Tropical cloud forest streams are one of the most threatened and understudied ecosystems in the world. Understanding how these ecosystems function is essential for effective conservation. In this study, macroinvertebrate community composition, functional feeding group analysis, ecosystem attributes, and physicochemical parameters were used to evaluate biophysical stream conditions of 3 low-order Neotropical cloud forest streams at Reserva Las Gralarias in Mindo, Ecuador. Additionally, food web structure was analyzed via stable isotope analysis and aquatic insect emergence rate was also examined. The microbiome provides multiple benefits to animal hosts that can profoundly impact health and behavior. Microbiomes are well-characterized in humans and animals in controlled settings, yet assessments of wild bird microbial communities remain vastly understudied. This study examines the multiple factors that affect the microbiome of a burrow-nesting Procellariiform bird species, Leach’s storm-petrel. 16S rRNA-based Illumina Mi-Seq analyses are used to assess the composition and structure of bird and burrow-associated bacterial communities. Results indicate that sex and skin site contribute to bird-associated bacterial community variation, and MHC heterozygosity impacts these bacterial assemblages in a sex and site-specific manner, potentially having …

Thousand Cankers Disease (TCD) is a disease complex involving the fungal pathogen Geosmithia morbida, an insect vector Pityophthorus juglandis, and the hosts, Juglans spp. and Pterocarya spp. Signs and symptoms of TCD include crown thinning due to branch dieback, yellowing and wilting of the leaves, appearance of epicormic shoots, numerous entrance/exit holes, gallery formation by P. juglandis, and the development of small, dark brown cankers underneath the bark. The first experiment was designed to determine the concentration of protected fish oil product (PFO), as GromegaTM, to be added that would sufficiently decrease the polyunsaturated fatty acid (PUFA) ratio in the sows’ milk and colostrum. Of the 3 diets tested (0.25%, 0.5% and 1%) with a control (0% PFO), only the 1% PFO diet had an effect on the PUFA concentrations. The science of identification by bite mark analysis has recently been called into serious question. (Reesu and Brown 2016) Human dentition is truly variable, but often not unique. When animal bites are considered, a proper ID of the perpetrator is nearly impossible. Primary distortion (when the bite is made) and secondary distortion (during decomposition or healing) both further disrupt landmarks that might be used for identification. An increased likelihood of developing obesity-related knee osteoarthritis may be associated with increased peak internal knee abduction moment. Increases in step width may act to reduce this moment. This study focused on how step width influenced the knee joint during stair ascent by healthy and obese participants. Participants ascended stairs while walking at their preferred speed and under one of two step width conditions – preferred and increased. Obese participants experienced greater mediolateral and vertical ground reaction forces (GRFs), as well as increased peak knee extensor moments and push-off peak internal knee adduction moments. In 1996, a crop tree enhancement study involving green ash (Fraxinus pennsylvanica) in a 16-year-old, naturally regenerated, mixed-species, pole-sized, bottomland hardwood stand was initiated at Ames Plantation in West TN. Protoplasts are appropriate targets for genome editing, DNA functional screens and transgenesis. This project focuses on the comparison of inexpensive mesophyll protoplast isolation via the use of food-grade enzymes and transformation between diploid Panicum hallii Vasey (PAH) and polyploid cellulosic feedstock Panicum virgatum L (switchgrass), a relative of PAH. PAH has great potential as a C4 model species for crop and bioenergy research. Here an inexpensive switchgrass and PAH mesophyll protoplast isolation and transformation system was developed; the first protoplast system for PAH. The use of lignocellulosic biomass as a feedstock for integrated biorefining is of current high interest, as separation into its component parts affords process streams of cellulose, hemicellulose and lignin, each of which can serve as a starting point for the production of biobased chemicals and fuels. The objective is to determine the effect of reducing nitrogen input through feeding low rumen degradable protein (RDP) and rumen undegradable protein (RUP) proportions on milk production, nitrogen efficiency and metabolism in heat-stressed cows. Forty-eight mid-lactating, Holstein cows were assigned to treatments using a randomized block design in a 2x2 factorial arrangement of treatments (n = 12/treatment). Livestock projects are an important part of 4-H youth development because they teach life skills like responsibility and hard work. However, many youth are unable to participate or do not have the interest. This study sought to discover what motivates some youth to become involved in a livestock project and why others do not.

