High-frequency accelerometer recording of key predatory behaviors in vipers: validation and case study with Timber Rattlesnakes (Crotalus horridus)

Tri-axial accelerometers (ACTs) are becoming increasingly common in studies of animal behavior wherein direct observation of subjects in nature is constrained or impossible. ACTs are small (< 1 g) piezo-electric (spring-like) sensors that measure three-dimensional acceleration (upward, downward, and side-to-side) derived from subject motion. When leveraged with advanced machine learning techniques, these data can enable precise automated classification of a wide range of movement-mediated behaviors. Until recently, ACTs were largely reserved for larger-bodied organisms or those most amenable to the temporary external attachment of devices. Ongoing ACT miniaturization has now expanded the breadth of organisms amenable to these methods. This project aims to expand on a recently developed framework for ACT monitoring in wild-ranging snakes, a group that has been mostly overlooked in biologging applications. We are currently conducting extensive captive validation trials for robust model training and testing to enable classification of predatory behaviors, including striking and ingestion of prey items, in Timber Rattlesnakes (Crotalus horridus). Following captive validation, we will translate this method to the field with a population of C. horridus in the lower Piedmont of middle Georgia to evaluate the efficacy of externally attached ACTs for remote and continuous monitoring of …

Contamination of oysters with a variety of viruses is one key pathway to trigger outbreaks of massive oyster mortality as well as human illnesses, including gastroenteritis and hepatitis. Much effort has gone into examining the fate of viruses in contaminated oysters, yet the current state of knowledge of nonlinear virus-oyster interactions is not comprehensive because most studies have focused on a limited number of processes under a narrow range of experimental conditions. A nexus approach contributes to the strategic allocation of resources to secure food, energy, and water for the world population. Integrated models considering the complex interactions across food, energy, and water (FEW) enhance decision-making and strategic planning towards resilience. The U.S. Forest Service has monitored longleaf pine cone production at sites throughout the southeastern United States for over 60 years. Data from the multi-decadal surveys have supported our understanding of the variability of stand-level cone production as it relates to environmental and ecological processes, and more broadly, how longleaf pine operates as a masting species. Cones from longleaf pine are counted each spring using visual surveys that follow a standard protocol. The 2019 Society for Neuroscience Professional 1Development Workshop on Teaching reviewed current tools, approaches, and examples for teaching computation in neuroscience. Robert Kass described the statistical foundations that students need to properly analyze data. Pascal Wallisch compared MATLAB and Python as programming languages for teaching students. Adrienne Fairhall discussed computational methods, training opportunities, and curricular considerations. Walt Babiec provided a view from the trenches on practical aspects of teaching computational neuroscience. Mathew Abrams concluded the session with an overview of resources for teaching and learning computational modeling in neuroscience.

Type IV pili (T4P) are thin, hair-like bacterial appendages composed of protein subunits polymerized into a helical fiber. T4P perform diverse functions such as host cell adhesion, biofilm formation, natural competence, and twitching motility. While T4P are well characterized in Gram-negative bacteria, they have more recently been found in Gram-positive bacteria as well. In this work we aimed to solve the crystal structure of the type IV major pilin protein PilA2 from Clostridium perfringens, the predominant pilus subunit which makes up about 99% of the pilus fiber. Mechanotransduction is the process by which a mechanical stimulus is converted to a cellular signal. This process is heavily influential of cell morphology, differentiation, and behavior. However, altered levels of mechanical stimuli are also found in many pathological contexts. For example, cancerous cells have stiffer surrounding tissue than healthy cells, and research suggests that this alters cell behavior and promotes metastasis. Many wildlife populations are experiencing a variety of environmental pressures due to the direct and indirect consequences of a changing climate. In the northeast, USA, moose Alces alces are declining in large part because of the increasing parasitism by winter tick Dermacentor albipictus, facilitated by high host density and optimal environmental conditions. To test this hypothesis, and better understand the influence of this interaction on the stability of the regional population, we constructed a population viability model using data collected through comprehensive survival and productivity studies in 2002–2005 and 2014–2018 in northern New Hampshire. The contamination of fruits with human pathogens is a reoccurring concern in the fresh produce industry. Atmospheric cold plasma (ACP) is a potential alternate to customary approaches for non-thermal decontamination of foods. In this study, the efficacy of a dielectric barrier discharge ACP system against Salmonella (Salmonella Typhimurium, ATCC 13311; Salmonella Choleraesuis, ATCC 10708) and Escherichia coli (ATCC 25922, ATCC 11775) was explored. To understand how foraging decisions impact individual fitness of herbivores, nutritional ecologists must consider the complex in vivo dynamics of nutrient–nutrient interactions and nutrient–toxin interactions associated with foraging. Mathematical modeling has long been used to make foraging predictions (e.g. optimal foraging theory) but has largely been restricted to a single currency (e.g. energy) or using simple indices of nutrition (e.g. fecal nitrogen) without full consideration of physiologically based interactions among numerous co-ingested phytochemicals. Physiology and Disease is a Biology elective at IMSA that has been developed to be mostly student-centered. Some examples of student projects include modeling heart structure to reflect function and creating LED arduino monitors to measure heart rate. Students also measure their lung capacity and blood pressure to demonstrate correlation of these values with heart rate, and trace the correlation back to neuronal controls. Projects such as these integrate other disciplines such as engineering and conform to NGSS Science and Engineering standards and NGSS Cross cutting Concepts standards. Students take responsibility for their own learning and articulate better on tests.

