storage at 4OC in DNA/RNA Shield using modified Quick-gDNA MiniPrep. Plethysmographic Variability Index (PVI) is a derivative index of pulse oximetry that allows evaluating an individual's intravascular volume status. Perfusion Index (PI) represents the strength of pulse signal at the anatomic site of measurement from which PVI is calculated using changes in PI over respiratory cycles. Plethysmographic Variability Index has been used to detect hypovolemia and predict fluid responsiveness in mechanically ventilated human patients however, fewer studies are available in spontaneously breathing patients. Human umbilical cord blood (UCB) progenitor cells with high aldehyde dehydrogenase activity (ALDHhi), can stimulate endogenous islet regeneration after transplantation into mice with steptozotocin (STZ)-induced diabetes. However, UCB ALDHhi cell are extremely rare, and expansion will be required to develop cell-mediated strategies to treat patients with diabetes. To increase the number of progenitor cells available for clinical application, we expanded ALDHhi UCB cells under clinically applicable, serum-free hematopoietic-restricted conditions. 6 day expansion resulted in a 15-fold increase in total cell number, and a 3-fold increase in the number of HPC retaining high ALDH (ALDHhi HPC) …

Tradeoffs in life history evolution result from conflicts in the time and energy that can be simultaneously invested in activities such as growth, reproduction, and survival. Reproduction is an energetically costly activity for organisms, and is known to elicit alterations in the daily activity patterns of individuals. I investigated reproductive tradeoffs in the Arizona Bark Scorpion,Centruroides sculpturatus. To better understand the nature of tradeoffs in this species, I examined the influence of reproductive state on the predatory efficiency and thermal preference of reproductive females. Grass shrimp,Palaemonetes pugio, can survive in brackish waters and estuarine ecosystems despite the frequent oscillations and fluctuations in salinity, temperature and oxygen. AdultP. pugiohave the ability to osmoregulate (Romney and Reiber 2011), change cardiac parameters to tolerate temperatures (not yet published, Mika and Reiber) and oxyregulate (Guadagnoli and Reiber 2013). Manipulation of cardiac parameters allows for these methods of regulation. However, cardiac contraction and internal convection of oxygen do not occur until later stages of embryonic development. Studies focused on these morphological and physiological advantages may provide further understanding of the regulatory mechanisms within grass shrimp embryos, larvae and adults. …

The goal of this dissertation was to exploit the tractability of the honey bee (Apis mellifera) to understand how the physiological and cellular mechanisms that determine the onset and duration of senescence are shaped by behavioral development and behavioral intensity. These data reveal how behavior can damage cells and consequently limit lifespan. The honey bee represents the ideal model to address these factors because age, behavior, functional senescence, and lifespan are easily manipulated independently of each other while in its natural environment. Sleep is a universal phenomenon in vertebrates and lack of sleep has been linked with various abnormal behaviors (Singh et al 2013). Studies have shown that a strong linkage exists between stress and sleep, or lack thereof. In fact, the Better Sleep Council’s 2009 survey revealed that 65% of Americans lose sleep due to elevated stress (Wells and Vaughn 2012). Continual (chronic) elevated stress levels have been linked with serious negative health effects. The overall goal of this dissertation was to examine inflammatory and regenerative responses to acute skeletal muscle damage and to define molecular mediators of repair. Study I examined the effects of an oral anti-inflammatory supplement on exercise-induced muscle damage (EIMD) and systemic inflammation in a human model. Quercetin has been shown in animal and in vitro models to downregulate nuclear factor-kappa beta (NF-κB) nuclear transactivation and monocyte chemoattractant protein 1 (MCP-1) secretion, which regulate muscle regeneration and inflammatory signaling between muscle and immune cells after injury. Introduced species are the greatest threat to biodiversity after habitat loss. Understanding the processes that permit organisms to become successful invaders may provide opportunities to prevent or limit their dispersal and establishment and thereby alleviate some of their harmful effects. The goal of my dissertation research has been to investigate whether invasive species have distinctive interactions with parasites, and some of the mechanisms that may underlie that variation. I used one of the world's most successful vertebrate invaders as a case study: the house sparrow (Passer domesticus; Introduction).

Understanding the efficiency and digestibility of dietary nutrients in dairy heifers is essential for minimizing costs and environmental impacts of the dairy industry. The objective of this study was to interpret the effects of manipulating rumen-degradable protein (RDP) in a precision feeding system. In spite of significant progress in our knowledge of mechanisms that control blood pressure, our understanding of the pathogenesis of hypertension, its genetics, and population efforts to control blood pressure, hypertension remains the leading risk factor for mortality worldwide. It’s estimated that 1 out of every 3 adults has hypertension. Hypertension is a major risk factor for cardiovascular disease and stroke, and is considered a primary or contributing cause of death to more than 2.4 million US deaths each year. The pesticide, 3-trifluoromethyl-4-nitrophenol (TFM), has been highly successful in the control of sea lamprey (Petromyzon marinus) populations in the Great Lakes. Treatments with TFM involve applying it to streams, where it targets larval sea lamprey which live burrowed in the stream substrate. While the toxic mechanism of TFM has been elucidated, and its effects on sea lamprey described, its effects on non-target fish species such as rainbow trout (Oncorhynchus mykiss) and lake sturgeon (Acipenser fulvescens) are not as well understood.
