Mosses are a dominant understory component in the Arctic and because of sparse canopy cover, contribute to spectral signals used in remote sensing estimates of various ecologically important characteristics such as productivity, phenology, and vegetation mapping. However, little is known about their contributions to community level spectra or how moisture content influences those spectral signals. Unlike vascular plants, mosses cannot actively regulate moisture content and are highly susceptible to desiccation. Previous research has shown that moss reflectance is sensitive to tissue moisture content. Despite the many governmental and medicinal restrictions created to combat the opioid epidemic in the United States, opioid abuse and overdose rates continue to rise. The development of an aptamer-based voltammetric sensor and biosensor is described in this dissertation. The aim was to develop a low-cost, sensitive, and specific aptamer-based sensor for on-site, label-free determination of codeine and fentanyl in biological fluids. To do this, the surfaces of screen-printed carbon electrodes (SPCE) were modified with gold nanoparticles (AuNPs), followed by the addition of single-stranded DNA aptamers. Microbial biofilms are held together by extracellular polymeric substances (EPS), which can be secreted by many organisms. EPS production can facilitate intercellular communication and inter-guild microbial mutualisms, intraspecific gamete exchange, nutrient sequestration, and desiccation resistance. Benthic microbial mats (periphyton) of the Florida Everglades and other karstic wetlands contain abundant EPS generated by mat-producing filamentous blue-green algae and many other species of mat-dwelling algae, cyanobacteria, and bacteria, including the most abundant Everglades diatom, Mastagloia calcarea. Understanding how organisms allocate limited resources across physiological systems is a major challenge in biology. This study revealed that high energetic demand of electric signals of male electric fish (Brachyhypopomus gauderio) is matched by a metabolic trade-off with other cellular functions. We used thyroxine (T4) to modulate the fish’s signal metabolism, partitioned the energy budget pharmacologically, and measured energy consumption using oxygen respirometry. In males, total energy consumption was unchanged pre- and post-T4 treatment, while signal metabolism rose and the standard metabolic rate fell in an even trade-off. In this dissertation I investigate the biodiversity of marine deep-water crustaceans of the Gulf of Mexico and adjacent waters, focusing on pelagic crustaceans. Taxonomic and molecular techniques were utilized to document adult and larval crustacean specimens to better under their taxonomy, life history, evolutionary relationships and cryptic biodiversity. The use of molecular techniques to study organisms from habitats with limited accessibility provides tremendous potential. Lakes are browning globally from increased inputs of colored dissolved organic carbon (cDOC) driven by climate and land-use change. Browning changes lake ecosystems, especially through reductions in water transparency. I used long-term limnological and plankton records and upper surface sediment diatom assemblages to infer a ~1600-year history of browning from diatom assemblages in a 166 cm core from a subtropical, oligotrophic lake known to experience cyclical browning related to climate oscillations. Tree root systems have evolved multidimensional functioning, specializing in water and nutrient acquisition via different strategies. Root functional strategies vary among tree species and are adapted to the abiotic and biotic soil environment. This dissertation explores how three facets of root system functional strategies – morphology and chemistry, biotic associations with fungi, and respiration rates – vary within and among tree species along environmental gradients. Anxiety-related disorders are some of the most pervasive mental health disorders affecting adult and youth populations. Despite growing evidence of the neurobiology associated with anxiety-related disorders, a consensus on the neurobiological mechanisms of anxiety-related disorders remains to be elucidated. We first provide background literature on the reasoning behind this dissertation in Chapter 1. In Chapter 2, we conducted a neuroimaging meta-analysis on posttraumatic stress disorder to identify convergent structural and functional alterations associated with this anxiety-related disorder among adults. The family Sergestidae provide a unique system for studying the evolution of bioluminescence, with species possessing one of three distinct forms of photophores. This study capitalizes on molecular data to construct a genus-level phylogeny of sergestid shrimp. “Genome skimming” was implemented, capturing mitochondrial genomic data across 19 species. Additional individuals were incorporated through Sanger sequencing of four partial gene regions. The -sergestes group of genera was recovered as non-monophyletic, with the -sergia group of genera being recovered as monophyletic. This dissertation examined the role of medial prefrontal cortex (mPFC) and the hippocampus (HC) in episodic memory, and provides a novel approach to identify the midline thalamus mediating mPFC-HC interactions in humans. The mPFC and HC are critical to the temporal organization of episodic memory, and these interactions are disrupted in several mental health and neurological disorders. The chloroperoxidase secreted from Caldariomyces fumago catalyzes broad spectrum of reactions. The crystallography combined with X-ray diffraction analysis was conducted to reveal recombinant CPO expressed in a modified Aspergillus niger system. Our results indicated that despite functional similarities with wild type CPO, recombinant CPO is over glycosylated with more mannose. Besides, ten iodide ion binding sites were identified in rCPO and six of them were found to be well superimposed on previously reported structure of the wild type CPO. Although fire-adapted ecosystems in Everglades require regular burning to maintain wetland ecosystems, land management and climate-change have altered natural fire-regime. Due to changes in climate and hydrology, historical fire-regimes may become irrelevant. To understand changing fire return intervals, I look at patterns in ecosystem recovery, where fast recovery is indicative of resilience and adaption with an objective of understanding post-fire recovery time in Everglades. I evaluated how post-fire recovery rates were influenced by hydrology and fire-history (1948-2019) by measuring changes in normalized difference vegetation index following fires that burned between 2005-2019 within Everglades. The mammalian high-mobility-group protein AT-hook 2 (HMGA2) is a small DNA-binding protein and consists of three positively charged “AT-hooks” and a negatively charged C-terminal motif. It is a multifunctional nuclear protein linked to obesity, human height, stem cell youth, human intelligence, and tumorigenesis. Previous results showed that HMGA2 is a potential therapeutic target of anticancer and anti‐obesity drugs through inhibiting its DNA‐binding activities. Here a miniaturized, automated AlphaScreen ultra‐high‐throughput screening assay is developed to identify inhibitors targeting HMGA2‐DNA interactions. After screening the LOPAC1280 library, several compounds are identified that strongly inhibit HMGA2‐DNA interactions including suramin, a negatively charged antiparasitic drug. …

