The pathogenicity of Pseudomonas syringae is associated with the type III secretion system (T3SS), a complex of proteins assembled in the inner and outer bacterial membranes that traverses the plant cell wall to deliver bacterial proteins into the cytoplasm of plant cells. The effector proteins translocated into the plant cells are called Hops (Hypersensitive response and pathogenicity outer proteins). Bacterial effectors target plant immune proteins to suppress defense responses and enhance bacterial parasitism. Bioassays have been used extensively to assess various toxicity endpoints of drinking water disinfection byproducts (DBPs), but an emphasis on single compounds prevails. In this research, DBP mixtures were assessed using a cytotoxicity test with Chinese Hamster Ovary (CHO) cells and an Ames fluctuation test with Salmonella typhimurium TA-98 and TA-100 with and without S9 rat liver homogenate. Seven whole mixture DBP concentrates were formulated using reconstituted natural organic matter (NOM) extracts from the Upper Mississippi River using scaled disinfectant dosing protocols with free chlorine and monochloramine in the presence and absence of added bromide. These nematodes cause more than $3 billion worth of crop losses each year, in part due to lack of available control tactics, such as nematicides. Soybeans are grown on approximately 1.3 million ha in Arkansas generating an estimated $1.7 billion annually. Foliar diseases on soybean can result in economic losses. Growers spend significant time and money on disease scouting via crop consultants and often (subsequent) fungicide applications. Fungicide trials are often arranged in small plots designs. In these scenarios, spatial variability of foliar disease is minimized. While it is advantageous to minimize variance outside of treatment differences, another limitation with many small plot trials is ample disease pressure or having only lower severity. Foot soundness and udder conformation are essential for longevity in beef cattle. The objective of this study was to investigate the effects of foot angle (FA), claw set (CS), teat size (TS) and udder suspension (US) scores on cowherd performance. Data were analyzed for 1,685 observations on Angus-based cows over a four-year period. At weaning FA, CS, TS, and US were evaluated. Scores are based on nine-point scales. The dissertation describes asymmetric synthesis towards C29-C34 moiety of fragment A of the Antascomicin B and Thermal azole based Claisen rearrangements. In chapter 1, we describes asymmetric synthesis towards C29-C34 moiety of fragment A of the Antascomicin B. The non-immunosuppressant Rapamycin, Ascomycin, and Tacrolimus (FK506), strongly binds with FKBP12, the ligand FKBP12 complexes responsible for immunosuppressive activity. Antascomicin B structurally related to Rapamycin, Ascomycin, and Tacrolimus (FK506), binds strongly to FKBP12, yet does not shown immunosuppressive activity. The work presented in this dissertation explores the structural dynamics in the chloroplast signal recognition particle pathway. Findings include cpSRP shows scanning functionality similar to that in the cytosolic SRP with the ribosome. The intrinsically disordered C-terminal tail of the Albino3 protein has some transient secondary structure. Upon binding to cpSRP43 in solution, separate secondary structure formation was identified in the C-terminal tail of Albino3. Finally, to increase efficiency of analyzing fluorescence time traces for this work, a modular software was produced.

Sequestration of atmospheric carbon in forested lands offsets carbon emissions from other industries. Conversion of private lands, particularly agricultural tracts in marginal areas, to forests can bolster carbon abatement. The United States government agencies administer some voluntary, incentive-based programs to encourage landowners to adopt production practices with positive environmental outcomes. This policy stream can be used to increase transition of marginal agricultural land to forests, thereby creating new carbon sinks. Colon inflammation is one condition closely related with colon cancer initiation. In experiment 1, 80 steers (197.0 kg initial body weight; BW for fall, 116.9 kg for spring), were stocked at 2.45 and 4.1 calves/ha in fall and spring, respectively in 16 tall fescue pastures [fall ergovaline (EV) = 1,475 ppb and spring EV = 1,173 ppb] under 2 treatments, mineral (MIN) (n = 8) and cumulative management (CM) (n = 8). Forage allowance did not differ (P = 0.76) between CM and MIN during fall but differed during spring (P ≤ 0.05, 2.55 vs. 3.22 kg DM/kg BW, for MIN and CM, respectively). One common way organisms deal with stressful environments is through gene expression changes, allowing for stress acclimation and resistance. Variation in stress sensing and signaling can potentially play a large role in how individuals with different genetic backgrounds are more or less resilient to stress. Barnyardgrass, one of the most problematic weeds in rice cropping systems, alone has been found to be resistant to four herbicide sites of action (SOA) in certain populations, leaving growers no options for control. This unsettling fact has led to research on other SOAs that have not previously been used in US rice. Trace minerals, particularly copper, zinc and manganese, play a role in ensuring optimal immune function. To examine the effects of diets containing different levels and sources of trace minerals on the inflammatory response, Cobb 500 broilers were fed four experimental diets containing 5 mg/kg Cu and 45 mg/kg of Zn and Mn or 10 mg/kg Cu and 90 mg/kg Zn and Mn in either the sulfate or hydroxychloride form. Highly conserved throughout evolution, lipins are dual functioning proteins found from yeast to humans. Functioning in the cytoplasm as phosphatidate phosphatase enzymes (PAP), lipins produce diacylglycerol that serves as a precursor for neutral fats and membrane phospholipids. Alternatively, nuclear lipins are responsible for the regulation of metabolic genes. Interestingly, both the mammalian lipin 1 paralog and the single Drosophila Lipin ortholog are highly phosphorylated proteins. Glycoside hydrolases are ubiquitous and one of the most catalytically proficient enzymes known, and thus understanding their mechanisms are crucial. Most research has focused on the interaction of the glycon of substrates and their inhibitors within the active site of glycoside hydrolases. The inhibitors employed to probe these interactions generally had small aglycons (i.e. a hydrogen atom, amidines, small aliphatic groups, or benzyl groups). Protein targeting is a vital cellular function. The signal recognition particle (SRP) pathway is a universally conserved targeting system present in the cytosol and used to co-translationally target many proteins to the inner membrane of prokaryotes and the endoplasmic reticulum of eukaryotes. The chloroplast has a homologous SRP system which post-translationally targets light harvesting chlorophyll binding proteins (LHCPs) to the thylakoid membrane for integration. The chloroplast SRP (cpSRP) is a heterodimer with a 54 kDa subunit equivalent to SRP54 in the canonical pathway. We are using whole genome resequencing to identify chromosomal regions associated with resistance or susceptibility to ascites, a form of pulmonary hypertension syndrome, meat-type chickens. Previous Genome Wide Association Studies (GWAS) based on Single Nucleotide Polymorphisms (SNPs) have identified regions on chromosomes 2, 9 and Z. Despite several GWAS and further genotyping, there are no reliable or potential markers for ascites phenotype. Crop improvement through biotechnology is an integrated effort, incorporating multiple approaches like integration of genes, editing of native genes, and removal of selection marker genes. Before streamlining the protocols, the efficiency and feasibility of the individual approach and their components must be tested. Variation in internal cooked color of ground beef is an economic concern for the ground beef market. Persistent pink color in hamburger patties can lead consumers to the perception of an undercooked product.
