A well-balanced intracellular iron trafficking in glial cells plays a role in homeostatic processes. Elevated intracellular iron triggers oxidative stress and cell damage in many neurological disorders, including migraine. This study aimed to investigate the effects of glyceryl trinitrate (GTN), on cellular iron homeostasis, matrixmetalloproteinase (MMP)-9, and calcitonin gene related peptide (CGRP) receptor (CRLR/CGRPR1) production in microglia, astrocyte, and meningeal cell cultures. Hephaestin (Hp) is a trans-membrane protein, which plays a critical role in intestinal iron absorption. Hp was originally identified as the gene responsible for the phenotype of sex-linked anaemia in the sla mouse. The mutation in the sla protein causes accumulation of dietary iron in duodenal cells, causing severe microcytic hypochromic anaemia. Although mucosal uptake of dietary iron is normal, export from the duodenum is inhibited. Neoplastic cells need essential metals, such as iron and copper, for cellular functions and rapid growth. Metal chelation and changes in their redox cycle in favor of oxidative stress may be critical for making these cells vulnerable to cell death. The HER2-overexpressing breast cancer cell line, MCF7-HER2, and its vehicle control, MCF7-vec cells, were treated with CuCl2, FeSO4, and Dp44mT. Reactive oxygen species (ROS) production in response to these chemicals was detected with flow cytometry, and cell viability was measured by MTT assay in the cells. Effects of different sodium nitrate and iron concentrations on the production of some metabolites and pigments and the activities of some nitrate assimilation enzymes [nitrate reductase (NR), nitrite reductase (NiR), glutamine synthetase (GS), and glutamate synthase (GOGAT)] were investigated in the growth medium of Spirulina platensis (Gamont) Geitler. For this purpose, S. platensis was cultivated on Zarrouk's medium containing different sodium nitrate concentrations (10-180 mM). A 100 mM sodium nitrate concentration stimulated growth of this organism as well as the production of pigments and other metabolites and enzyme activities.
