Introduction: Antimicrobial resistance (AMR) is an increasing public health concern for humans, animals, and the environment. However, the contributions of spatially distributed sources of AMR in the environment are not well defined.
Methods: To identify the sources of environmental AMR, the novel microbial Find, Inform, and Test (FIT) model was applied to a panel of five antibiotic resistance-associated genes (ARGs), namely, erm(B), tet(W), qnrA, sul1, and intI1, quantified from riverbed sediment and surface water from a mixed-use region.

The primary nitrite maximum (PNM) is a ubiquitous feature of the upper ocean, where nitrite accumulates in a sharp peak at the base of the euphotic zone. This feature is situated where many chemical and hydrographic properties have strong gradients and the activities of several microbial processes overlap. Near the PNM, four major microbial processes are active in nitrite cycling: ammonia oxidation, nitrite oxidation, nitrate reduction and nitrite uptake. The first two processes are mediated by the nitrifying archaeal/bacterial community, while the second two processes are primarily conducted by phytoplankton. Constraining the role of dust deposition in regulating the concentration of the essential micronutrient iron in surface ocean waters requires knowledge of the flux of seawater-soluble iron in aerosols and the replacement time of dissolved iron (DFe) in the euphotic zone. Here we estimate these quantities using seasonally resolved DFe data from the Bermuda Atlantic Time-series Study region and weekly-scale measurements of iron in aerosols and rain from Bermuda during 2019. Increasing greenhouse gas concentrations in the atmosphere, resulting in climate impacts, are raising concerns over the hydrologic cycle and its effects upon agricultural productivity. If rainfall patterns change, meeting an increased demand for fruits and vegetables will pose a challenge for domestic production regions in the United States (U.S.). Information on potential water supply scarcity in the current production regions provides decision makers with critical information for risk mitigation for future production. We used a hydrologic balance-based model of historic and future water availability to evaluate risk of available irrigation water to support major fruit and vegetable production the US. …

Multi-Element Fingerprinting Of Waters To Evaluate Connectivity Among Depressional Wetlands, Yuxiang Yuan, Xiaoyan Zhu, David M. Mushet, Marinus L. Otte

Establishing the connectivity among depressional wetlands is important for their proper management, conservation and restoration. In this study, the concentrations of 38 elements in surface water and porewater of depressional wetlands were investigated to determine chemical and hydrological connectivity of three hydrological types: recharge, flow-through, and discharge, in the Prairie Pothole Region of North America. Most element concentrations of porewater varied significantly by wetland hydrologic type (p < 0.05), and increased along a recharge to discharge hydrologic gradient. Significant spatial variation of element concentrations in surface water was observed in discharge wetlands. Generally, higher element concentrations occurred in natural wetlands compared to wetlands with known disturbances (previous drainage and grazing). Electrical conductivity explained 42.3% and 30.5% of the variation of all element concentrations in porewater and surface water. Non-metric multidimensional scaling analysis showed that the similarity decreased from recharge to flowthrough to discharge wetland in each sampling site. Cluster analysis confirmed that element compositions in porewater of interconnected wetlands were more similar to each other than to those of wetlands located farther away. Porewater and surface water in a restored wetland showed similar multi-element characteristics to natural wetlands. In contrast, depressional wetlands connected by seeps along a deactivated drain-tile path and a grazed wetland showed distinctly different multi-element characteristics compared to other wetlands sampled. Our findings confirm that the multi-element fingerprinting method can be useful for assessing hydro-chemical connectivity across the landscape, and indicate that element concentrations are not only affected by land use, but also by hydrological characteristics.

Kewaunee County, Wisconsin is an agricultural area dominated by concentrated animal feeding operations and manure fertilized cropland. The objective of this study was to characterize chemical and antibiotic resistance gene (ARG) profiles of 20 surface water locations in Kewaunee County to better understand relationships between agricultural contamination and ARG abundance over one year. Surface water (n = 101) and bed sediment (n = 93) were collected from 20 sites during five timepoints between July 2016 and May 2017. Nitrogen (N) uptake in streams is an important ecosystem service that reduces nutrient loading to downstream ecosystems. Here we synthesize studies that investigated the effects of urban stream burial on N-uptake in two metropolitan areas and use simulation modeling to scale our measurements to the broader watershed scale. We report that nitrate travels on average 18 times farther downstream in buried than in open streams before being removed from the water column, indicating that burial substantially reduces N uptake in streams. Simulation modeling suggests that as burial expands throughout a river network, N uptake rates increase in the remaining open …

A combination of local and regional factors and stochastic forces is expected to determine the occurrence of species and the structure of communities. However, in most cases, our understanding is incomplete, with large amounts of unexplained variation. Using functional groups rather than individual species may help explain the relationship between community composition and conditions. In this study, I used survey data from freshwater lakes and ponds to understand factors that determine the presence of the floating plant functional group in the northeast United States. A common member of the intestinal microbiota in humans and animals is Escherichia coli. Based on the presence of virulence factors, E. coli can be potentially pathogenic. The focus of this study was to isolate E. coli from untreated surface waters (37 sites) in Illinois and Missouri and determine phenotypic and genotypic diversity among isolates. Water samples positive for fecal coliforms based on the Colisure® test were streaked directly onto Eosin Methylene Blue (EMB) agar (37°C) or transferred to EC broth (44.5°C). EC broth cultures producing gas were then streaked onto EMB agar. Forty-five isolates were identified as E. coli …

Dissolved inorganic phosphorus (DIP) concentrations in surface water of vast areas of the ocean are extremely low (<10 nM) and phosphorus (P) availability could limit primary productivity in these regions. We explore the use of oxygen isotopic signature of dissolved phosphate (δ18OPO4) to investigate biogeochemical cycling of P in the Sargasso Sea, Atlantic Ocean. Additional techniques for studying P dynamics including 33P-based DIP turnover time estimates and percent of cells expressing alkaline phosphatase (AP) activity as measured by enzyme-labeling fluorescence are also used. In surface waters, δ18OPO4 values were lower than equilibrium by 3–6%, indicative of dissolved organic phosphorous (DOP) remineralization by extracellular enzymes. Between 2001 and 2009 the Water Corporation constructed three flood-compensating basins on farmland in the Vasse and Sabina river catchments, upstream of in the city of Busselton. The basins are designed to reduce the risk of flooding in urban areas of Busselton by temporarily storing and then regulating runoff during flood events. This report fulfils the reporting requirements of a contract between the Water Corporation and the Department of Agriculture and Food. Dissolved cobalt (dCo), iron (dFe) and aluminum (dAl) were determined in water column samples along a meridional transect (∼31°N to 24°N) south of Bermuda in June 2008. A general north-to-south increase in surface concentrations of dFe (0.3-1.6 nM) and dAl (14-42 nM) was observed, suggesting that aerosol deposition is a significant source of dFe and dAl, whereas no clear trend was observed. for near-surface dCo concentrations. Shipboard aerosol samples indicate fractional solubility values of 8-100% for aerosol Co, which are significantly higher than corresponding estimates of the solubility of aerosol Fe (0.44-45%).
