Background

The invasion associated marker (iam) has been detected in the majority of invasive Campylobacter jejuni retrieved from humans. Furthermore, the detection of iam in C. jejuni isolated from two important hosts, humans and chickens, suggested a role for this marker in C. jejuni's colonization of multiple hosts. However, no data exist regarding the occurrence of this marker in C. jejuni isolated from non-poultry food-animals such as cattle, an increasingly important source for human infections. Reliable density estimates of game and keystone species such as white-tailed deer (Odocoileus virginianus) are desirable to set proper management strategies and for evaluating those strategies over time. However, traditional methods for estimating white-tailed deer density have been inhibited by behavior, densely forested areas that can hamper observation (detection), and invalid techniques of estimating effective trapping area. We wanted to evaluate a noninvasive method of mark-recapture estimation using DNA extracted from fecal pellets as the individual marker and for gender determination, coupled with a spatial detection function to estimate density (Spatially Explicit Capture-Recapture, SECR). The study examined the influence of timing of prescribed burn on native warm-season grass stands in Tennessee. The purpose of the study was to determine the nutritional quality of forage as it relates to the timing of prescribed burns on native warm-season grass stands in Tennessee. The prescribed burns were conducted in March, April, May, and September. Given the white-tailed deer’s (Odocoileus virginianus; deer) popularity and potentially negative impact on forested systems; Arnold Air Force Base (AAFB) in Tullahoma, Tennessee, USA has made minimizing negative deer impacts on biodiversity a priority. To address these management issues, I initiated a study on AAFB to investigate deer survey techniques and the effects of deer density on forage availability across vegetative communities.

Elevated ambient temperatures negatively impacts pregnancy rates in agriculturally important females. An oocyte undergoing meiotic maturation exposed to a physiologically-relevant heat stress results in alterations of developmentally important processes and reportedly decreases subsequent developmental potential after fertilization. To address problems of reduced competence after exposure to heat stress for the first 12 h of meiotic maturation (hIVM), Payton (2009) assessed the transcriptome profiles of oocytes and associated cumulus cells by microarray analysis and reported heat-induced perturbations at the molecular level in the oocyte. The reproductive neuropeptide, kisspeptin (KP), may have a role in regulation of growth hormone (GH) secretion. Expression of KP and its receptor (Kiss1r) are developmentally and hormonally regulated. This study was conducted to determine the effects of KP on plasma concentrations of luteinizing hormone (LH) and GH in pubertal heifers during different functional reproductive stages. Heifers received a single injection of KP (100 pmole / kg) or saline intravenously during proestrus, estrus, and diestrus, and serial blood samples were collected for 3 hours to determine the response of both LH and GH after treatment. Kisspeptin ( KP ), a neuroendocrine regulator of reproduction, is hypothesized to be an integrator of metabolism and hormones critical to the regulation of reproduction. Lactation is associated with enhanced growth hormone (GH) responsiveness and reduced fertility. Our study was designed to determine the effects of lactation on KP-stimulated GH and luteinizing hormone (LH) secretion. Five non-lactating and five lactating dairy cows were used in the study. Experiments were conducted with lactating cows at weeks 1, 5 and 11 after parturition. The Effect Of Kisspeptin Receptor Agonist (Ftm080) On Luteinizing Hormone In Sheep, Brian K. Whitlock, Joseph A. Daniel, Maria Hes, Barbara P. Steele, James L. Sartin, S. Oishi, N. Fujii

Kisspeptin receptor (Kiss1r) agonists with increased half-life and similar efficacy to kisspeptin (KP) in vitro potentially may provide beneficial applications in breeding management of many species. However, many of these agonists have not been tested in vivo. This study was designed to test the effect of a Kiss1r agonist (FTM080) on luteinizing hormone (LH) in vivo. Sheep were treated with FTM080 (500 pmol/kg BW) or sterile water in a 2-ml bolus via the jugular cannula. Serial blood samples were collected every 15-min before (1 hr) and after (1 hr) treatment. Intravenous (IV) injection of FTM080 increased (P < 0.05) LH plasma concentrations through the 45-min sample following treatment. Moreover, the area under the curve of LH in the period from 0 to 60-min following FTM080 treatment was also increased (P < 0.05). These data provide evidence to suggest that FTM080 stimulates the gonadotropic axis of ruminants in vivo.

Consumption of toxic endophyte-infected (E+) tall fescue pastures is known to have a negative impact on bull reproductive performance. Since decreased cleavage rates of embryos fertilized with spermatozoa from bulls grazing E+ tall fescue pastures have been observed in several studies using differing sets of bulls, technicians, pastures, and other methods of inducing tall fescue toxicosis (ergotamine tartrate), it is hypothesized that spermatozoa function from bulls grazing E+ is impaired in ways undetectable by gross semen examination.

ABSTRACT

One cause of declines and extinctions of island species is carnivore introduction. Four carnivores, including the small Indian mongoose (Herpestes auropunctatus), are on the IUCN’s list of 100 of the World's Worst Invasive Alien Species. My thesis summarizes global patterns of carnivore introductions and examines ecological, evolutionary, and management impacts of this mongoose. I study abundances of reptiles and amphibians on mongoose-infested and mongoose-free islands in the Adriatic Sea to determine if factors other than mongoose presence can account for abundance differences.
