Contaminated sediments in marine environments have been shown to be good indicators of ecological risk and a means to assess anthropogenic impacts on marine habitats and the animals that inhabit them (Long et al. 1995, Rattner 2009). Estuarine sediments are especially complex media with regard to physical, chemical, and biological characteristics that trap, store, modify and sometimes release contaminants to the biota (Long et al. 1995). Large bodies of water, such as the Great Lakes in North America, are potential barriers to migrating bats. Islands may be used as stopover sites by bats crossing the Great Lakes as stopover sites, or included in crossing routes to reduce the distance flown over open water. I predicted that stopover use by migrating bats would result in higher activity on Great Lakes islands during late-season migration than at mainland sites. Successful captive-breeding and re-introduction programs must have the ability to breed a surplus of genetically suitable animals for release into the wild. Unfortunately, many individuals in captive breeding programs often do not reproduce even when they are apparently healthy and presented with genetically appropriate mates. Mate choice can affect multiple parameters of reproductive sperformance, including mating success, offspring production, survival, and fecundity. We investigated the role of mate preference and personality on the reproductive performance of male and female giant pandas (Ailuropoda melanoleuca) as measured by intromission success and litter production. Pacific lamprey (Entosphenus tridentatus), an endemic species to the Columbia River Basin, U.S.A, has experienced staggering decreases in returns to spawning territories in recent decades. As lamprey are threatened severely by a lack of passage at mainstem dams, lamprey specific passage structures have been designed and constructed to address the problem. The crop pest Drosophila suzukii, native to Southeast Asia, has been found in Ontario since 2010. This raises concern with respect to it establishing permanent populations, however, little is known about its cold tolerance. I investigated the low-temperature tolerance, including phenotypic plasticity, of D. suzukii. While acclimation increased cold tolerance, there was no evidence of short-term cold-hardening. Chill coma occurs at -1.2 °C, which will limit winter activity. Cold shock decreased the reproductive output of females, but this negative effect may be mitigated by re-mating. Drosophila suzukii is chill-susceptible and 80 % of the flies die after exposure …

Choline and vitamin B12 are essential nutrients for growth and performances of production animals. However, both nutrients are extensively degraded during digestion in the rumen. This thesis comprised three experiments. First, four cows equipped with a rumen cannula and catheters in the portal vein and a mesenteric artery received a post-ruminal bolus of: 1) cyanocobalamin (CN-CBL) alone (0.1 g) [gram], 2) CN-CBL (0.1 g) + casein (10 g) or 3) CN-CBL (0.1 g) + whey proteins (10 g). Quagga mussels (Dreissena rostiformis bugensis) act as ecosystem engineers in the southern basin of Lake Michigan, altering physical habitats and biogeochemical processes. Adapted to cold and oligotrophic conditions, profunda quagga mussels thrive on the soft substrate of deeper depths. At a 55 m site (10,000 mussels m-2) offshore from Milwaukee, WI, profunda mussel biomass (g m-2) was 1/3 of biomass (g m-2) measured at a 10 m comparison site (5,000 mussels m-2). Detrusor-sphincter dyssynergia after chronic spinal cord injury (SCI) leads to over-distention, high bladder pressures and vesico-ureteric reflux into the kidney, resulting in repeated infections and pyelonephritis. Given the importance/limited knowledge of SCI effects on the upper urinary tract, this study assessed expression of two proteins in the kidneys of contused (T9) male rats relative to controls that are indicative of tissue stress/damage (TGFβ, CD11b). 4,096 broiler chicks were randomly allocated to 128 floor pens (32 birds/pen). 