Purpose. We have previously shown favorable in vitro gut permeability for three novel dipeptide esters of glucosamine (GlcN) likely facilitated by the peptide transporter 1 (PepT1). Herein, we report the development of a novel assay for the determination of bioavailability of the peptide ester of interest, the anti-inflammatory properties of a glycine-valine ester derivative of GlcN (GVG) as well as its pharmacokinetics under healthy and inflammatory conditions.

Objective: Probiotics and prebiotics that affect gut microflora balance and its associated enzymeactivity may contribute to interindividual variation in isoflavone absorption after soy intake, possiblyenhancing isoflavone bioavailability. This study examined the effects of the consumption ofbioactive yogurt (a probiotic) or resistant starch (a known prebiotic) in combination with high soyintake on soy isoflavone bioavailability.Methods: Using a crossover design, chronic soy consumption was compared with soy plusprobiotic yogurt or resistant starch in older male and postmenopausal females (n 31). Research over the past two decades has provided significant epidemiological and other evidence for the health benefits of the consumption of soy-based foods. A large number of dietary intervention studies have examined the effects of soy isoflavones on risk factors for cardiovascular disease and hormone-dependent cancers. However, these report large variability in outcome measures, very limited reproducibility between studies and in some cases, controversy between results of clinical trials using dietary soy or soy protein and isoflavone supplementation. This highlights a major gap in our understanding of soy isoflavone uptake, metabolism, distribution, and overall bioavailability. There are many potential factors …

By identifying the relationship between calcium location in the plant cell and nutrient bioavailability, the plant characteristics leading to maximal calcium absorption by humans can be identified. Knowledge of plant cellular and molecular targets controlling calcium location in plants is emerging. These insights should allow for better strategies for increasing the nutritional content of foods. In particular, the use of preparation-free elemental imaging technologies such as synchrotron X-ray fluorescence (SXRF) microscopy in plant biology may allow researchers to understand the relationship between subcellular location and nutrient bioavailability. The importance of dissolved versus sediment-bound cadmium as uptake routes for the deposit-feeding polychaete Capitella species I and the toxicity and toxicokinetics of cadmium from these exposure routes were investigated. Effects were reported as changes in worm growth rate, egestion rate and allometry. Radioactive cadmium (109Cd) was used as a tracer to examine the uptake (5 d) and subsequent depuration (6 d) of cadmium. Both effects and kinetics were investigated in systems with and without sediment. Individual Capitella sp. One of the largest obstacles in drug development is to overcome solubility and bioavailability problems. Preformulation strategies such as nanoparticle formation are often employed but sometimes create new issues and are limited in their effectiveness and applications. Since the majority of drugs are marketed and sold as solid forms, drug delivery systems are not always desirable. This is where solid-state chemistry becomes important. Traditional solid-state chemistry approaches are often successful but are sometimes too restrictive and cannot be applied to certain compounds. Two aluminum compounds, AlCl_3 and Al(OH)_3, were used to determine the effect of Al compounds on pH and bioavailability of Al in 2 acid soils. Both soils were incubated for periods of 3, 10, and 30 days. Al-tolerant (ET8) and Al-sensitive (ES8) wheat seedlings were used as a testing plant to confirm bioavailability of Al^{3+} in soil solution. Both soils were acidified through the addition of Al compounds to detect the maximum Al^{3+} toxicity level of these compounds. The results showed that the AlCl3 compound increased the bioavailability of Al^{3+} in soil solutions. In contrast, AlCl_3 decreased bulk soil pH. …

