Galega officinalis L. is a medicinal plant being used as a galactagogue and an antidiabetic in folk medicine. A new effective micropropagation protocol was established from leaf, stem, node, and root explants of G. officinalis for in vitro plant regeneration and callus induction systems. Murashige and Skoog (MS) medium supplemented with 55 various concentrations and 4 different combinations [benzyl adenine (BA)/naphthalene acetic acid (NAA), BA/indole-3-acetic acid (IAA), thidiazuron (TDZ)/indole-3-butyric acid (IBA), and TDZ/IAA] of plant growth regulators was used for the regeneration systems. The present study addressed the successful in vitro seed germination, bulblet regeneration, increased bulblet size, hardening, and acclimatization of Persian lily (Fritillaria persica). Seed germination rate was recorded as 86.7% and 96.7% after 2 and 3 months of cold treatment at 4 °C, respectively. Bulblet explants taken from a germination experiment were cultured on Murashige and Skoog (MS) medium supplemented with 0.5-2.0 mg/L thidiazuron (TDZ). Maximum bulblet regeneration frequency and bulblets per explants were achieved on a medium containing 2.0 mg/L TDZ. In the present study, the effects of three ethylene inhibitors, silver nitrate (AgNO3), silver sulfate (Ag2SO4), and cobalt chloride (CoCl2), on the morphogenic and biochemical responses in CAB-6P and Gisela 6 cherry rootstocks were investigated. In both rootstocks, AgNO3 and Ag2SO4 promoted shoot regeneration while CoCl2 had such an effect only in Gisela 6. The rooting percentage of CAB-6P and Gisela 6 explants was diminished with AgNO3, Ag2SO4, and CoCl2. AgNO3 increased root number (40 μM) and root length (50 μM) in the CAB-6P rootstock while it had an inhibitory effect in Gisela 6. Ag2SO4 and CoCl2 enhanced root length …

An in vitro propagation method was established for both male and female genotypes of lentisk using actively growing shoot tips derived from forcefully lignified shoots. The effects of growth regulators on in vitro morphogenesis were investigated. Since rooting of the regenerated shoots for both genotypes was not achieved, an in vitro micrografting method was developed for the production of plantlets. Moreover, genetic stability of 3-, 6-, and 24-times subcultured clones of both genotypes was assessed and compared with the mother plants using fluorescent-based amplified fragment length polymorphism (AFLP) analysis.
