Oxygen activated cold-atmospheric-pressure-argon plasma jet (APPJ) has gained prominence over the regular argon plasma especially in disinfection and decontamination. As an objective of the current research, an oxygen-enriched argon system was built, where plasma produced through a vessel metallic tube that is introduced into alumina one. A sinusoidal high voltage signal of 25 kHz was used to generate plasma jet. Potential impact of oxygen enriched APP jet (Ar/O2) in decontamination of different microbial cells was observed. Escherichia coli (E. coli) are popular fecal indicator bacteria (FIB) used by many beach management programs in Wisconsin to evaluate the health risks posed by beaches. Prolonged survival of E. coli in beach sand undermines beach management practices that rely on FIB enumeration in surface water because sand and water are routinely mixed through wave action. Studies have shown that sand is a favorable environment for E. coli due to temperature, moisture, nutrient content, and protection from UV radiation. Contamination of meat by Escherichia coli (E. coli) can take its source from the live animal or by cross contamination. The study determined the prevalence of resistance of E. coli isolated from locally produced meats in the Tema Metropolis. Beef (n=200), chicken (n=200) and pork (n=200) were randomly selected and evaluated for the presence of E. coli using the procedure in the Bacteriological Analytical Manual of USA-FDA. The disc diffusion method was used for antibiotic susceptibility test of E. coli (n=55) isolates. Locally produced beef (67%), chicken (41%) and pork (23%) were contaminated by E. coli. The neuronal calcium sensor 1 (NCS-1), an EF-hand Ca binding protein, and Ric-8A coregulate synapse number and probability of neurotransmitter release. Recently, the structures of Ric-8A bound to Gα have revealed how Ric-8A phosphorylation promotes Gα recognition and activity as a chaperone and guanine nucleotide exchange factor. However, the molecular mechanism by which NCS-1 regulates Ric-8A activity and its interaction with Gα subunits is not well understood. Given the interest in the NCS-1/Ric-8A complex as a therapeutic target in nervous system disorders, it is necessary to shed light on this molecular mechanism of action at atomic level. World-wide consumption of fresh-cut produce has seen a dramatic increase over the past few decades. However, fresh-cut produce has been implicated in several illness outbreaks due to contamination with foodborne bacterial pathogens. Post-harvest contamination during handling and processing is considered rare but several influencing factors are often overlooked. Raw, ready-to-eat (RTE) seafood products, such as ceviche, poke, and sushi, have experienced growing demand globally; however, these products have the potential to be contaminated with foodborne pathogens. The objective of this study was to determine the prevalence of Escherichiacoli/coliforms, Salmonella, and Listeria in ceviche, poke, and sushi dishes sold at the retail level in Orange County, CA, USA. Additional organisms detected during testing were also considered in the results. Fecal contamination has continued to plague local communities around the United States and the world, especially in highly populated areas like Southern Florida. Newer techniques are beginning to be used to track microbes properly as they are found in waterways. Bacteriophages are the natural enemy of bacteria. They infect and kill bacterial cells similarly to how animal viruses infect and kill our (eukaryotic) cells. Being viruses, bacteriophages need to get inside of their host to be able to replicate and survive. To do this, phages use receptor binding proteins (RBPs) to bind to various molecules on the surface of bacterial cells, which allows them to attach and initiate the first step of infection. An under-investigated target of lysine acetylation is the bacterial ribosome. Although lysine acetylations on the ribosome are common and conversed in diverse bacterial species, little work has been done to understand how lysine acetylation affects the bacterial ribosome. The goal of this work is to determine if lysine acetylation has functional impact on the bacterial ribosome.I have identified in vitro and in vivo effects of nonenzymatic, AcP-dependent lysine acetylation on translation and the ribosome. In vitro acetylation of transcription-translation reactions causes a translation-specific defect that is unaffected by the addition of the CobB deacetylase. However, the current production methods for GlcN-based products are not eco-friendly and pose risks to individuals with shrimp allergies. ABC transporters are central in many cellular functions including nutrient uptake, signal transduction membrane assembly, and cellular detoxification. Both structural and functional studies have revealed insights into the high-affinity uptake mechanism of the MetNI methionine ABC transporter. Using the energy from ATP binding and hydrolysis, the MetNI-Q system can import L-Met, D-Met, and other methionine derivatives against concentration gradients. Many mechanistic studies of ABC transporter propose a model in which cognate binding proteins sequester substrates in the periplasm and deliver them to the transporter. The increasing prevalence of antibiotic-resistant bacteria is an emerging problem for humans. Clinical misuse, overuse in agricultural and food settings, and limited numbers of new antibiotics have accelerated the proliferation of antibiotic-resistant bacteria. To confront this threat, scientists must develop new therapeutics that kill these antibiotic-resistant bacteria. In this study, we used Escherichia coli to analyze antibiotic and bacteriophage susceptibility. E. coli is a common, mostly benign, enteric, gram-negative bacteria. E. coli is used as an indicator for water quality to determine if water poses a health risk for pathogens. Past research has shown that E. coli is present in high numbers in freshwater beach sands distinct from fecal pollution events, yet the precise mechanism for their persistence in not well understood. Persistent E. coli populations in sand can resuspend into adjacent water and lead to increased beach closures when no threat is present. With the ever-growing crisis of antibiotic resistant bacteria, bacteriophage provide an important potential alternative therapy to treat infections when antibiotics are ineffective. For this reason, it is important to understand the mechanisms bacteriophage use to enter their bacterial host cell. The cellular envelope of the gram-negative bacterium E. coli contains a protein system known as Tol-Pal, which utilizes energy from the Proton Motive Force to carry out several cellular functions, including some associated with cellular division and outer-membrane stability. Recent studies in this laboratory showed that an extracellular cutinase from A. mediterranei (AmCut) was able to degrade the plastics polycaprolactone and polybutylene succinate. Such plastics can be slow to degrade in soils due to a lack of efficient polyester degrading organisms. AmCut also showed potential for the biocatalytic synthesis of esters by reverse hydrolysis.