2,048 day-old male broilers were placed in the east end of a barn, and the following week 2,048 day-old male broilers were placed in the west end. At each placement day, half of the chicks were Cobb 500 and half were Ross 708, and each pen contained only one breed source. East end birds received coccidiostat in the feed, west end received coccidial vaccine, and each end was under separate environmental control. Haplosporidium nelsoni is a protozoan parasite that causes the devastating disease MSX in the Eastern oyster, Crassostrea virginica, along the East coast of the United States. Until recently, H. nelsoni had not reached epizootic levels in Maine. However, in the summer of 2010, H. nelsoni was responsible for significant mortalities in cultivated Eastern oysters in the Damariscotta River Estuary. Since the initial outbreak, surveys performed by the Department of Marine Resources (DMR) show that MSX is still prevalent in the Damariscotta River Estuary. The objective of this study was to evaluate the intestinal porcine epithelial cell-jejunum 2 (IPEC-J2) cell line as a model to study the innate immune function of live pigs. Growth rates of IPEC-J2 cells in T-75 flasks and 96-well plates were evaluated using a hemocytometer and spectrophotometer for cell quantity measurements to determine growth rate and doubling time. Growth rates of IPEC-J2 cells in T-75 flasks and 96-well plates were 0.4016 and 0.2851 times of doubling/day respectively with a doubling time of 1.73 d and 2.43 d, respectively. Resident killer whales, Orcinus orca, of the Northeastern Pacific form stable kinship-based matrifocal associations and communicate with group-specific repertoires of discrete calls (dialects) that reflect these associations. The gradual fission of matrilines is usually consistent with dialect variations among groups that may manifest as differences in call usage at the repertoire level or subtle structural differences of the calls themselves. Therefore, matrilines that are more closely related tend to be more acoustically similar. The objective of this study was to determine the central effect of the VT4R antagonists (SR-49059 and H-5350 (Manning compound)) on corticosterone (CORT) levels during stress and food intake. A 22-gauge stainless steel guide cannula was surgically implanted into the lateral ventricle of the birds. A preliminary study was done to test the antagonists and their role on food intake. Birds were injected with saline, NPY (4µg), NPY (4µg)+SR-49059 (250ng), or NPY (4µg)+Manning compound (250ng). Birds injected with saline had the lowest 1h food intake (17.7g1.6). Tall fescue (Lolium arundinaceum Schreb) is the most prevalent forage in the Mid-south United States due to its observed hearty nature and ability to withstand heavy grazing. This is due to a symbiotic relationship with an endophytic fungus (Neotyphodium coenophialum). This fungus produces ergot alkaloids, which can bind to many adrenergic, serotonergic, and dopaminergic receptors. One of the primary functions of dopamine is to inhibit prolactin response. Decreased growth, reproduction, lactation, and depressed serum prolactin concentrations are commonly observed in beef cattle grazing endophyte-infected tall fescue. The aim of this study was to examine relationships between bull temperament, behavior, growth, endocrine, and reproductive parameters measured over an 84 d test period. Bulls (n = 65) were reared in 6 pens separated on BW and age. Pen scores were assigned on d -1, 27, 55 and 83. On d 0, 28, 56, and 84, exit velocity, BW, time it took each bull to leave the chute, bull order through chute, and hair and blood samples were recorded or collected. Research trials were conducted to evaluate the effects of different levels of dietary grain sorghum on broiler live performance, carcass yield and shank (leg) and breast skin coloring. Iso-caloric diets were formulated where sorghum replaced corn at rates of 0% (control), 20, 40, 60, 80 and 100% for a total of 6 diets. For each of the two trials, 1500 Cobb 500 male broiler chicks were randomly allocated to 60 pens with 25 birds per pen (10 pens/diet) and grown to 46 days for the first research trial and 41 days for the second. Two trials were performed using one-day-old male Cobb x Cobb 500 broilers to determine how dietary phytate and phytase levels as well as phytase phase feeding impacted bird performance parameters, tibia characteristics, and malonaldehyde (MDA) content of the liver, breast and thigh tissues. The first experiment consisted of 1,008 birds randomly placed in 48 floor pens within two commercial broiler houses at the Applied Broiler Research Farm (ABRF; 21 birds per pen; 0.76 ft2 per bird). First, we determined the post-denning behaviors of female black bears in order to help management agencies protect bears