European Fire Ants On Mount Desert Island, Maine: Population Structure, Mechanisms Of Competition And Community Impacts Of Myrmica Rubra L. (Hymenoptera: Formicidae), Jeffrey Garnas
Electronic Theses and Dissertations
In the early to mid-201h century, Mvrmica rubra Latreille established in various communities in Maine, mostly along the coast. Since its establishment, the ant has spread both locally via vegetative colony budding and regionally via human commerce to no fewer than 30 Maine communities, including one inland site. Studies were undertaken in the summer of 2002 to address questions of the ants' population structure in its introduced range by testing for intercolony aggression within and between local infestations. Using captive nests maintained in their original nest soil, M. rubra was tested against its close neighbors, neighbors of lorn within the …
Environmental Conservation On Agricultural Working Land: Assessing Policy Alternatives Using A Spatially Heterogeneous Land Allocation Model, Kelly M. Cobourn
Environmental Conservation On Agricultural Working Land: Assessing Policy Alternatives Using A Spatially Heterogeneous Land Allocation Model, Kelly M. Cobourn
Electronic Theses and Dissertations
Multifunctionality refers to the ability of agricultural systems to produce an array of non-market goods and services in addition to market commodities. This thesis focuses explicitly on the provision of environmental benefits, through reduced soil erosion and fertilizer applications, by agricultural producers. Soil erosion and nutrient contamination from agricultural production are the foremost contributors to ground and surface water degradation in the United States. Reducing their production implies gains in social welfare, but may generate significant private losses to producers. The objective of this analysis is to quantify the tradeoff between environmental improvements and producer welfare and to examine the …

Wetland And Nest Scale Habitat Use By The Four-Toed Salamander (Hemidactylium Scutatum) In Maine, And A Comparison Of Survey Methods, Rebecca J. Chalmers
Electronic Theses and Dissertations
Conserving amphibian populations requires knowledge of a species and its habitat relationships. The four-toed salamander (Hemidactylium scutatum) is listed as Special Concern in Maine and 11 additional states and provinces, Threatened in Illinois, and Endangered in Indiana (Appendix A). Little is known of H. scutatum ecology despite the species' extensive range. Infrequent sightings of H. scutatum throughout its range may indicate either low numbers or that the species' behavior make detection difficult. Records for H. scutatum in Maine existed from only 32 sites before my study, and the total number of occurrences of this species in Maine is …
Characterization Of A Catechol-Type Siderophore And The Detection Of A Possible Outer Membrane Receptor Protein From Rhizobium Leguminosarum Strain Iari 312., Brianne Lee Clark
Characterization Of A Catechol-Type Siderophore And The Detection Of A Possible Outer Membrane Receptor Protein From Rhizobium Leguminosarum Strain Iari 312., Brianne Lee Clark
Electronic Theses and Dissertations
Many gram-negative bacteria produce and secrete siderophores under iron-deficient conditions. Siderophores are low molecular weight compounds (600-1500 Daltons), which chelate ferric iron with an extremely high affinity, and the complex is actively transported across the outer and inner membranes of gram-negative bacteria. There are two main classes of siderophores: catechol and hydroxamate. Catechol-type siderophores chelate ferric iron via hydroxyl groups, and hydroxamate-type siderophores chelate ferric iron via a carbonyl group with an adjacent nitrogen. Rhizobia fix atmospheric nitrogen symbiotically in leguminous plants using the iron-containing enzyme nitrogenase. To satisfy their iron requirements, many rhizobia are known to produce siderophores. Rhizobium …
Biology And Ecology Of Larval Lobsters (Homarus Americanus): Implications For Population Connectivity And Larval Transport, Eric R. Annis
Biology And Ecology Of Larval Lobsters (Homarus Americanus): Implications For Population Connectivity And Larval Transport, Eric R. Annis
Electronic Theses and Dissertations
The connectivity of marine populations and the degree to which they are considered open or closed has important implications for the ecology, management, and resilience of commercially harvested species. Larval exchange is a primary determinant of the level of connectivity between populations, and this thesis examines the intrinsic and extrinsic factors influencing larval transport and the distribution of larvae of the American lobster (Homarus americanus). The potential for larval transport is directly proportional to the planktonic larval duration. Our field data suggest development times in situ were up to three times faster than previous laboratory development times, indicating that potential …
Nest Success And Chick Survival Of Black Terns In Maine: Effects Of Predation On Breeding Productivity, Shane R. Heath
Nest Success And Chick Survival Of Black Terns In Maine: Effects Of Predation On Breeding Productivity, Shane R. Heath
Electronic Theses and Dissertations
Nest predation is a prominent cause of reproductive failure in wetland-nesting birds, including black terns. As a result, predator avoidance should play an important role in nest site selection. I examined intraspecific variation in nest success to identify factors affecting nest predation of black tern colonies in central Maine. I measured variables related to proximity of wetland features and nest aggregation for 231 successful and 124 depredated black tern nests during the period 1998-2002. I defined candidate models based on logistic regression and selected models with Akaike's Information Criterion adjusted for small sample sizes (AICc) to determine the …
Nrage Regulates Life And Death Of Neural Progenitors, Stephen E. Kendall
Nrage Regulates Life And Death Of Neural Progenitors, Stephen E. Kendall
Electronic Theses and Dissertations
The development of the central nervous system requires the orchestration of numerous instructive and permissive cues. These factors are secreted from signaling centers and function in a concentration dependent manner that effects the proliferation, survival and differentiation of neural progenitors (NP) and their differentiated progeny. The family including bone morphogenetic proteins (BMPs) are key regulators of NP expansion and survival. However, once NP become committed to a neuronal fate and begin to differentiate they compete for limited amounts of neurotrophin that facilitate further growth and survival. Here we show that the p75 neurotrophin receptor interacting protein NRAGE is expressed in …

