Influence Of An Internal Parasite Control On Cattle Grazing Behavior And Production, Jace Stott

Six herds of 45 to 90 cow/calf pairs grazing on upland range were used to examine the efficacy of an injectable extended release eprinomectin parasite control on production traits, activity behavior, and efficacy against internal parasitism. In 2016, treatment cows were given a subcutaneous injection of LongRangetm. In 2017, all cows in the study were treated with a short acting Synanthictm treatment and only treatment cows were given an additional LongRangetm treatment. In both 2016 and 2017, fecal egg counts were significantly lower (P < 0.05) in eprinomectin treated cows compared to control cows. Calf gains were 4.8 kg and 8.7 kg greater (P < 0.1) for the calves of dams treated with eprinomectin compared to calves of control cows in 2016 and 2017, respectively. Activity characteristics of cattle were inconclusive, with treated cows having lower (P < 0.01) grazing and traveling times compared to control cows in 2016, and more (P < 0.01) grazing and traveling in 2017.

Background: There are significant limitations in existing methods for the genome-wide identification of genes whose expression patterns affect traits.

Results: The transcriptomes of five tissues from 27 genetically diverse maize inbred lines were deeply sequenced to identify genes exhibiting high and low levels of expression variation across tissues or genotypes. Transcription factors are enriched among genes with the most variation in expression across tissues, as well as among genes with higher-than-median levels of variation in expression across genotypes. Drip-irrigation is increasingly applied in maize (Zea mays L.) production in sub-humid region. It is critical to quantify irrigation requirements during different growth stages under diverse climatic conditions. In this study, the Hybrid-Maize model was calibrated and applied in a sub-humid Heilongjiang Province in Northeast China to estimate irrigation requirements for drip irrigated maize during different crop physiological development stages and under diverse agro-climatic conditions. Using dimensionless scales, the whole growing season of maize was divided into diverse development stages from planting to maturity. Although organisms make resource selection decisions at multiple spatiotemporal scales, not all scales are ecologically relevant to any given organism. Ecological patterns and rhythms such as behavioral and climatic patterns may provide a consistent method for identifying ecologically relevant scales of habitat selection. Using elk (Cervus canadensis) as an example species, we sought to test the ability of behavioral patterns to empirically partition diel scales for modeling habitat selection. We used model selection to partition diel scales by shifts in dominant behavior and then used resource selection probability functions to model elk habitat selection hierarchically at diel scales within seasons. …

Establishing vegetation on roadsides following construction can be challenging, especially for relatively slow growing native species. Topsoil is generally removed during construction, and the surface soil following construction (“cut-slope soils”) is often compacted and low in nutrients, providing poor growing conditions for vegetation. Nebraska Department of Transportation (NDOT) protocols have historically called for nitrogen (N) and phosphorus (P) fertilization when planting roadside vegetation following construction, but these recommendations were developed for cool-season grass plantings and most current plantings use slower-establishing, native warmseason grasses that may benefit less than expected from current planting protocols. Common ragweed emerges early in the season in Nebraska, USA and is competitive with soybean; therefore, preplant herbicides are important for effective control. Glyphosate has been used as a preplant control option; however, confirmation of glyphosate-resistant (GR) common ragweed in Nebraska necessitates evaluating other herbicide options. Evaluation of seed protein alleles in soybean populations showed that an increase in protein concentration is generally associated with a decrease in oil concentration and yield.

Soybean [Glycine max (L.) Merrill] meal is one of the most important plant-based protein sources in the world. We established and elaborated on a method to enrich the membrane proteome of mature pollen from economically relevant crop using the example of Solanum lycopersicum (tomato). To isolate the pollen protein fraction enriched in membrane proteins, a high salt concentration (750 mM of sodium chloride) was used. The membrane protein-enriched fraction was then subjected to shotgun proteomics for identification of proteins, followed by in silico analysis to annotate and classify the detected proteins.