Worldwide, many large marine predator populations are in decline. These populations can be difficult to study due to the extensive home ranges and migration patterns often exhibited by these species. Molecular tools are therefore necessary to measure specific parameters on these populations that would otherwise be nearly impossible to obtain. Starting in the late 1800’s orchids were heavily poached, leaving many species to reach critically low numbers. Coupled with habitat loss and urbanization many orchid populations were extirpated in southern Florida. Due to lack of endosperm, orchid seeds are reliant on specific mycorrhizal fungi to obtain nutrients to enable growth and development resulting in very low germination rates in nature. The dinoflagellate Karenia brevis, blooms annually in the Gulf of Mexico, producing a suite of neurotoxins known as the brevetoxins. The cellular toxin content of K. brevis, however, is highly variable between or even within strains. I investigated biochemical differences between high (KbHT) and low (KbLT) toxin producing cultures both derived from the Wilson strain, related to energy-dependent quenching (qE) by photosystem II, and the content of reduced thiols of the proteome. In this dissertation, synthesis and reactivity of (β-iodovinyl)sulfone and β-ketosulfone groups incorporated into purine nucleosides at C8 or C2 positions and DNA incorporation of their 5' triphosphates have been developed. Moreover, synthesis of novel antibiotic arsinothricin (AST) as well as polycyclic aromatic hydrocarbons (PAHs) have been discussed. The 8-(1-iodo-2-tosylvinyl)-2'-deoxyadenosine and 8-(1-Iodo-2-tosylvinyl)adenosine were synthesized employing iodovinylsulfonation of 8-ethynyl precursors with TsNa/I2/NaOAc. As higher education becomes more attainable to all populations, college students from low-income backgrounds are at an increased risk of food insecurity due to the financial burdens that come with the transition to college. Food insecurity on U.S. college campuses ranges up to 59%, quadrupling the average national household food insecurity rate of 14%. In college students, food insecurity is correlated with unhealthy eating, alcohol use, and mental health issues.

Historically, human neuroimaging has studied brain regions “activated” during behavior and how they differ between groups of people. This approach has improved our understanding of healthy and disordered brain function, but has two key shortcomings. First, focusing on brain activation restricts how we understand the brain, ignoring vital, behind-the-scenes processing. In the past decade, the focus has shifted to communication between brain regions, or connectivity, revealing networks that exhibit subtle, consistent differences across behaviors and diagnoses. To successfully navigate the complex visual world, animals must extract relevant information from the deluge of light-carried signals that arrive at their eyes. Early vision filters are passive, energy-saving gates that block out irrelevant signals. The remaining incoming signals are then subject to active filtering by visual attention systems which are energetically expensive, especially for smaller animals, which are subject to similar survival challenges as larger animals.

The coffee ring effect (CRE) phenomenon is originated from the nonuniform solvent evaporation of a sessile droplet deposited onto the flat substrate. Once the evaporation starts, the droplet suspended particles move outwards by the radial flow and concentrate in the edge region of the evaporating droplet, resulting in the formation of the so-called CRE stains. Neuroimaging research has identified brain alterations linked with the human immunodeficiency virus (HIV) that contribute to cognitive declines characterizing the disease. Given cannabis’s (CB’s) anti-inflammatory properties, use prevalence among people living with HIV (PLWH), and impact on neurocognition, my dissertation utilizes a between-groups study design to interrogate separate and interactive effects of HIV and CB on fMRI measures of brain activity. We investigate (1) task-based brain activity at the regional-level, (2) insular resting-state functional connectivity (rsFC) at the circuit-level, and (3) large-scale brain network interactions at the systems-level. Heavy and/or chronic cannabis use has been associated with neurocognitive impairment and decline, often in domains such as memory and executive functioning. On the other hand, exercise has been linked to positive effects on brain and cognitive health across the lifespan, as well as to better substance use outcomes. Despite this, little is known about the ways in which exercise could help prevent or ameliorate adverse cannabis-related outcomes among adolescents.

Plumeria L. (Apocynaceae) is a genus of flowering trees and shrubs that are often cultivated in tropical gardens worldwide. The majority of its species occur in the Greater Antilles with many as single-island endemics. The only comprehensive revision for the genus was done by Robert E. Woodson Jr. (1937) who recognized only seven species and created a ‘Plumeria obtusa complex’ with a plethora of synonyms.
