Analysis Of An Outdoor Pilot-Scale Rotating Algae Biofilm Reactor For Power Optimization, Ash-Enhanced Productivity, And Nutrient Uptake, Peter F. Jeppesen
All Graduate Theses and Dissertations, Fall 2023 to Present
Outdoor testing of a rotating algae biofilm reactor (RABR) was performed to see how well it could remove nutrients from water and produce algae. By adjusting the operation settings, we found the lowest power needed to remove nitrogen and phosphorus. The lowest power values needed were 1.36 kWh per kilogram of nitrogen and 20.1 kWh per kilogram of phosphorus. The algae production rate reached a maximum of 7.09 grams per square meter per day. Detailed analysis showed that the reactor worked like a continuous flow system, and the performance was influenced by factors including light and temperature. The results also …
Biophysical Model Of Retraction Motor Neurons And Their Modification By Operant Conditioning, Maria Rasheed
Biophysical Model Of Retraction Motor Neurons And Their Modification By Operant Conditioning, Maria Rasheed
Dissertations & Theses (Open Access)
Operant conditioning (OC) is a form of associative learning in which an animal modifies its behavior based on the consequences that follow that behavior. Despite its ubiquity, the underlying mechanisms of OC are poorly understood. Insights into the mechanisms of OC can be obtained by studying Aplysia feeding behavior as it can be modified by OC. This behavior is mediated by a central pattern generator (CPG) network in the buccal ganglia that contains a relatively small number of neurons. This CPG generates rhythmic motor patterns (BMPs) that move food into the gut by closing a tongue-like structure (i.e., radula) during …
Bcl2 Mediated Targeted Drug Delivery For The Treatment Of Kidney Fibrosis And Stomach Cancer, Humayra Afrin
Bcl2 Mediated Targeted Drug Delivery For The Treatment Of Kidney Fibrosis And Stomach Cancer, Humayra Afrin
Open Access Theses & Dissertations
Apoptosis, the programmed death of cells, is primarily regulated by a delicate balance between pro-apoptotic and anti-apoptotic signals. The Bcl-2 (B-cell lymphoma 2) family of proteins acts as anti-apoptotic agents, promoting cell survival. Dysregulation of these proteins is a common occurrence in conditions such as cancer and fibrosis, where overexpression of anti-apoptotic members can foster tumor cell survival and fibroblast activation. In this study, our aim was to explore the therapeutic potential of Bcl-2 inhibitors, both as a small molecule (specifically Navitoclax (Navi)), inhibitor and as Bcl-2 siRNA, for targeted treatment. Intravenous administration of Navi often leads to thrombocytopenia, necessitating …

Since humans last went to the moon on the Apollo missions, there has been a fascination with inhabiting and colonizing other planetary objects, starting with the Earth’s Moon, Mars, and recently Jupiter’s moon, Europa. However, there is still lack of knowledge and science behind many of the extraterrestrial environmental effects on biological organisms living on Earth. Therefore, there is a need to study how such environments would affect these organisms. Moreover, how can we attain sustainable living in such environments as well through self-providing life support systems (LSS), without the need to provide for additional necessities back and forth from …

Pulsed Electric Fields Sensitize Methicillin-Resistant Staphylococcus Aureus To Antibacterial Therapies And Stimulate Host Immune Responses, Alexandra E. Chittams-Miles
Biomedical Sciences Theses & Dissertations
This research explores the impact of nanosecond pulsed electric fields (nsPEF) on two fronts: their immune stimulatory effects and their potential as a novel strategy to enhance the sensitivity of Methicillin-resistant Staphylococcus aureus (MRSA) to clinically relevant antibiotics. While pulsed electric fields have been reported to have an immune stimulatory effect, the mechanisms responsible for these effects have yet to be determined.
Our investigation addresses the rising concern of MRSA derived skin and soft tissue infections (SSTIs). Consistent with other publications, we found that nsPEF alone cause modest inactivation of planktonic MRSA. We then investigated the effects of nsPEF in …

Under Pressure: The Soft Robotic Clap-And-Fling Of Cuvierina Atlantica, Daniel Mead
USF Tampa Graduate Theses and Dissertations
Evolution over billions of years has led to unique animals of all types. The tiny sea butterfly is one that remains mostly anonymous because of its size and low place on the food chain, but it is a swimming creature that when examined closely reveals a surprise. Shockingly, it has a swimming motion nearly identical to small insects flying at intermediate Reynolds numbers, referred to as the clap-and-fling. The centimeter long pteropod flapping briskly at 5Hz escapes attention with its size and speed. Modeling the clap-and-fling motion of the sea butterfly at a larger scale allows the benefits and uniqueness …