The medicinal legume Mucuna pruriens (L.) DC. has attracted attention worldwide as a source of the anti-Parkinson’s drug L-Dopa. It is also a popular green manure cover crop that offers many agronomic benefits including high protein content, nitrogen fixation and soil nutrients. The plant currently lacks genomic resources and there is limited knowledge on gene expression, metabolic pathways, and genetics of secondary metabolite production. Identifying and characterizing alternative splicing (AS) enables our understanding of the biological role of transcript isoform diversity. This study describes the use of publicly available RNA-Seq data to identify and characterize the global diversity of AS isoforms in maize using the inbred lines B73 and Mo17, and a related species, sorghum. Identification and characterization of AS within maize tissues revealed that genes expressed in seed exhibit the largest differential AS relative to other tissues examined. N leaching is closely linked to weather factors, especially rainfall. Prediction of N leaching in cropping systems is critical to improvement of crop management and reduction of N leaching. Corn (Zea mays L.) residue grazing or harvest provides a simple and economical practice to integrate crops and livestock, but limited information is available on how widespread corn residue utilization is practiced by US producers. In 2010, the USDA Economic Research Service surveyed producers from 19 states on corn grain and residue management practices. Total corn residue grazed or harvested was 4.87 million ha. Approximately 4.06 million ha was grazed by 11.7 million livestock (primarily cattle) in 2010. The fall armyworm, Spodoptera frugiperda (J. E. Smith) and the corn earworm, Helicoverpa zea (Boddie) are among the main pests of maize. Both species exhibit cannibalistic behavior and quite often share the same feeding guild in maize (maize ears), which can result in several interspecific and intraspecific interactions. Paired interaction scenarios of intraspecific and interspecific larvae were assessed in arenas in the presence and absence of food to characterize movements resulting from interactions of these insects. As researchers and land managers increasingly seek to understand plant invasions and the external (climate) and internal (plant genetics) conditions that govern the process, new insight is helping to answer the elusive question of what makes some invasions successful and others not. Aflatoxins are among themost powerful carcinogens in nature. The major aflatoxin-producing fungi are Aspergillus flavus and A. parasiticus. Numerous crops, including peanut, are susceptible to aflatoxin contamination by these fungi. There has been an increased use of RNA interference (RNAi) technology to control phytopathogenic fungi in recent years. Aflatoxin contamination is a major constraint in food production worldwide. In peanut (Arachis hypogaea L.), these toxic and carcinogenic aflatoxins are mainly produced by Aspergillus flavus Link and A. parasiticus Speare. The use of RNA interference (RNAi) is a promising method to reduce or prevent the accumula-tion of aflatoxin in peanut seed. In this study, we performed high-throughput sequencing of small RNApopulations in a control line and in two transformed peanut lines that expressed an inverted repeattargeting five genes involved in the aflatoxin-biosynthesis pathway and that showed up to 100% less aflatoxin B1 than the controls. Early vigor is an important trait for many rice (Oryza sativa L.)- growing environments. However, genetic characterization and improvement for early vigor is hindered by the temporal nature of the trait and strong genotype × environment effects. Gene flow is an important component in evolutionary biology; however, the role of gene flow in dispersal of herbicide-resistant alleles among weed populations is poorly understood. Field experiments were conducted at the University of Nebraska-Lincoln to quantify pollen-mediated gene flow (PMGF) from glyphosate-resistant (GR) to -susceptible (GS) common waterhemp using a concentric donorreceptor design. More than 130,000 common waterhemp plants were screened and 26,199 plants were confirmed resistant to glyphosate. Habitat selection by animals is influenced by and mitigates the effects of predation and environmental extremes. For birds, nest site selection is crucial to offspring production because nests are exposed to extreme weather and predation pressure. Predators that forage using olfaction often dominate nest predator communities; therefore, factors that influence olfactory detection (e.g., airflow and weather variables, including turbulence and moisture) should influence nest site selection and survival. The past few years have witnessed a paradigm shift in molecular systematics from phylogenetic methods (using one or a few genes) to those that can be described as phylogenomics (phylogenetic inference with entire genomes). One approach that has recently emerged is phylo-transcriptomics (transcriptome-based phylogenetic inference). As in any phylogenetics experiment, accurate orthology inference is critical to phylo-transcriptomics. To date, most analyses have inferred orthology based either on pure sequence similarity or using gene-tree approaches. An improved understanding of the impact of tillage systems on soil hydraulic properties is necessary to conserve and manage soil water under a changing climate. The objective of this study was to specifically measure soil hydraulic properties (total porosity, water infiltration, saturated hydraulic conductivity, and water retention characteristics) in no-till, chisel plow, disk, and moldboard plow systems under rainfed continuous corn (Zea mays L.) after 35 yr on silty clay loam soils in eastern Nebraska. Increased rainfall variability due to climate change threatens the efficacy of critical soil ecosystem services. One strategy to negate effects of too much or not enough rainfall is to improve soil water properties. Practices that offer “continuous living cover” can enhance soil water storage and other soil hydrologic properties relative to annual crop systems, but to what extent such benefits can accrue, under different conditions, remains under-quantified. Peanut (Arachis hypogaea) is a botanically indeterminate plant where flowering, fruit initiation, and pod maturity occurs over an extended time period during the growing season. As a result, the maturity and size of individual peanut pods vary considerably at harvest. Immature kernels that meet commercial edible size specifications negatively affect quality during processing due to their increased propensity for off flavors, higher moisture and water activity, and variable roasting properties. As peanuts progress toward maturation, late season flowers set within 40 days till harvest will not have sufficient time to develop into mature, marketable pods prior to harvest. …

Molecular mechanisms that plants use to sense and respond to Fe deficiency by coordinated expression of Feuptake genes are not fully understood. With Next Generation Sequencing technologies, high-throughput RNA sequencing (RNAseq) was conducted to examine gene expression in neonates of Diabrotica virgifera virgifera (LeConte) (Western Corn Rootworm, WCR) challenged with individual proteins of the binary Bacillus thuringiensis insecticidal proteins, Cry34Ab1 and Cry35Ab1, and the combination of Cry34/Cry35Ab1, which together are active against rootworm larvae. Integrated results of three different statistical comparisons identified 114 and 1300 differentially expressed transcripts (DETs) in the Cry34Ab1 and Cry34/35Ab1 treatment, respectively, as compared to the control. No DETs were identified in the Cry35Ab1 treatment. Putative Bt binding receptors previously identified in other insect species were not identified …