Cellular Immune Response And Gene Expression Profiling In Crohn's Dise, Claudia Romero
Cellular Immune Response And Gene Expression Profiling In Crohn's Dise, Claudia Romero
Electronic Theses and Dissertations
Despite the chronic debate in the etiology of crohn's disease (cd), a debilitating inflammatory bowel disease (ibd) closely related to ulcerative colitis (uc), an emerging interest in a possible mycobacterial role has been marked. Granuloma and pathologic manifestations in cd resemble aspects found in tuberculosis, leprosy and paratuberculosis. The latter, a chronic enteritis in cattle, goat, sheep and primates, which is similar to human enteritis, also known as cd, is caused by a fastidious, slow growing mycobacterium avium subspecies paratuberculosis (map). Due to the similarities between cd and paratuberculosis, a mycobacterial cause in cd has been proposed. Recent discovery of …
Characterization Of Protein Prenyltransferases And Protein Prenylation In Plasmodium Falciparum, Thiago Gaspar Dasilva
Characterization Of Protein Prenyltransferases And Protein Prenylation In Plasmodium Falciparum, Thiago Gaspar Dasilva
Electronic Theses and Dissertations
Malaria kills at least one million people each year, mostly children - a death every 30 seconds. Almost one half of the world population is at risk from malaria. Antimalarial drugs are the only means for the treatment of about 500 million annual global malaria cases. Because of prevalent drug-resistance it is extremely urgent to identify new drug targets. Many proteins involved in eukaryotic signal transduction and cell cycle progression undergo post-translational lipid modification by a prenyl group. Protein prenyltransferases, which catalyze the post-translational prenyl modification, have been established as a target for anticancer therapy. Research done in our laboratory …

Aesthetic Values Of Five Primary Wood Transporting Methods Common To Northern New England, Michael C. Eckley
Electronic Theses and Dissertations
Throughout northern New England and across the country, increasing populations and the exurbanization of rural forested landscapes have had a tremendous impact on forest management. As forested areas become more populated, society has become more exposed to the sights and sounds associated with different forest operations. As a result, aesthetics are increasingly driving public reaction to and concern about forestry practices, especially timber harvesting. How people perceive forestry harvesting can be significant in defining the future of forest management, particularly in more populated woodlands. The objective of this study was to better understanding public values as they relate to timber …

Population Genetics Analysis Of The Grass Shrimp Palaemonetes Pugio Using Single Strand Conformation Polymorphism, Melody A. Flowers
Electronic Theses and Dissertations
Little is known about the genetic structure of grass shrimp, Palaemonetes pugio, populations or the effects the parasite Microphallus turgidus has on it. The objective of this study was to determine the amount of genetic diversity of the shrimp populations based on locality and parasite load. In order to examine the genetics of P. pugio, shrimp DNA was extracted and scanned using single stand conformation polymorphism. Results revealed P. pugio populations are highly conserved among localities. This suggests a significant amount of gene flow is taking place. Results also indicate there is no correlation between the most common host haplotype …

Electronic Theses and Dissertations
Author's abstract: Q-fever is a zoonosis caused by a worldwide-distributed bacterium Coxiella burnetii. Ticks are vectors of the Q-fever agent but play a secondary role in transmission because the agent is also transmitted via aerosols. Most Q-fever studies have focused on farm animals but not ticks collected from dogs in animal shelters. In order to detect the Q-fever agent in these ticks, a nested PCR technique targeting the 16S rDNA of Coxiella burnetii was used. A collection of 450 ticks from the animal shelter were screened via nested PCR and 144 (32%) were positives. The positive PCR products were also …