from human disturbances as well as set spring hunts that minimize the taking of females with dependent young. Feral horses occupy 31.6 million acres throughout western North America. Feral horses share similar habitats with a wide range of animal species, including pronghorn. Since horses are larger and often more aggressive than other animals of this region, they are generally socially dominant over all other native ungulate species. Pronghorn share water sources with horses in areas where both occur. In situations where horses exclude pronghorn from water, pronghorn fitness may be impaired, especially during the hottest months of the year when water is limited. Next-generation sequencing (NGS) has revolutionized the field of genetics by providing a means for fast and relatively affordable sequencing. With the advancement of NGS, whole- genome sequencing (WGS) has become more commonplace. However, sequencing an entire genome is still not cost effective or even beneficial in all cases. In studies that do not require a whole-genome survey, WGS yields lower sequencing depth and sequencing of uninformative loci. Targeted sequencing utilizes the speed and low cost of NGS while providing deeper coverage for desired loci. The genus Fusarium has a wide host range and causes many different forms of plant disease. These include seed rot and seedling blight diseases of cultivated plants. The Fusarium-caused diseases of wild plants are less well-known. In this study we examined Fusarium sp. n-caused disease development on non-dormant seeds of the important rangeland weed Bromus tectorum as part of broader studies of the phenomenon of stand failure or ‘die-off’ in this annual grass. The superfamily Haploporoidea Nicoll, 1914 comprises two families, the Atractotrematidae Yamaguti, 1939 and the Haploporidae Nicoll, 1914, and the subfamily Cadenatellinae Gibson et Bray, 1982. All members are parasites of the alimentary tract or gall bladder of marine, estuarine, and freshwater herbivorous fishes. Overstreet and Curran (2005a) provided a review of the Haploporidae and recognized four subfamilies, the Chalcinotrematinae Overstreet et Curran, 2005, Haploporinae Nicoll, 1914, Megasoleninae Manter, 1935, and Waretrematinae Srivastava, 1937. Observations of Bromus tectorum L. (cheatgrass or downy brome) monocultures have shown that populations are susceptible to stand die-off or replacement failures. Die-offs, where the seed bank from the previous year fails to emerge, occurs in cheatgrass stands and it is unclear the trigger or cause. The fungus Fusarium has been identified in plant and seed samples from die-offs and may drive die-off activity through pathogenicity. Pinyon-juniper woodland encroachment threatens ecosystem function and diversity on sagebrush steppe. Decreased fire frequency likely favors proliferation of pinyon-juniper woodlands and subsequent decline in desirable understory species. Increased tree cover produces hazardous canopy fuel loads that contribute to severe crown fires and threaten life and property at the wildland-urban-interface. Mechanical mastication converts large canopy fuels into small woody debris, altering wildfire dynamics from a potential crown fire to a more controllable surface fire. Currently the size and frequency of wildfires are increasing at a global scale, including arid ecosystems that exhibit great sensitivity to disturbance. Fire effects on plant pollination and reproductive success in deserts are largely unknown. Plant dependence on animal pollinators for reproduction can increase the risk of reproductive failure if pollination services are hindered or lost. Species that depend on few taxonomically related pollinator species are expected to be most negatively affected by disturbances that disrupt pollination interactions. Invasive species are one of the leading causes of biodiversity loss and their presence can significantly impact the structure and function of native ecosystems. In this dissertation, the impacts of the invasive honeysuckle shrub, Lonicera maackii, on exotic earthworm populations, leaf litter decay responses, and inorganic and organic nutrient pools in an urban woodland park are assessed. Chapter 1 provides a brief review of honeysuckle's effects on forest ecosystems. Chapter 2 describes honeysuckle’s effects on exotic earthworm populations and the seasonal importance of macroinvertebrates to leaf litter decomposition in urban woodlands.