Resveratrol (RV) is a naturally occurring phytoalexin of the stilbenoid family produced by some plant species, and present in grape skin, peanuts, and red wine. It has been found to exhibit anti-cancer, anti-inflammatory, anti-viral, anti-aging, cardio protective, and anti-oxidant properties. Bioavailability is a huge setback that limits the potentials of RV. As a result, efforts have been made to design and synthesize RV esters and aliphatic acids in an attempt to increase its bioavailability, solubility in water, and possibly improving its biological activities. Epidemiologic studies have demonstrated a significant association between exposure to particulate matter (PM) and atherosclerosis. Polycyclic aromatic hydrocarbons (PAHs) present in particulate matter, are well known to induce oxidative stress and lipid peroxidation via generation of reactive oxygen species (ROS). Lipid peroxidation involves regulating endothelial nitric oxide synthase via inhibition of its activity, and as a result, mediates dilation of coronary arterioles is involved with the pathogenesis of atherosclerosis. This research combines observation, experimentation, and modeling to evaluate the influence of lakes on dissolved organic matter (DOM) quantity, quality and export in subalpine watersheds of the Sawtooth Mountain Lake District, central Idaho.
First, I conducted an empirical study of the hydrologic and biogeochemical controls on DOM dynamics in stream-lake fluvial networks. I hypothesized that lakes would decrease temporal variability (i.e., buffer) and alter the characteristics of DOM from inflow to outflow. I tested these hypotheses by evaluating DOM temporal patterns and measuring annual export in seven-paired lake inflows and outflows. I then evaluated how ultraviolet (UV) exposure affected DOM …

A laboratory study was conducted to determine the toxicity of indoxacarb and chlorantraniliprole to Eastern subterranean termites, Reticulitermes flavipes (Kollar) (Isoptera: Rhinotermitidae) resulting from topical applications and exposure to treated soil. Soils with varying organic matter (0.57-3.64%) and chemical characteristics were used in termiticide bioassays. Lethal dose resulting from topical application indicated that chlorantraniliprole was two- to 11-foldmoretoxic than indoxacarb. Lethal concentration assays yielded opposite results where concentrations of indoxacarb in soil that caused either 50 or 90% mortality of R. flavipes workers at 48 and 144 h were two- to six-fold lower than chlorantraniliprole. The present study was aimed at providing data to be used at predicting exposure-based effects of 2,4,6-trinitrotoluene (TNT) aged in soil on endpoint organisms representing two trophic levels. These data can be used to define criteria or reference values for environmental management and conducting specific risk assessment. Long-term exposure tests were conducted to evaluate sublethal toxicity and uptake of aged soil-based explosives, with TNT as the main contaminant. In these tests, plants were exposed for 55 d, and biomass and explosives residues were determined. Worms were exposed for 28 and 42 d, and biomass, number, and tissue residues were determined. …

The analysis of dissolved organic matter ( DOM) by Fourier transform ion cyclotron resonance mass spectrometry ( FTICR- MS) has gained wide interest recently, driven primarily by its ultrahigh resolving power and mass accuracy. Accurate calibration of mass spectra is a key step to successfully decipher the DOM components. We propose a simple and accurate method to internally calibrate the peaks in the complex spectra without the need to add a calibrant. The usefulness of two surrogate methods for rapidly determining the bioavailability of PAHs in hydrocarbon-contaminated marine sediments was assessed. Comparisons are made between the PAHs accumulated by the benthic bivalve, Tellina deltoidalis, and the extractable-PAHs determined using a 6-h XAD-2 resin desorption method and a 4-h gut fluid mimic (GFM) extraction method. There were significant positive relationships between PAH bioaccumulation by the bivalves and sediment PAH concentrations. These relationships were not improved by normalising the sediment PAH concentrations to the organic carbon concentration. The lower portion of Tims Branch (TB), a second-order stream system on the Savannah River site (SC, USA), receives influx of mixed waste-contaminated sediments from Steed Pond, a former settling basin for target processing wastes for over three decades. The magnitude and distribution of U, Ni, and other metals and the potential for trophic movement were studied to facilitate risk assessment and determine potential remedial action. Total and sequential extraction of TB soils demonstrated contaminant heterogeneity both spatially and between operationally defined fractions. Metal concentrations were elevated within riparian zone soils in contrast to stream sediments, suggesting off-site transport. Leaf …

