Heparin and heparin related drugs have remained the anticoagulant of choice for medical and surgical indications. These drugs are comprised of poly-component sulfated chains of varying length and biophysical structure with direct and indirect pharmacological effects. Heparins produce their therapeutic effects by multiple mechanisms involving both the cellular and humoral targets. These drugs represent natural products which are isolated from mammalian tissue rich in mast cells. Currently used heparin and LMWHs are obtained from porcine mucosal tissue. Fibroblast growth factors (FGFs) are involved in various cellular processes such as cell growth,proliferation, differentiation, migration, angiogenesis, wound healing and embryonic development. Human acidic fibroblast growth factor (hFGF1) binds non-selectively to all the four FGF-receptors and is therefore considered as a powerful mitogen with broadest specificity. However, pharmacological applications of hFGF1 are restricted due to the low thermal stability of the growth factor. hFGF1 has low thermodynamic stability under physiological temperatures which leads to impairment of cellular signaling process thereby preventing its potential mitogenic properties. hFGF1 has a heparin binding pocket at the C-terminus which comprises of positively charges residues. …

Fibroblast growth factors are polypeptide members of the FGF family, which to date comprises of at least 22 members. They belong to a group of growth factors and are involved in a variety of cellular processes including wound healing, angiogenesis, differentiation and development (organogenesis). Amongst FGF members, human acidic FGF-1 and basic FGF-2 are the most characterized. FGF-1 and FGF-2 are known to share more than 80% sequence similarity and have an identical structural fold. The results of hematological analyses may be affected by preanalytical factors such as sampling procedure, type and concentration of anticoagulant, or conditions and time of blood storage. In the present study the effects of various concentrations of heparin (0-200 IU/cm3) and different times (2, 24, or 48 h) and temperatures (4 or 22 °C) of blood storage on the values of hematological parameters in Cyprinus carpio were evaluated. Increase in heparin concentration resulted in a decrease in frequency of erythroblasts, a reduction of leukocyte and thrombocyte counts, and a decrease in oxidative metabolic activity of phagocytes. Heparin is a polydispersed sulfated molecule that is part of the family called glycosaminoglycans found in the extracellular matrix and cell surfaces. The management of recurrent pregnancy loss (RPL) still remains a great challenge, and women with polycystic ovarian syndrome (PCOS) are at a greater risk for spontaneous abortion. Treatment with low-molecular-weight heparin (LMWH) has become an accepted treatment option for women with RPL; however, the subgroup of women, who are likely to respond to LMWH, has not been precisely identified. Disodium EDTA used as anticoagulant for common carp blood caused a significant increase and high variability in hematocrit readings comparing to the heparinized samples. Na2EDTA induced erythrocyte swelling, causing cell membrane disruption (hemolysis). The nuclei released from destroyed cells changed shape from oval to round, and underwent gradual swelling and vacuolation, followed by karyolysis. The degree of observed changes was similar at all Na2EDTA concentrations (0.1, 0.5, and 1.0 mg/mL); on the other hand, percentage of abnormal and destroyed cells showed considerable differences among blood sampled from various carp individuals. The Effects Of Anticoagulant Choice And Sample Processing Time On Hematologic Values Of Juvenile Whooping Cranes, Joan Maurer, Betsy Reichenberg, Cristin Kelley, Barry K. Hartup

Blood collected from juvenile whooping cranes (Grus americana) in 2007 and 2008 was divided and placed in either the anticoagulant lithium heparin (LiHep) or tri-potassium ethylenediaminetetraacetic acid (K3EDTA) for diagnostic hematology. Thin smears were prepared from the anticoagulated blood in the field with no delay and in the laboratory after a 4-6-hour delay, and then used to determine differential and total leukocyte counts. Manual heterophil and eosinophil counts were greater in LiHep-treated samples compared to K3EDTA samples (P < 0.05), but there was no difference in the total leukocyte concentration or differential leukocyte counts between anticoagulants based on blood smears prepared with no delay (n = 15). Marked differences were noted in relative heterophil (P < 0.05) and lymphocyte (P < 0.05) counts and total leukocyte (P < 0.05) concentrations of K3EDTA-treated samples processed after the delay (n = 7), suggesting a negative effect on lymphocyte integrity from the anticoagulant. Microscopically, lymphocytes were more intact and easily differentiated from thrombocytes in LiHep-treated samples than K3EDTA, but modest thrombocyte clumping in the LiHep samples was a concern. Either anticoagulant appears adequate for diagnostic hematology in juvenile whooping cranes based on this limited analysis, but blood smears should be prepared immediately under controlled conditions for best results.

Abstract The hyaluronic acid receptor for endocytosis (HARE)/ Stabilin-2 is the primary systemic scavenger receptor for hyaluronan (HA), the chondroitin sulfates (CS), dermatan sulfate (DS), and nonglycosaminoglycan (GAG) ligands such as acetylated low-density lipoprotein (AcLDL), pro-collagen propeptides, and advanced glycation end products. We recently discovered that HARE is also a systemic scavenger receptor for heparin (Hep) (Harris EN, Weigel JA, Weigel PH. 2008. Mycoplasma hyopneumoniae, the causative agent of porcine enzootic pneumonia, colonises the respiratory cilia of affected swine causing significant economic losses to swine production worldwide. Heparin is known to inhibit adherence of M. hyopneumoniae to porcine epithelial cilia. M. hyopneumoniae cells bind heparin but the identity of the heparin-binding proteins is limited. Proteomic analysis of M. hyopneumoniae lysates identified 27 kDa (P27), 110 kDa (P110) and 52 kDa (P52) proteins representing different regions of a 159 kDa (P159) protein derived from mhp494. Reactivity of factor IXa with basic pancreatic trypsin inhibitor is enhanced by low molecular weight heparin (enoxaparin). Previous studies by us have suggested that this effect involves allosteric modulation of factor IXa. We examined the reactivity of factor IXa with several isolated Kunitz-type inhibitor domains: basic pancreatic trypsin inhibitor, the Kunitz inhibitor domain of protease Nexin-2, and the first two inhibitor domains of tissue factor pathway inhibitor. Mycoplasma hyopneumoniae is the causative agent of porcine enzootic pneumonia, a chronic and economically significant respiratory disease that affects swine production worldwide. M. hyopneumoniae adheres to, and adversely affects the function of ciliated epithelial cells of the respiratory tract, and the cilium adhesin (P97), is intricately but not exclusively involved in this process. Although binding of pathogenic bacteria to glycosaminoglycans is a recognised step in pathogenesis, knowledge of glycosaminoglycan-binding proteins is lacking in M. hyopneumoniae.
