Opaque fluids have a limited irradiation transmission. Thus, their decontamination employing near UV irradiation poses significant technical challenges. In the present study, a thin UV-transparent/waterproof glue layer coated with a 1.5 wt% of TiO2 and a new PhotoReactor Cell were implemented. TiO2 irradiation in the PhotoReactor Cell was effected on the TiO2 particle side, not directly in contact with the fluid, allowing the postulation of an “h+” site mobility mechanism on photocatalysis.

The wake of a freely flying European starling (Sturnus vulgaris) was measured using high speed, time-resolved, particle image velocimetry, simultaneously with high speed cameras which imaged the bird. These measurements have been used to generate vector maps in the near wake that can be associated with the bird’s location and wing configuration. A kinematic analysis has been performed on select sequences of measurements to characterize the motion of the bird, as well as provide a point of comparison between the bird of the present study and other birds or flapping wings. Mercury (Hg), as a trace element cycling in the environment, poses a serious health threat to both humans and wildlife due to its toxicity. Atmospheric deposition is the main source of Hg to most remote environments. The Adirondack Park in New York State of the United States receives moderate Hg deposition, and is a region characterized by relatively high concentrations of Hg in the terrestrial and especially aquatic biota. The detection of oligomers and aggregates formed by two amyloid proteins, insulin and amyloid-beta (AB), is of particular importance due to the role which these species play in Diabetes and Alzheimer's disease, respectively. However, existing techniques are limited in the ability to detect insulin and AB; oligomers due to the fact that these early aggregates are transient, present at low concentrations, and difficult to isolate. Improvements must be made to existing techniques or alternative techniques must be explored in order to identify and quantify the size of these oligomeric and aggregate species without disrupting their structure.

Chlorinated aliphatic hydrocarbons (CAHs), such as tetrachloroethene (PCE) and trichloroethene (TCE), are ubiquitously pollutants in aquifer sediments and groundwater due to their heavy usage in industry and inappropriate disposal in the last century. Among about 1300 NPL (National Priorities List) sites, PCE and TCE are the two most frequently detected hazardous contaminants.

Engineered bioremediation, including biostimulation and bioaugmentation, is a promising technology to clean those PCE and/or TCE contaminated sites. This study describes the design, fabrication and applications of a novel thermoelectric microfluidic bio-sensor. The bio-sensor is used for real time detection of the L-glutamate (L-glu) dynamics and metabolism for brain tumor cells immobilized in a microfluidic device. The microfluidic device is fabricated using a polymer/glass laminating technique (Xurography). An antimony-bismuth thin-film thermopile (primary sensing element) is integrated to the microfluidic device. Glutathione (GSH) plays an essential role in the intracellular antioxidant defense against the oxidant radicals, especially the ·OH radical. To understand the early and progressive cellular changes in Alzheimer's disease (AD) development, we investigated reduced glutathione/oxidized glutathione (GSH/GSSG) status in a double mutated AD transgenic mouse model (B6.Cg-Tg), which carries Swedish amyloid precursor protein mutation (APPswe) and exon 9 deletion of the PSEN1 gene. Likewise, S-glutathionylation (Pr-SSG) is a specific post-translational modification (PTM) of cysteine residues by the addition of glutathione. Non-invasive electroencephalography (EEG) based brain-computer interface (BCI) is able to provide alternative means for people with disabilities to communicate with and control over external assistive devices. A hybrid BCI is designed and developed for following two types of system (control and monitor).

An investigation into the effect of moisture augmentation by manipulation of food waste proportion or wastewater treatment plant biosolids proportion was undertaken to determine the effects on production of methane and other biogases from municipal solid waste (MSW). Laboratory microcosm experiments were performed to determine the effect of various proportions of influent waste streams on the production of biogas. Results indicated that moisture augmentation through the addition of food waste to MSW increases the overall bio-gas and hydrogen gas formed during fermentation. Poly-N-substituted glycines (peptoids) are a very versatile family of synthetic molecules that can be customized for any number of applications. In this study, we chose to use peptoids as a foundation for sandwich ELISA microarray analysis with a long term goal of creating an early detection device for complex diseases such as cancer. The peptoids were designed to self-assemble into microspheres to be used in coatings on the surface of the microarray substrates to increase the surface area available for antibody attachment. Methods of molecular biology are commonly applied to investigate the expression and role of these molecular factors. This project presents a robust three-step protocol for examining gene expression in the mouse tibia. The protocol begins with isolating RNA from a flash frozen tibia sample. This study was done to determine the effectiveness of a commercially available bioaugmentation product, BiOWiSHTM-Aqua FOG, for remediating petroleum-contaminated sandy soil. Biodegradation enhancement by BiOWiSHTM-Aqua FOG was evaluated in laboratory microcosms by directly measuring total petroleum hydrocarbon (TPH) and indirectly using respirometry. Attempts were made to enrich hydrocarbon-degrading bacteria in BiOWiSHTM-Aqua FOG, and the resulting enrichment cultures were screened using respirometry as well. Potential hydrocarbon-degrading bacteria in BiOWiSHTM-Aqua FOG were isolated. Team Zeus’ senior project was to design, build and test a working throwing frame system for Bridge II Sports, a nonprofit organization in North Carolina. The throwing frame is used to allow athletes and individuals with disabilities to throw shot-put, discus, and javelin using a frame to sit against. Team Zeus was specifically tasked to build a frame that is lighter weight and more transportable than products that are currently on the market. Stenosis is one of the most common causes for spinal surgery. Laminectomy decompression and fusion are surgical procedures prescribed for this condition. The intention of this work was to investigate the effects of a laminectomy decompression, followed by fusion, on a lumbar functional spinal unit (FSU) through in vitro dynamic (±8Nm at 0.125Hz) and quasi-static (±7.5Nm at 0.1Hz) biomechanical tests, for flexion, extension, bending and rotation motions.