For an undergraduate Honor's project, I worked under the direction of Dr. Jeffery Hall to determine the bioavailability and toxicity of iron from Milorganite® fertilizer, an aerobically processed organic fertilizer. The Milorganite® Company is a subsidiary of the Milwaukee Municipal Sewage District and produces a fertilizer of high iron content (approximately 5-7%). The high iron content has resulted in concern about the toxic potential, which until now was unknown. The toxicity of common compounds of lead, cadmium and zinc was evaluated in waters similar to that found in the world's largest lead producing area in Missouri. Static, acute toxicity tests were performed using fathead minnows (Pimephales promelas) and water fleas (Daphnia magna) respectively. Test organisms were subjected to varying amounts of sulfide, carbonate, chloride and sulfate salts of lead, zinc and cadmium mixed in hard, alkaline waters typical to this region. Median lethal concentrations were calculated using nominal versus measured metal concentrations. Two medication formulations are compared using noncompartmental pharmacokinetic (PK) variables. However, more than the ratio of mean effects is of interest. A difference in formulation coeficients of varication (CV), within- or between-subject, is sought. The experimental design chosen is a 2 sequence crossover design of the form ABBA and BAAB, where A and B are two medication formulations. A mixed linear model is defined that contains random effects for subjects and for subject by formulation interactions. Characterizing The Outlines Of Degraded Fine-Particles By Fractal Dimension, Andrew Hunt, David L. Johnson

Fractal dimension has been used extensively as a descriptor of the rugged outlines of fine-particles. Potentially, it may be a useful parameter for characterizing the outlines of fine-particles which have been subjected to some form of chemical degradation. Here, fractal dimension values have been computed for the outlines of microscopic lead fine-particles both before and after weak hydrochloric acid dissolution experiments. Values obtained for the post-dissolution rugged profiles were greater than those of the pristine fracture grains which had a Euclidean form. The profiles of the degraded fine-particles could be characterized by a single fractal dimension value, or they exhibited …

Methods currently used to determine iron bioavailability have disadvantages for both the subjects and researchers involved. One safe and simple method that needs further evaluation, the serum iron absorption method, involves an initial blood drawing, dosages of iron, and blood drawings taken at intervals thereafter. Generally, the rise in serum iron or area under the curve is used to determine iron uptake. Two experiments were conducted using the serum iron absorption method in an effort to improve the utility of this method for the measurement of iron bioavailability. Two Experiments were conducted to investigate the effect of iron status and iron source on iron bioavailability using the rat model. Beef was proportionally mixed with spinach (Experiment I) or soy protein isolate (Experiment II) to test if meat enhances the absorption of iron from spinach or SPI. Five diets with the iron from meat:spinach (or SPI) ratios of 100:0, 75:25, 50:50, 25:75 and 0:100 were prepared and fed to rats. FeS04 was used as a reference. Three separate experiments were conducted to investigate the effects of heat treatment on meat heme and nonheme iron bioavailability in anemic rats. Male weanling Sprague-Drawley rats were first made anemic, then subsequently fed with experimental diets. In the first experiment, dietary iron sources were raw or autoclaved meat mixed with ferrous sulfate to give the ratios of iron from meat and ferrous sulfate equal to 100:0, 75:25, 50:50, 25:75, and 0:100 respectively. Iron bioavailability of diets containing oxidized turkey and oxidized beef meat was investigated in two experiments. In both studies male, weanling rats were made anemic by consumption of a low iron diet and bleeding. The experimental diets were prepared so that meat was the only source of iron. Hemoglobin regeneration served as the basis for measuring iron utilization. In experiment 1, lyophilized, uncooked turkey meat was allowed to oxidize at 20-22°C for 0, 48, 96, 144, 216, or 264 hours and then fed to the rats. Two separate experiments were used to investigate iron bioavailability of mechanically-deboned meats using hemoglobin regeneration to measure iron utilization. In both studies, male weanling rats were made anemic by bleeding and being fed a low-iron diet. Experimental diets were control fed to the animals and final hemoglobin concentrations were recorded. Animals were sacrificed and liver iron concentrations determined. The first experiment measured the iron bioavailability of mechanically-deboned and hand-deboned beef plate was better utilized by rats than the iron from mechanically- deboned shank and mechanically-deboned plate.