The gene for AmCut has an upstream leader sequence whose transcript is not present in the purified enzyme. In this study, we show using predictive modelling, that this sequence codes for an N-terminal signal peptide that directs transmembrane expression via the Sec secretion pathway.

The impacts of management-intensive grazing (MIG) of cattle on concentrations of total Escherichia coli, total suspended solids (TSS), and nitrate-nitrite nitrogen (NO3+NO2-N), and occurrence of E. coli O157:H7 and selected antibiotic resistance genes (ARGs) in stream water and/or sediments were evaluated. Cattle were grazed for twoweek periods in May in each of three years. Overall, grazing increased total E. coli in downstream water by 0.89 log10 MPN/100 mL (p < 0.0001), and downstream total E. coli concentrations were higher than upstream over all sampling intervals. Antimicrobial agents are used for suppressing some bacterial infections during the rearing of poultry. In this study; the antibiotic resistance properties of Extended Spectrum ßeta-lactamase (ESßL), Amp-C, carbapenemase-producing Escherichia coli (E. coli) strains isolated from chicken breast meat were determined by disc diffusion and E-test methods. Additionally, it is aimed to determine antibiotic resistance genes by classical polymerase chain reaction (PCR). In this context, E. coli strains were isolated from 7 of 100 chicken breast meat (fillets) and identified by a rapid identification system (BBL Crystal). During the spring of 2021, the Kansas State University Swine Early Wean Facility (SEW) experienced a notable increase in piglet morbidity and mortality. Piglet diarrhea was observed approximately 2 to 3 weeks post-weaning along with an increase in number of sudden mortalities. Escherichia coli can cause health problems in humans and livestock. It is frequently found in faeces and soil, both of which can contaminate grass harvested for silage-making. To determine the survival of E. coli 0157:H7 during ensilage, and its influence on ensilage, unwilted precision-chop grass was ensiled in laboratory silos with the following additive treatments: (a) no additive, (b) E. coli 0157:H7 (Ec - non-infectious strain, at log10 4.5 colony forming units (cfu) g-1), formic acid (FA), and (d) Ec + FA. The decrease in subtherapeutic antibiotic administration in poultry has increased the need to address production challenges caused by pathogens, such as E. coli. One potential way to improve bird health and reduce bacterial infection is through the addition of litter amendments that absorb moisture. The present study employs a data review on the presence and aggregation of oxytetracycline (OTC) and resistance (AMR) bacteria in wastewater treatment plants (WWTPs), and distribution of the contaminated effluent with the aid of shallow and deep ocean currents. The study aims to determine the fate of OTC, AMR bacteria in seafood, and demonstrate a relationship between AMR levels and human health. This review includes (1) OTC, (2) AMR bacteria, (3) heavy metals in aquatic environments, and their relationship. Few publications describe OCT in surface waters. Formaldehyde fumigation in poultry hatch cabinets has been utilized for sanitation of hatching eggs for over a century. Formaldehyde is utilized to minimize pathogenic microbes on the surface of the egg as well as the microbial bloom during the hatching process. While formaldehyde is effective, its use is regulated in the United States and Europe due to its carcinogenic nature. Formaldehyde has been shown to damage the cuticle of the egg, cause embryonic death, and damage the epithelial lining of the respiratory tract of freshly hatched chicks, predisposing them for respiratory infection. Salmonella is a bacteria most commonly known for causing the eponymous food-related illness. Due to their rapid reproduction rate and their ability to be propogated and maintained in a lab setting, they are commonly used in lab studies so that we can better understand how Salmonella causes disease in organisms that are more difficult to study.
