Soybean Glycine max (L.) Merr., is an important crop grown in South Dakota, with an estimated production of $2.33 billion in 2015. However, soybean production in South Dakota is compromised by four pests, the fungal pathogens Diaporthe longicolla (Hobbs) Santos, Vrandecic, and Phillips and Diaporthe caulivora (Athow and Caldwell) Santos, Vrandecic, and Phillips, the soybean cyst nematode Heterodera glycines Ichinohe (SCN), and the soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae). Additionally, these pest can co-occur in fields within South Dakota and the implications associated with interactions among these pests are unknown. Diverse cropping systems and no-till (NT) are beneficial in improving soil organic carbon (SOC) and other soils properties. The present study was conducted to evaluate the interactions and impacts of long-term tillage and diverse crop rotation systems on soil quality indicators that include a range of soil chemical, physical, hydrological, and microbial properties. From 2015-2017, we evaluated a newly established bighorn sheep (Ovis canadensis) herd in the Deadwood Region of the Black Hills, South Dakota. Our objectives were to 1a) determine annual survival rates, 1b) determine cause-specific mortality, 1c) estimate population size, 2a) assess genetic diversity, 2b) assess disease prevalence, 3) evaluate movement patterns post-release, 4a) evaluate 3rd-order habitat selection, and 4b) estimate herbaceous biomass at foraging sites post-release of translocated bighorn sheep. Irrigation water is scarce. Hence, accurate estimation of crop water use is necessary for proper irrigation managements and water conservation. Satellite-based remote sensing is a tool that can estimate crop water use efficiently. Several models have been developed to estimate crop water requirement or actual evapotranspiration (ETa) using remote sensing. One of them is the Mapping EvapoTranspiration at High Resolution using Internalized Calibration (METRIC) model. Macrophages nonspecifically take up extracellular fluids, solutes and macromolecules by macropinocytosis. Understanding the mechanisms of macropinosome maturation will inform the study of lipid uptake, viral entry, antigen processing and presentation, as well as regulation of cell growth. Colony stimulating factor-1 receptor (CSF-1R) is internalized by small vesicle endocytosis, trafficked to nascent macropinosomes and degraded. These CSF-1R positive macropinosomes mature through a sequence similar to endosomes, progressing from EEA1 and Rab5 to Rab7 positive vesicles before fusing with lysosomes. The first study was conducted on commercial Italian semi-hard cheese samples that were analyzed microbiologically to understand the effect of fast and slow rate of cooling on cheese microflora during ripening process at different temperatures. A cheese plant noticed an active growth of non-starter lactic acid bacteria, especially the heterofermentative bacteria, in ripened cheese blocks, which weren’t fully cooled. For that, three cheese sample sets, each having 8 samples, were received from the cheese plant for microbiological analysis. Remote ischemic postconditioning (rIpost) is a potent vasculoprotective stimulus that is thought to reduce reperfusion injury associated with heart disease. However, certain animal models of cardiometabolic risk factors such as dyslipidemia, diabetes, and hypertension suggest the beneficial effects of rIpost to lessen reperfusion injury may be diminished. Weed control is challenging to farmers who are transitioning from production systems that use synthetic herbicides to organic systems. A 2-year field study examined air-propelled corncob grit abrasion for in-row weed control efficacy and effect on corn yield. Grit was applied based on corn vegetative developmental stages with one (V1, V3 or V5), two (V1 + V3, V1 + V5, or V3 + V5), or three (V1 + V3 + V5) applications. Macrophages contribute to plaque formation in atherosclerosis. Macrophages take up modified low-density lipoproteins and store excess cholesterol and triglycerides in lipid droplet organelles. Evidence of lipid-laden macrophages or “foam cells” is apparent on histology sections of diseased arteries, and this lipid-laden appearance can be recreated in cell culture upon exposure of cultured macrophages to acetylated LDL (Ac-LDL). Under nutrient stress, neutral lipids in lipid droplets are hydrolyzed by lipolysis, autophagy, or both. However, these processes are not well understood in macrophages. To determine the effects of a diet that provides 30% energy from protein with ½ as lean, red meat on risk factors of metabolic syndrome in humans. This pilot study was a 3- month, randomized, control, intervention trial with 33 participants (Beef-Intervention n=18; DASH-Control n=15) who displayed markers of metabolic syndrome. Native grasses and forbs are being considered for planting along South Dakota roadsides to create a favorable habitat for wildlife while also allowing landowners a source of forage and biomass production. South Dakota is host to 54,900 