This senior project investigates the relative efficacy of ozonated water and chlorinated water for the purpose of sanitizing freshly-harvested organic produce. The aim of this study was to assist the Cal Poly Organic Farm in deciding whether or not to invest in an ozone generator. The crops used for testing were grown organically in the same field. Carrots and cilantro were harvested and promptly treated with 5 ppm ozone solution or 25 ppm sodium hypochlorite solution. The issue of sediment removal from the San Gabriel Mountains has been a complex issue that has created problems with beach replenishment, habitat destruction and the need to spend millions of dollars at regular intervals to avoid safety hazards. Most recently 11 acres of riparian habitat, including 179 oaks and 70 sycamores, were removed for sediment placement. The risks associated with exposure to radiation make it critical that digital imaging systems give the best possible images for a given dose to the patient. The DQE is the most widely accepted measure of performance and dose efficiency for digital radiography systems, however it is not commonly measured in a clinical environment as part of routine quality assurance. The objective of this thesis was to investigate BiOWiSHTM-Aqua, a commercial dry solid formulation containing a consortium of bacteria and yeast, as a biopesticide for treatment of Botrytis cinerea, a gray mold that affects strawberries. BiOWiSHTM-Aqua was compared with another commercial product specifically used as a fungicide and bacteriocide, Serenade® Garden Disease Control Spray (concentrated Bacillus subtilis strain QST 713). Both laboratory tests as well as in vivo lab tests were conducted. BiOWiSHTM-Aqua results varied widely from plate to plate, regardless of experimental conditions. The heart is a complex electro-mechanical system which is intrinsically and intimately linked to physiology and pathology. Cardiac muscle tissue underlies the dynamics of the heart; understanding cardiac muscle tissue allows insight into the working of the heart at a fundamental level. Indeed, models allow a theoretical understanding of systems which necessarily exceeds that of experiment. A simple anaerobic digestion model has been developed for a continuously-stirred tank reactor (CSTR), which links the specific biogas production rate to the food/microorganism ratio (F/M). The model treats the various microbial populations involved in the sequential biological processes involved in anaerobic digestion as a composite and links the entire biomass specific growth rate directly to the specific biogas production rate. The model was calibrated by determining the specific gas production rate for a range of F/M values using a municipal wastewater seed sludge. This study investigated a series of independent unassisted and device-assisted transfers from a wheelchair to vehicle mock-up and vice versa while simultaneously capturing kinematic, kinetic and electromyographic (EMG) data of impaired volunteers. The study provides a venue for observation and evaluation of upper extremity (UE) joint stresses, muscular force and functional demands associated with transfers in persons with spinal cord injury (SCI) to ultimately prevent UE injury, minimize excessive stress, preserve functionality and limit pain. If people with SCI lose function of their UEs, due to pain and/or degeneration, they must then rely on others for everyday tasks.

This senior project report discusses the design, construction, and evaluation of a portable artificial insemination chute for the use of the Cal Poly Beef Department. The design was based on the suggested modifications from operators of several other chutes currently in use.

The main areas considered for the design were; Ergonomics, cattle safety, factors effecting maximum breeding conception rates, and Portability.

The breeding chute proved superior over traditional chutes in a variety of ways. Improvement occurred in the areas of consideration as follows, user operation was simplified, animal stress was decreased, and maximum breeding efficiency was obtained.

Anthropogenic activities have increased phosphorus (P) loading in tributaries to the Laurentian Great Lakes resulting in eutrophication in small bays to most notably, Lake Erie. Changes to surface water quality from P loading have resulted in billions of dollars in damage and threaten the health of the world’s largest freshwater resource. To understand the factors affecting P delivery with projected increasing urban lands and biofuels expansion, two spatially explicit models were coupled.