Finding remnant populations of species that are of conservation concern can be difficult, particularly in aquatic habitats. Models of ecological niches can aid in the discovery of refuges. Remnant populations of native freshwater mussels (unionids) have been found in Lakes Erie and St Clair. Our goals were to predict undiscovered refuges in Lake Ontario based on habitat analysis from Lake Erie and to conduct surveys to test those predictions. We built a presence-only model on environmental data including attributes of the benthic zone and shoreline where mussels occurred in Lake Erie. Historically crop models have been used to evaluate crop yield responses to nitrogen (N) rates after harvest when it is too late for the farmers to make in-season adjustments. We hypothesize that the use of a crop model as an in-season forecast tool will improve current N decision-making. Quantitative measurements of root traits can improve our understanding of how crops respond to soil and weather conditions, but such data are rare. Our objective was to quantify maximum root depth and root front velocity (RFV) for maize (Zea mays) and soybean (Glycine max) crops across a range of growing conditions in the Midwest USA. The fate of bioavailable nitrogen species transported through agricultural landscapes remains highly uncertain given complexities of measuring fluxes impacting the fluvial N cycle. The net primary productivity (NPP), also known as the net ecosystem metabolism, of an estuary is a value indicative of the growth and activity of an estuary’s primary producers, relative to the metabolic activity of its consumers. When NPP is high, estuaries exhibit autotrophic conditions that have the capacity to support fruitful bivalve aquaculture. The gross primary production (GPP) metric is useful in determining trends in the terrestrial carbon cycle. Models that determine GPP utilizing the light use efficiency (LUE) approach in conjunction with biophysical parameters that account for local weather conditions and crop specific factors are beneficial in that they combine the accuracy of the biophysical model with the versatility of the LUE model. Students will test Madagascar hissing cockroach’s capacity as a vector for transmission of microorganisms. By comparing a cockroach exposed to human contact (handled by students) and a cockroach with limited exposure (not handled), students can assess the ability of cockroaches to transmit microorganisms from one location (hands) to another (agar plate where the microorganism will be grown). This will allow students to determine if the Madagascar hissing cockroach, the classroom pet, is a potential vector for microorganisms. Students then will be able to question and relate the concept of insects and objects as vectors for common pathogen transfer.

In response to recent educational imperatives in the United States, modeling and systems thinking have been identified as being critical for science learning. In this paper, we investigate models in the classroom from two important perspectives: (1) from the teacher perspective to understand how teachers perceive models and use models in the classroom and (2) from the students perspective to understand how student use model-based reasoning to represent their understanding in a classroom setting. Soil erodibility is a key factor for estimating soil erosion using physically based models. In this study, a new parameterization approach for estimating erodibility was developed for the Rangeland Hydrology and Erosion Model (RHEM). The approach uses empirical equations that were developed by applying piecewise regression analysis to predict the differences of erodibility before and after disturbance (i.e., wildfire, prescribed fire, and tree encroachment) and across a wide range of soil textures as a function of vegetation cover and surface slope angle. The aims of the original proposed project remain the same, that is, to test the hypothesis that Managed Aquifer Recharge (MAR) for stormwater harvesting is a technically feasible, socially and environmentally acceptable, economically viable, and permittable option for developing new water supplies for arid Western urban ecosystems experiencing increasing population, and climate change pressures on existing water resources. Peptide substrate reporters are fluorescently labeled peptides that can be acted upon by one or more enzymes of interest. Peptide substrates are readily synthesized and more easily separated than full-length protein substrates; however, they are often more rapidly degraded by peptidases. As a result, peptide reporters must be made resistant to proteolysis in order to study enzymes in intact cells and lysates. This is typically achieved by optimizing the reporter sequence in a single cell type or model organism, but studies of reporter stability in a variety of organisms are needed to establish the robustness and broader utility of these …

Improved prediction of optimal N fertilizer rates for corn (Zea mays L.) can reduce N losses and increase profits. We tested the ability of the Agricultural Production Systems sIMulator (APSIM) to simulate corn and soybean (Glycine max L.) yields, the economic optimum N rate (EONR) using a 16-year field-experiment dataset from central Iowa, USA that included two crop sequences (continuous corn and soybean-corn) and five N fertilizer rates (0, 67, 134, 201, and 268 kg N ha-1) applied to corn. Models are valuable tools for explaining and testing systems. Small-scale models can be especially useful for educational purposes. For models to be useful, they have to accurately depict the larger system that they are describing. A novel man-made system, known as an agrivoltaic structure, is being constructed at Biosphere 2 near Oracle, Arizona. The word agrivoltaic is a combination of agriculture and photovoltaics, or solar farming. My research involved creating a small-scale version of this system for educational purposes. The majority of the studies on VO2 kinetics in pediatric populations investigated gender differences in prepubertal children during submaximal intensity exercise, but studies are lacking in adolescents. The purpose of this study was to test the hypothesis that gender differences exist in the VO2 and heart rate (HR) kinetic responses to moderate (M) and heavy (H) intensity exercise in adolescents. Twenty-one healthy African-American adolescents (9 males, 15.8 ± 1.1 year; 12 females, 15.7 ± 1 year) performed constant work load exercise on a cycle ergometer at M and H. The land-atmospheric exchanges of carbon dioxide (CO2) and methane (CH4) are major drivers of global warming and climatic changes. The greenhouse gas (GHG) fluxes indicate the dynamics and potential storage of carbon in terrestrial and wetland ecosystems.
