Purification And Characterization Of An Antibacterial Substance Produced By Pest-Originated Serratia Marcescens Mm3, Serpi̇l Uğraş, Kazim Sezen, Hati̇ce Kati, Zi̇hni̇ Demi̇rbağ

The aim of this investigation was to determine, purify, and characterize an antibacterial substance produced by Serratia marcescens Mm3 (Sm-Mm3) isolated from a common pest of hazelnut in Turkey, Melolontha melolontha, which is also a significant pest all over the world. A highly active antibacterial substance produced by Sm-Mm3 was purified by ammonium sulfate precipitation and high-performance liquid chromatography, respectively, and identified by spectrometric analysis (TOF-MS). Finally, we showed that this molecule is heat-stable, stable in the pH range of 5-9, and insensitive to organic solvents and proteinase K. The molecular mass was determined as 479 Da. Leuconostoc mesenteroides subsp. cremoris strain W3, isolated from wine, was previously found to produce a bacteriocin with inhibitory activity towards malolactic bacteria and hence inhibit malolactic fermentation in model wine medium. The bacteriocin, which we termed mesentericin W3, was purified by multistep chromatography and found in a previous study to be similar to mesentericin Y105, albeit with a low recovery (3.2%). The effects of physical and chemical sublethal treatments on the antibacterial activity of enterocin KP produced by Enterococcus faecalis KP against Escherichia coli O157:H7 and Salmonella Typhimurium were investigated. Enterocin KP was not active against intact cells of E. coli O157:H7 or S. Typhimurium. However, the use of enterocin KP together with ethylenediaminetetraacetic acid (50 mM), sodium tripolyphosphate (50 mM), sublethal heating (60 °C for 10 min), cold shock (-20 °C for 2 h), or acid stress (mixture of 40% lactic acid, 16% propionic acid, 16% acetic acid) in BHI medium decreased the cell number of E. coli O157:H7 by …