The Endangered Species Act (ESA), established in 1973, was a landmark piece of environmental legislation and remains the standard for endangered species conservation. Implementation of the ESA has often been framed as pitting economic development against species conservation, inciting passions for and against endangered species conservation. The strength of opposing public opinions is highlighted by high-profile controversies such as those around the snail darter, northern spotted owl and the greater sage grouse. In the state of Tennessee, statute 54-5-134, cutting hay along controlled access highway right-of-way, gives Tennessee agriculturalists the right to harvest hay along interstate medians and shoulders. Current maintenance is contracted out to private mowing companies and paid for by Tennessee taxpayers. Panicum virgatum (switchgrass), a perennial grass native to North America, is a leading biomass feedstock candidate for the manufacture of cellulosic ethanol. Switchgrass is considered a viable option for biofuel production due to its cheap production cost and ability to grow on marginal land. Biofuel derived from switchgrass has been shown to be very energy efficient, producing 540% more renewable energy versus nonrenewable energy expended. Switchgrass-derived biofuel is also estimated to have greenhouse gas emissions that are 94% lower than emissions from gasoline (Schmer et al 2008). Probiotics die over time during processing, storage and digestion, resulting in reduced health benefits to the consumer. Microencapsulation of microorganisms is an effective way to improve probiotic viability by restricting cell exposure to extreme conditions through the gastrointestinal tract until release in the colon. In this work, appearance and survival of encapsulated probiotic species from two genera was explored. Lactococcus lactis and Bifidobacterium longum were suspended in calcium alginate microbeads by spraying droplets of alginate-probiotic mixture into calcium chloride solution. Cardiac arrest is a prevalent condition with a poor prognosis, attributable in part to persistent myocardial dysfunction following resuscitation. The molecular basis of this dysfunction remains unclear. We induced cardiac arrest in a porcine model of acute sudden death and assessed the impact of ischemia and reperfusion on the molecular function of isolated cardiac contractile proteins. Cardiac arrest was electrically induced, left untreated for 12 min, and followed by a resuscitation protocol. With successful resuscitations, the heart was reperfused for 2 h (IR2) and the muscle harvested. In failed resuscitations, tissue samples were taken following the failed efforts (IDNR). Actin …

In this study, we determined the effect of nutritive sweeteners at 0 to 0.50 moles/kg on lipid oxidation in a model cookie system. Confocal microscopy using Bodipy 493 as a fat soluble dye showed that the fat formed a continuous phase surrounding the starch granules regardless of sugar type. The impact of glucose concentration on lipid oxidation was monitored by lipid hydroperoxides and headspace hexanal during storage at 55°C. Low concentrations of glucose (0.09) were strong inhibitors. At equal molar concentrations, reducing sugars (glucose and fructose) inhibited lipid oxidation, greater than a two months increase in lag phase compared to …

Self-injurious behavior (SIB) is a behavioral pathology seen in a small percentage of humans and non-human primates. In one previous study, macaques with SIB had more sleep disruption than controls, but observations were limited. Two studies were conducted: a baseline study to investigate nighttime activity in rhesus macaques (Macaca mulatta) displaying SIB and controls, and a probiotic study to assess probiotic Bifidobacterium infantis 35624 for high nighttime activity. T. brucei is unique in several ways that distinguish this organism from other eukaryotes. One of the unique features of T. brucei is the organism’s mitochondrial DNA, which is organized in a complex structure called kinetoplast DNA (kDNA). Understanding how the cellular cytoskeleton is maintained and regulated is important to elucidate the functions of many structures such as the mitotic spindle, cilia and flagella. Katanin p60, microtubule-severing enzymes from the ATPase associated with cellular activities (AAA+) family, has previously been shown in our lab to be inhibited by free tubulin as well as α- and β-tubulin carboxy-terminal tail (CTT) constructs. Here we investigate the inhibition ability of several different tubulin CTT sequences. We quantify the effect of the addition of these constructs on the severing and binding activity of katanin. Beginning in the early 1600s, dam construction in New England obstructed anadromous fish access to spawning grounds during migration. As a result, anadromous forage fish populations have declined, which has impacted freshwater, marine, and terrestrial ecosystems. To determine the impacts of dams on anadromous forage fish and freshwater ecosystems, I used historical and current data to estimate population changes in alewives (Alosa pseudoharengus) from 1600-1900. A significant reduction in spawning habitat occurred in New England as a result of 1,642 dams constructed between 1600 and 1900, resulting in 14.8% and 16.6% lake and stream habitat remaining by 1900, …

Over the last century anadromous alewife (Alosa psuedoharengus) and blueback herring (Alosa aestivalis), collectively called river herring, suffered drastic declines throughout their range from Newfoundland (Canada) to North Carolina (USA). A 2011 petition to include river herring in the Endangered Species Act (ESA) was rejected, partly due to inadequate information towards identifying coast-wide population status. Additionally, knowledge gaps were identified with basic ecology of the river herring life cycle in freshwater, including species reproductive strategies.