hectares (145,000 acres) of right-of-way that are managed by the South Dakota Department of Transportation. Breeding Bird Survey (BBS) data for grassland bird species has shown the most rapid population decline of any other bird group. Current roadside survey techniques, however, may fall short of providing accurate numbers of rare grassland bird species such as chestnut-collared longspur (Calcarius ornatus), lark bunting (Calamospiza melanocorys), Sprague’s pipit (Anthus spragueii), and Baird’s sparrow (Ammodramus bairdii). We hypothesized that the regrowth in Zea might be controlled by two dominant complementary genes. F1 hybrids were created by crossing Zea diploperennis Iltis, Doebley & R. Swift foxes (Vulpes velox) were reintroduced into Badlands National Park between 2003 and 2006 after being nearly extirpated from South Dakota in the early 1900’s. Genetic analysis provided strong evidence that the reintroduction was successful, but viability analysis indicated the population may be in jeopardy with a high probability of extinction. Recently, the population has declined due to various biotic and abiotic factors (e.g., recent weather patterns, effects of plague [Yersinia pestis], and increased coyote [Canis latrans] numbers). This thesis investigates the economic implication of changing slaughter volume patterns across the cash and contract markets in the US cattle industry. Weekly time series data that spans from November 2002 to December 2015 was used in the analysis. Empirical evidence suggests that Mandatory Price Reporting (MPR) played an important role in changing slaughter volumes across the cash and contract markets. Additionally, price of feeder cattle, cattle quality characteristics (Choice-Select spread and the percentage of animals grading choice or better) among others influence feedlots’ decision to use a particular marketing arrangement.

However, there have been only a few reports of SVA associated with disease in last two decades. Main limitations of dried distillers’ grains with solubles (DDGS) as meal for monogastric animals is the presence of high fiber content, reduced fat content, and poor amino acid balance. This causes a reduced economic value of DDGS for these species due to its low inclusion rates. Landscape homogenization and the degradation of riparian areas has greatly impaired stream ecosystems throughout North America. Conservation programs may repair riparian ecosystems to indirectly improve water quality and instream habitat heterogeneity in hopes to elicit biological responses. However, focused manipulations on isolated stream fragments have rarely achieved biological goals. Prairie streams with restored riparian areas were appraised (chemical, physical, and biological variables) to quantify the indirect effects of prevalent grassland conservation practices on aquatic resources. Premise of the study: Chloroplast primers were developed from genomic data for the taxonomically challenging genus Castilleja. We further tested the broader utility of these primers across Orobanchaceae, identifying a core set of chloroplast primers amplifying across the clade.
Methods and Results: Using a combination of three low-coverage Castilleja genomes and sequence data from 12 Castilleja plastomes, 76 primer combinations were specifically designed and tested for Castilleja. The primers targeted the most variable portions of the plastome and were validated for their applicability across the clade. This research quantifies risk reduction and performance of the producer accumulator contract in corn and soybean markets. To quantify performance, we use three alternative theoretical pricing models to estimate historical producer accumulator contract specifications in corn and soybean markets. We then compare the performance of the producer accumulator to eight alternative agricultural marketing strategy portfolios that are also used in new generation grain contracts. The spotted wing fruit fly (SWD), Drosophila suzukii (Matsumura) (Diptera: Drosophilidae), is a pest of small fruit crops. Unlike most other Drosophila species, this insect can oviposit into ripe fruits, rendering them unmarketable. Drosophila suzukii is spreading quickly throughout the continental United States including South Dakota, and causing serious damage to horticultural crops, particularly those within the fruit industry. This study determines the D. suzukii host plant both commercial crops and native plants, and defines non-crop host plants by season in South Dakota. Soybean, a leguminous plant and rhizobia establishes symbiotic relationships, forming root nodules. Nodule organogenesis starts with cortical cell division forming infection and parenchyma zones which houses N-fixing bacteria and block O2 for nitrogenase enzyme function, respectively. The mechanism by which root cortical cells give rise to two structurally and functionally different nodule tissues is not known. One approach to address this knowledge gap is to evaluate global gene expression patterns in these two tissue types during nodule development.
