Metabolic Inhibition Of A Toluene-Enriched Microbial Population Due To Lead (Pb2+): Verification Of A Free Metal Ion Toxicity Model, Patrick J. S. Marbas

A dissolved oxygen probe and an ion specific electrode were used to study the lead-induced metabolic inhibition in a toluene-enriched microbial population. Predicted toxicity values were compared to the actual toxicity responses using a free metal ion toxicity model (TM) which linked metabolic inhibition with lead activity. Experimentally derived values for the model parameters (lead activity and a lead distribution coefficient) were used in the TM. It was postulated that cellular metabolism is disrupted by the conformational changes to the cell's plasma membrane produced by lead ion adsorption. The predicted toxicity values were higher than the actual toxicity response. This …

The effectiveness of management practices in improving quality of runoff from agricultural land areas has been reported based primarily on results from plot- and field-scale studies. There is limited information available on watershed scales, particularly when the dominant agricultural land use is pasture. Experimental autoimmune myasthenia gravis (EAMG) is an animal model of human myasthenia gravis (MG). In mice, EAMG is induced by immunization with Torpedo californica acetylcholine receptor (AChR) in complete Freund's adjuvant (CFA). However, the role of cytokines in the pathogenesis of EAMG is not clear. Because EAMG is an antibody-mediated disease, it is of the prevailing notion that Th2 but not Th1 cytokines play a role in the pathogenesis of this disease. A study was performed to investigate the factors that affect the accuracy of the procedures used by the Virginia Department of Transportation for design of drilled shaft sound wall foundations. Field load tests were performed on eight inch and nine inch diameter drilled shafts, and the results were compared to theoretical solutions for ultimate lateral load capacity. Standard Penetration Tests were run in the field and laboratory strength tests were performed on the soils from the test sites. It was found that published correlations between blow count and friction angle for sands and gravels can be used to estimate friction …

Experimental autoimmune encephalomyelitis (EAE) is an inflammatory autoimmune disease of the central nervous system which serves as a model for the human disease multiple sclerosis. We demonstrate here that encephalitogenic T cells, transduced with a retroviral gene, construct to express interleukin 4, and can delay the onset and reduce the severity of EAE when adoptively transferred to myelin basic protein-immunized mice. Thus, T lymphocytes transduced with retroviral vectors can deliver "regulatory cytokines" in a site-specific manner and may represent a viable therapeutic strategy for the treatment of autoimmune disease.

Lake Mead, Colorado River, Arizona-Nevada, is one of the most heavily used reservoirs in the western United States, providing abundant recreational opportunities as well as downstream domestic and agricultural water for over 22 million users. Based on average nutrient levels and productivity, Lake Mead is classified as mildly mesotrophic. The interflow of the Colorado River dominates the limnology of much of the 106 km-long reservoir, and may still be identified at Hoover Dam under certain conditions. Runoff from land areas amended with poultry (Gallus gallus domesticus) manure can contain elevated concentrations of metals such as Cu, Fe, and Zn. Vegetative filter strips (VFS) can reduce runoff concentrations of animal manure components, but reported studies have typically focused on nutrients and solids rather than metals. This experiment assessed the impact of VFS length (0 to 12 m) on concentrations and mass losses of Cu, Fe, K, Na, Ni, and Zn in runoff from fescuegrass (Festuca arundinacea Schreb.) plots (1.5 m wide × 6 and 12 m long) treated with poultry litter. Site-specific management (SSM, also known as precision farming, precision agriculture, prescription farming, etc.) is a management strategy that seeks to address within-field variability and to optimize inputs such as pesticides and fertilizers on a point-by-point basis within a field. By reducing over-application and under-application of nutrients and pesticides, this strategy has the potential to improve profitability for the producer and also to reduce the threat of groundwater or surface water contamination from agrichemicals. We designed, constructed, and tested a single-stream insect odormeter for the measurement of bovine manure odors. The biosensing element in the odormeter is a group of six gravid face flies (Musca autumnalis DeGeer) in a 360 mL chamber. A microphone, mounted above the chamber, senses sounds made by the flies’ activity. The microphone signal is processed by an analog root-mean-square power-measuring circuit connected to a computer data acquisition system. The apparatus allows electronic measurement of insect activity. Initial experiments demonstrate the feasibility of this system for odor detection (yes/no). Concentrations of indicator bacteria such as fecal coliform (FC) and fecal streptococcus (FS) are often used to assess the suitability of waters for their intended use(s) and to allocate resources for water quality improvement measures. There is evidence, however, that concentrations of FC and FS can be influenced by variables such as season and flow rate during sampling, which could lead to biased results. The objective of this study was to assess the impacts of season and flow rate on concentrations of FC and FS. A new principle of an amperometric actuation technique was incorporated in the ISFET glucose sensor. The ISFET is fabricated by the CMOS process and the platinum working electrode is deposited by the lift-off process. A sensor with a specially designed ladder type working electrode exhibits improved operation in response time, response magnitude and detection range. An expectation concerning the reduction of sensor size is also discussed.

The forces between a silica probe and an n-type TiO2 single-crystal electrode were measured using an atomic force microscope in an aqueous electrolyte solution. These interactions were a strong function of the solution pH, the presence of specifically adsorbed anions, and the TiO2 electrode potential. For a series of pH values, a strong electrostatic repulsion was seen at high pH and decreased as the pH was reduced. At pH values below 5.5, the interaction became attractive. A series of force measurements between SiO2 and n-type TiO2 showed a repulsive interaction when TiO2 was held at negative electrode potentials, which transformed …