To gain an understanding of the ecology of drift algae in the Indian River Lagoon system along the east coast of central Florida, four questions were addressed: 1) What is the composition and rate of accumulation of drift? 2) How much movement and turnover occurs within drift accumulations? 3) Do growth rates differ for drift versus attached algae? 4) Is there a difference in photosynthetic performance in drift versus attached algal species? Manipulative field and laboratory experiments were conducted to address these questions with the green macroalga Codium decorticatum and the red macroalga Gracilaria tikvahiae. Changes in pigment concentration and …
Multigene Metabolic Engineering Via The Chloroplast Genome, Oscar Nemesio Ruiz
Multigene Metabolic Engineering Via The Chloroplast Genome, Oscar Nemesio Ruiz
Electronic Theses and Dissertations
The vast majority of valuable agronomic traits are encoded polygenetically. Chloroplast genetic engineering offers an alternate approach to multigene engineering by allowing the insertion of entire pathways in a single transformation event, while being an environmentally friendly approach. Stable integration into the chloroplast genome and transcription of the phaA gene coding for β-ketothiolase was confirmed by Southern and northern blots. Coomassie-stained gel and western blots confirmed hyperexpression of β-ketothiolase in leaves and anthers, with high enzyme activity. The transgenic lines were normal except for the male sterile phenotype, lacking pollen. Scanning electron microscopy revealed a collapsed morphology of the pollen …

Recruitment And Restoration Of The Oyster Crassostrea Virginica In Areas With Intense Boating Activity In Mosquito Lagoon, Florida, Lisa Michele Wall
Electronic Theses and Dissertations
Productivity, diversity and survival of estuaries are threatened by explosive coastal population growth and associated recreational activities. One major area of recreational growth has been the number of small pleasure craft motoring in shallow waters at high speeds. On the east coast of Central Florida in the Indian River Lagoon system, intense boating activity occurs year-round and intertidal reefs of the eastern oyster Crassostrea virginica with dead margins (piles of disarticulated shells) on their seaward edges are commonly found adjacent to major boating channels. The cause(s) of the dead margins is unclear. However, the disarticulated shells may be reducing reef …
Genetic Analysis Of Rhoa Signaling During Epithelial Morphogenesis In Drosophila, Amanda Fitch Leppert
Genetic Analysis Of Rhoa Signaling During Epithelial Morphogenesis In Drosophila, Amanda Fitch Leppert
Electronic Theses and Dissertations
Epithelial morphogenesis is contingent upon cell shape changes. Cell shape changes are the driving force for the metamorphosis of the adult Drosophila leg from the leg imaginal disc precursor. Genetic analysis has identified several Drosophila genes involved in regulating cell shape changes during leg disc morphogenesis. These include members of the RhoA signaling pathway and the product of the Stubble-stubbloid (Sb-sbd) locus, a transmembrane serine protease. Mutations in the Sb-sbd gene interact genetically with the members of the RhoA signaling pathway, however the nature of the relationship between Sb-sbd serine protease activity and RhoA signaling is not understood. To identify …
Extracullular Atp Regulates Il-1beta Release From Microglial Cells Via Purinergic Receptor After In Vitro Trauma, Chengya Liang
Extracullular Atp Regulates Il-1beta Release From Microglial Cells Via Purinergic Receptor After In Vitro Trauma, Chengya Liang
Electronic Theses and Dissertations
Traumatic brain injury (TBI) induces a state of microglialactivation, which includes upregulation of macrophage functions and release inflammatory mediators such as certain inflammatory cytokines. Current literature suggests that interleukin-1Beta is an important cytokine mediator, which is dramatically increased after brain injury. Previous studies indicate that ATP is released by traumatically injured astrocytes and serves as a cell-to-cell mediator through purinergic receptors after in vitro injury. However, the mechanism of interleukin-1Beta release after traumatic brain injury remains poorly defined and is difficult to study using in vivo models. Using an in vitro model for traumatic brain injury (cell strain or stretch), …

Possible Use Of P20 Antigen In Serodiagnosis Of Inflammatory Bowel Disease, Shinchieh Yang
Electronic Theses and Dissertations
Crohn's disease (CD) is an idiopathic, chronic, relapsing inflammatory disorder, which is most commonly involved terminal ileum and colon. The incidence and prevalence of CD has dramatically increased during the last 50 years; however, the etiology and mechanism of this disorder remain unveiled. Besides genetic susceptibility, recent integrated researches investigated the role of environmental triggers such as microflora, measles viruses and mycobacteria in the pathogenesis of CD. The association between M. avium subsp paratuberculosis (MAP) and CD has been heightened because of clinical resemblance to Johne's disease (JD), a granulomatous enteritis in ruminants caused by MAP. Isolation of MAP from …
