Spleen stromal cells are critical determinants of dendritic cell (DC) development in spleen. The spleen stromal line, namely STX3, supports DC differentiation in vitro from overlaid bone marrow cells while the lymph node stromal line, namely 2RL22, does not. Here we have characterised the hematopoietic support capacity of each stroma, and analyzed lineage origin of the stromal cell lines by gene profiling using microarrays. Stromal co-culture experiments were performed using bone marrow cells as a source of hematopoietic progenitors. A characteristic immature myeloid-like CD11c+CD11b+CD86+MHC-II−/loB220−CD8α− DC is produced after 14 days in STX3 cocultures, while 2RL22 cocultures produce only monocyte/macrophage-like cells. …

Postharvest surveillance for Vibrio vulnificus by a commercial processing facility was conducted from May 2001 to September 2003. Harvest areas included the Delaware Bay, Long Island Sound and Prince Edward Island. Occurrence followed a seasonal distribution. Low densities were observed in June, increased through August, and became rare by September. Given the ubiquitous nature of marine Vibrios, it was not surprising to find V. vulnificus everywhere, which was investigated. Observations confirmed the importance of strict time/temperature and product handling practices by shellfish harvesters, dealers and processors. Mishandling led to a potentially dangerous and uncharacteristically high V. vulnificus observation.

A Simple Spectrophotometric Streptavidin-Biotin Binding Assay Utilizing Biotin-4-Fluorescein., Mark Waner, David Mascotti

A new assay for biotin binding capacity of Streptavidin (SA) is presented in this work. The assay is based on the large decrease in the extinction coefficient at 493 nm that accompanies binding of biotin-4-fluorescein (B4F) to SA. This decrease is attributed to formation of a charge transfer complex between the B4F-donor and one or more SA residues. We show that one may observe the stoichiometric binding via monitoring the absorbance at 493 nm using either SA or B4F as the titrant. The sensitivity of the assay is at the lower end of similar fluorimetric and photometric assays. Though the …

It is not uncommon to find old-growth giant sequoia trees in their native range that are 1,500 years old and over 15 feet in diameter at breast height. The ability of individual giant sequoia trees to survive over such long periods of time has often been attributed to the species high resistance to disease, insect, and fire damage. Three methods of tanoak (Lithocarpus densiflorus [Hook. & Arn.] Rehd.) control involving the application of the amine or ester form of triclopyr were evaluated in this coast redwood uneven-aged forest management study of herbicides. A cut-stump application with the amine form of triclopyr (Garlon 3A), frill cut with the amine form of triclopyr, basal-bark (outer surface) with the ester form of triclopyr (Garlon 4), and an untreated control were replicated three times. Fire management has always meant fire suppression to the managers of the chaparral covered southern California National Forests. Today, Forest Service fire management programs must be cost effective, while wilderness fire management objectives are aimed at recreating natural fire regimes. A cost-effectiveness analysis has been developed to compare fire management options for meeting these objectives in California's chaparral wilderness. This paper describes the analytical procedure using examples from a study currently being conducted for the Los Padres National Forest, and discusses some preliminary results.

Field studies were conducted for 15 species of Iochroma Benth. and the nested genus Acnistus Schott to quantify the diversity of pollination systems and to assess the potential contribution of pollinator behavior to the persistence of closely related species in sympatry. We combined measures of pollinator visitation and pollen deposition to estimate the importance of major groups of pollinators for each species, and we calculated proportional similarity in the pollinator assemblage among species. In E. coli, RecA polymerizes on ssDNA produced after DNA damage and induces the SOS Response. The RecA-DNA filament is an allosteric effector of LexA auto-proteolysis. LexA is the repressor of the SOS Response. Not all RecA-DNA filaments, however, lead to an SOS Response. To determine habitat characteristics that influence Sciurus niger (Eastern Fox Squirrel) abundance and distribution within a suburban/urban landscape in the midwestern United States, I documented the density and placement of fox squirrel leaf nests in 20 woodlots in the Indianapolis metropolitan area. Marion County, IN. The woodlots varied in size (0.94 to 19.5 ha), approximate age, shape, and degree of isolation from other woodlots and suitable squirrel habitat in the surrounding area. The plant cell wall is a dynamic structure that changes in response to developmental and environmental cues through poorly understood signaling pathways. We identified two Leu-rich repeat receptor-like kinases in Arabidopsis thaliana that play a role in regulating cell wall function. Mutations in these FEI1 and FEI2 genes (named for the Chinese word for fat) disrupt anisotropic expansion and the synthesis of cell wall polymers and act additively with inhibitors or mutations disrupting cellulose biosynthesis. Five treatments in different ratio of cow dung : spent mushroom substrate were prepared as feed materials with four replicates for each treatment namely; 80:20 (T1), 60:40 (T2), 50:50 (T3), 40:60 (T4) and 20:80 (T5). After 7 weeks of vermicomposting, macronutrient elements of vermicompost produced were measured and the process continues with composting when all the earthworms were taken out from the treatments. We evaluated the relative decomposition of residue from two pairs of Bt and non-Bt corn hybrids from different seed manufacturers buried in the root zone of adjacent Bt and non-Bt corn plots over a period of 384 d. This paper reports on the results of a machinability study in the milling of various grades of this material. Three types of samples were studied: 100%, 75% and 50% phenolic samples.