Bidirectional In-Situ Analysis And Visualization Of Fluid Structure Interactions With Applications In Blood Flow, Nazariy Tishchenko
Graduate Research Theses & Dissertations
Interactive blood flow simulation has applications in the understanding of complex fluids, training medical professionals, diagnostics, and surgical procedure planning. Bidirectional in situ visualization and analysis are much needed to streamline the analysis of large-scale patient-specific blood flow simulations. The in situ component ensures the live visualization of the simulation results while the simulation is running, and the bidirectional component allows for real-time control and configuration of the simulation parameters (i.e., setting and retrieving the simulation parameters and data based on need). In this project, a highly accurate multiphysics simulation was instrumented to work with an existing bidirectional in situ framework. …
Assessing Stormwater Management Pond Water Quality, Function, And The Potential Biotic Effects To Receiving Waters, Mitchell Elstone
Assessing Stormwater Management Pond Water Quality, Function, And The Potential Biotic Effects To Receiving Waters, Mitchell Elstone
Theses and Dissertations (Comprehensive)
The use of stormwater management ponds (SWMPs) has been increasing over the past five decades. However, an in-depth understanding of the daily performance of SWMPs and functionality during cold periods is limited. This is in part because mandated monitoring is relatively infrequent, and the assumption that SWMPs are inactive between storm events and during the winter. The goals of this research were to better understand daily stormwater (SW) characteristics, the performance of SWMPs based on current forms of evaluation and assess the potential for SWMP effluent to impact downstream biota. Influent and effluent samples from two SWMPs were collected daily …
Determining The Effects Of Glycocalyx Modifications On The Electrophysical Properties Of Human Mesenchymal Stem Cells, Rominna E. Valentine Ico
Determining The Effects Of Glycocalyx Modifications On The Electrophysical Properties Of Human Mesenchymal Stem Cells, Rominna E. Valentine Ico
Electronic Theses, Projects, and Dissertations
Human mesenchymal stem cells (hMSCs) have gained popularity in clinical trials due to their multipotent differentiation characteristics, ability to secrete bioactive molecules, migrate into diseased or damaged tissues, and their immunosuppressive properties. HMSC cultures are heterogeneous, containing stem cells, partially differentiated progenitor cells, and fully differentiated cells. One of the major challenges with hMSCs therapeutic potential is the inability to select specific cell subpopulations due to an insufficient number of biomarkers. Often the biomarkers used, like those for fluorescence-activated cell sorting, are not sufficient to define hMSCs because they overlap with other cell types. Consequently, there is a need to …

Deep Ocean Vehicle Applications And Modifications, Nichole "Nikki" T. Arm
Master's Theses
This project had two primary goals: (1) to explore opportunities to further a deep-ocean vehicle’s reach using alternative pressure spheres, and (2) to implement an existing deep-ocean vehicle (lander) in active scientific research.
I gained a greater understanding of the limitations and design choices made for existing pressure spheres using Finite Element Analysis (FEA). My simplified FEA model predicted sphere failure for the existing 30% Fiber Glass 70% Nylon injection molded spheres at an external pressure of 3,954psi or 2,690m ocean-depth (only a 7.38% error compared to the tested minimum failure depth), so I determined it a valid model. I …

Analyzing Pseudomonas Aeruginosa With Bacteriophage Tags Using Photoacoustic Flow Cytometry, Jennifer C. Schinke
Electronic Theses and Dissertations
The number of daily bacterial infections is climbing and the CDC explains that this is due to the antibiotic-resistant threat in the United States. Finding a faster way of bacterial identification is necessary as it currently takes 1-4 days for a medical lab to culture and identify bacteria. Photoacoustic flow cytometry (PAFC) can be used as an alternative method resulting in swift identification within an hour (Edgar, 2019). Pseudomonas aeruginosa, cell line PA01, will be coated in up to a few hundred red dyed phages making it detectible by the photoacoustic flow cytometry system. Bacteriophages (phages) are viruses that …
Resource Recovery From Crew Metabolic Wastes In Lunar And Martian Habitats: Reuse Of Effluent From An Anaerobic Membrane Bioreactor Treating Fecal Organic Matter As Fertilizer Source For Plants, Alexandra A. Smith
USF Tampa Graduate Theses and Dissertations
As humans set their sights on exploration of the Moon and Mars human focused challenges like providing potable water, supplying nutritious food to the astronaut crew, and managing their waste present themselves. There are no readily available resources on the Moon and Mars, therefore nothing should be considered unusable including the organic wastes (i.e., feces and urine) produced by the crew. Fecal matter is not currently recycled and is treated as solid waste. After collection, it is discarded out of the International Space Station where it is incinerated upon re-entry. This approach will not work on the Moon’s surface. There …

