Traumatic spinal cord injury (SCI) produces a complex syndrome that is expressed across multiple endpoints ranging from molecular and cellular changes to functional behavioral deficits. Effective therapeutic strategies for CNS injury are therefore likely to manifest multi-factorial effects across a broad range of biological and functional outcome measures. Thus, multivariate analytic approaches are needed to capture the linkage between biological and neurobehavioral outcomes. Injury-induced neuroinflammation (NI) presents a particularly challenging therapeutic target, since NI is involved in both degeneration and repair. Here, we used big-data integration and large-scale analytics to examine a large dataset of preclinical efficacy tests combining five …

Mhcherrypan, A Novel Model To Predict The Binding Affinity Of Pan-Specific Class I Hla-Peptide, Xuezhi Xie

The human leukocyte antigen (HLA) system or complex plays an essential role in regulating the immune system in humans. Accurate prediction of peptide binding with HLA can efficiently help to identify those neoantigens, which potentially make a big difference in immune drug development. HLA is one of the most polymorphic genetic systems in humans, and thousands of HLA allelic versions exist. Due to the high polymorphism of HLA complex, it is still pretty difficult to accurately predict the binding affinity. In this thesis, we presented a new algorithm to combine convolutional neural network and long short-term memory to solve this …

Methods To Integrate Genetic And Clinical Data For Disease Subtyping, Diana Mabel Diaz-Herrera Jan 2020

Methods To Integrate Genetic And Clinical Data For Disease Subtyping, Diana Mabel Diaz-Herrera

Wayne State University Dissertations

Enormous efforts have been made to collect genetic and clinical data from cancer patients to advance the understanding of disease development and progression. Processing and analyzing these flows of data is challenging. Many computational methods have been proposed to help different fronts of biology and medicine. The integration of clinical and genetic data using computational methods towards personalized medicine is considered the future for oncology studies, and this thesis contributes in this direction. This thesis presents three new data integration approaches to elucidate granular and meaningful disease sub-types from high-dimensional complex genetic and clinical variables, which is an essential step …

We share a widely-held vision that tomorrow's health care will be delivered by a distributed team using a knowledge based process that is focused on prevention and wellness. Caregivers will be able to treat and monitor patients where and when needed, capturing data as a natural byproduct of care delivery. Clinical teams with a wide range of skills and expertise will deliver consistent, quality care based on timely, situation-specific knowledge and guidance derived from outcomes evidence. That same knowledge will support advances in healthcare research and education. Home care and remote medicine will increase, reducing the need for high-cost hospitalization. …