Development Of A Computational Model To Investigate Pathways And The Effects Of Treatment In Fanconi Anemia, Sabrina Kellett
Development Of A Computational Model To Investigate Pathways And The Effects Of Treatment In Fanconi Anemia, Sabrina Kellett
Biological Sciences Undergraduate Honors Theses
Fanconi Anemia (FA) is a rare type of anemia that is not easily studied and can have very detrimental effects. This disease compromises the bone marrow, resulting in decreased hemopoiesis. Symptoms of FA also include abnormalities in the brain and spinal cord, incorrect formation of the kidneys, abnormal formation of the heart and lungs, and a dramatically increased risk of developing cancer. FA can be caused by various mutations in any of the 22 genes that encode for proteins involved in what is called the FA DNA repair pathway. In healthy individuals, this pathway specifically repairs interstrand cross-links (ICLs) recognized …

Pulsed power treatment has been used to induce regulated cell death (RCD) in cells or ablate tumors in animals. A subset of pulsed power as electroporation with high voltage and pulse duration of milliseconds is used for biomedical treatment to induce pores in the plasma membrane of cells. Nanosecond Pulsed Electric Fields (nsPEFs)– an extension of electroporation, uses waveforms with pulse durations on the order of 10-900 nanoseconds. nsPEF treatment has demonstrated intracellular effects for potential biomedical applications. In this work, nsPEF treatment is used to demonstrate changes that affect viability, plasma membrane permeability ROS (Reactive Oxygen Species) in the …
Amphisbaenian Head Movement And Burrowing Forces In Damp Granular Media, Jacob Newell
Amphisbaenian Head Movement And Burrowing Forces In Damp Granular Media, Jacob Newell
Williams Honors College, Honors Research Projects
Damp granular media is a difficult environment to study because it is both practically complex and it lacks equations which fully describe its behavior. In this study, an oscillatory lateral head movement and its effects while penetrating damp granular media were tested using a robophysical model. This experimental research was inspired by the burrowing behavior of the clade Amphisbaenia, a group of usually limbless squamates that employ a variety of different burrowing behaviors, but it can apply to a wide range of burrowers. This research could help with both human burrowing technologies and the further investigation of animal behaviors.
Winter Dynamics Of Storm Water Management Ponds And Winter Tolerance In Three Aquatic Plant Species, Patrick Strzalkowski
Winter Dynamics Of Storm Water Management Ponds And Winter Tolerance In Three Aquatic Plant Species, Patrick Strzalkowski
Theses and Dissertations (Comprehensive)
The vast majority of the research into the performance of stormwater management ponds (SWMPs) has been performed in warm regions or during the warmer seasons in temperate regions. It is presumed that SWMPs are inactive in the winter as any potential stormwater is trapped in snow and ice. The main goal of this thesis was to test this presumption and to study the dynamics and performance of three SWMPs during the winter. Remote water level loggers were installed into the three SWMPs and daily grab samples from the influents and effluents were taken and analyzed for total phosphorus (TP), chloride, …
Functionality Of The Gracilis Collateral Is Impaired Following Arteriogenesis And Cell Transplantation, Ada A. Tadeo
Functionality Of The Gracilis Collateral Is Impaired Following Arteriogenesis And Cell Transplantation, Ada A. Tadeo
Biomedical Engineering
Peripheral Arterial Disease involves narrowed arteries, reducing blood flow to limbs. Increasing blood flow to those extremities is possible by redirecting the blood to flow through natural bypasses (i.e. collateral arteries), which can enlarge via arteriogenesis to maintain blood supply once the prominent arteries have become occluded. This study aims to investigate how arteriogenesis affects collateral function, if myoblast transplantation can stimulate collateral growth, and how that in turn may affect collateral function. Femoral artery ligation was performed to mimic the blockage that occurs in patients with ischemic diseases on lean mice and mice with diet induced obesity (DIO). A …

Biological understanding of complex diseases such as stroke and obesity is critical for the advancement of medicine. Further knowledge discovery can provide effective biomarkers to improve disease diagnosis and prognosis, identify driver mutations, predict individual genetic susceptibility for early prevention and effective disease management, and facilitate development of personalized drugs. Stroke is the second leading cause of death and long-term disability in the world. Thus, stroke management is a time-sensitive emergency. The initial hours after stroke onset map the trajectory of subsequent neurologic complications. Cerebral edema develops hours to days after acute ischemic stroke and may result in midline shift …
Engineering Of Ideal Systems For The Study And Direction Of Stem Cell Asymmetrical Division And Fate Determination, Martina Zamponi
Engineering Of Ideal Systems For The Study And Direction Of Stem Cell Asymmetrical Division And Fate Determination, Martina Zamponi
Biomedical Engineering Theses & Dissertations
The cellular microenvironment varies significantly across tissues, and it is constituted by both resident cells and the macromolecules they are exposed to. Cues that the cells receive from the microenvironment, as well as the signaling transmitted to it, affect their physiology and behavior. This notion is valid in the context of stem cells, which are susceptible to biochemical and biomechanical signaling exchanged with the microenvironment, and which plays a fundamental role in establishing fate determination and cell differentiation events. The definition of the molecular mechanisms that drive stem cell asymmetrical division, and how these are modulated by microenvironmental signaling, is …
The Synthesis And Characterization Of Ionic Liquids Using Nitrogen-Based Cations For Transdermal Delivery, Jo Galloway
The Synthesis And Characterization Of Ionic Liquids Using Nitrogen-Based Cations For Transdermal Delivery, Jo Galloway
Honors Theses
The purpose of this research was to explore the synthesis mechanisms of water-soluble ionic liquids with nitrogen-containing cationic bases for future use in transdermal drug delivery and forensic science applications. Ionic liquids are salts with an organic cation and either an organic or inorganic anion. They have asymmetric structures, which means that the molecules don’t pack together as neatly as other salts do, therefore, they don’t crystallize as easily, and their melting points are lower. Ionic liquids have melting points below 100°C, and many are liquid at room temperature. Mechanistic studies reveal that the potency of ILs in enhancing transdermal …
A Tool For Biometric Interpretation Of Forensic Str Dna Profiles, Ahmad Jamal Baroudi
A Tool For Biometric Interpretation Of Forensic Str Dna Profiles, Ahmad Jamal Baroudi
Graduate Theses, Dissertations, and Problem Reports
Rapid DNA biometric identification applications are becoming more essential and widely used in human identity validation processes. Despite their powerful identification capabilities, processing a sample to generate a forensic DNA profile still takes longer compared with other rapid biometric technologies. Methods used to speed up the analysis could lead to signal artifacts similar to those arising from low copy or degraded DNA samples, making the electropherogram unsuitable for forensic interpretation and analysis. The goal of this research effort is to apply biometrics and mathematical approaches to forensic STR (Short Tandem Repeat) profiles. To accomplish this goal, a multi-function software tool …

Histological Comparison Of Shark Dermis Across Various Ecomorphologies, Olivia Schuitema
UNF Graduate Theses and Dissertations
The integument plays essential roles in the structural support, protection, and hydrodynamic capability among fishes. Most shark skin research has been done on the external epidermal layer containing the dermal denticles, while the larger dermis layer has been mostly ignored. The dermis layer in sharks is composed of two layers, the upper stratum laxum and the lower stratum compactum, holding supportive collagen and elastin fibers. There may be morphological and compositional differences in the dermis layers across various species of sharks, although the extent of such is unknown. These potential differences may be represented by various dermal thicknesses and different …
Exosome- And Microrna-Based Therapeutic Approach For Tendinopathy, Angela Wang Ilaltdinov
Exosome- And Microrna-Based Therapeutic Approach For Tendinopathy, Angela Wang Ilaltdinov
Dissertations and Theses
Tendinopathy, characterized by degeneration and chronic inflammation, is a significant clinical burden. Current treatments focus on symptom management but do not sufficiently address its underlying pathology; however, stem cell-based approaches aimed at repairing diseased tissues may overcome this limitation. Therapeutic effects of stem cells may be due in part to paracrine actions, including some mediated by exosomes – extracellular vesicles secreted by cells that play a role in cell communication. MicroRNA (miRNA), small non-coding RNA carried by exosomes, are likely responsible for many exosome effects. Exosomes and miRNA therapies show promise in treating diseases such as cancer and arthritis, but …

Impact Of Orthophosphate As A Corrosion Inhibitor And Chloramine Disinfectant On Drinking Water Biofilm Communities, Mitchell Cooke
Theses and Dissertations (Comprehensive)
A drinking water distribution system (DWDS) must maintain conditions within quality standards which assure the effective and safe transport of finished drinking water from treatment plants to the household tap. Although safe to drink, finished water is not sterile, and may contain hundreds of microorganisms in a single milliliter. These microorganisms are present from the source waters, such as lakes, rivers and aquifers, and have passed through early treatment steps. Final treatment steps, such as the maintenance of disinfectant residuals, are used to further minimize viable cells present and focus on the reduction of harmful organisms. Microbial cells entering the …

The Investigations Of Nps Modulated Immunity And Immunometabolism, Brittney Leigh Ruedlinger
Biomedical Sciences Theses & Dissertations
Cancers remain in the top noncommunicable diseases responsible for premature mortality. The heterogeneity among cancers and within tumors makes treating them ever more challenging. Our misfortune for developing cures is mocked by cancer, with the lowest probability of success (PoS) through clinical trials and FDA approval. At the basic level, there are generally two broad gaps impeding cancer eradication: the unidentified shared mechanism(s) exploited by all cancers and the therapeutic approach to intervene. Nanosecond pulse stimulation (NPS) offers a unique approach since its broad impacts intersect those often hijacked by oncogenesis. Metabolic pathways, known for dysfunctions among cancers, share a …
