Rainbow Trout Oncorhynchus mykiss are routinely stocked in Black Hills streams and reservoirs to enhance angling opportunities for the public, however in most cases, hatchery-reared Rainbow Trout do not successfully recruit to establish natural populations. One exception is the Deerfield Reservoir system, where it is estimated that up to 25% of the Rainbow Trout population consists of naturally produced, wild Rainbow Trout. While recruitment of wild Rainbow Trout to the Deerfield Reservoir fishery does occur, annual stockings of 12,000 hatchery Rainbow Trout have continued. Catastrophic flooding of the Missouri River in 2011 has had lasting effects on floodplain habitats (i.e. floodplain lakes) and side-channel habitats (e.g. canals, side-channel embayments, stilling basins, and tributaries) in Lake Sharpe, SD. Floodplain and side-channel habitats are rare habitat in Lake Sharpe, a mainstem Missouri River reservoir, and are thought to be crucial habitat for prey and sport fish. Hipple Lake, the only warm-water floodplain embayment in Lake Sharpe, is in danger of losing connectivity to the reservoir because of sedimentation resulting from the 2011 flood. Nitrogen (N) is a vital factor of corn (Zea mays) production. Previous work in South Dakota has shown that there is uncertainty as to whether nitrogen requirements are the same for corn raised under no-till (NT) versus conventional till (CT) production systems. The objective of this study was to evaluate whether N requirements continue to be greater under long-term NT versus CT production systems in southeastern South Dakota, while also considering effects from cover crops and crop rotation. Studies were conducted to determine the effectiveness of ultrasonication and hydrodynamic cavitation, in a continuous mode, alone and in combination with thermal treatments, on heat resistant aerobic sporeformers and spores in skim milk, and skim milk concentrate. The first study was conducted to investigate the effectiveness of a continuous ultrasonication process to reduce thermoduric Bacillus coagulans cells in skim milk. A 25% reduction was obtained by lab pasteurization alone, whereas ultrasonication alone resulted in an inactivation of 92% after 12 passes (80 sec exposure time per pass). Ultrasonication when combined with pasteurization resulted in further increased inactivation to 99.98%. It …

Pediomelum esculentum (commonly prairie turnip) is a perennial legume of the Great Plains, consisting of a deep taproot and large edible tuber, and has served as a nutritious staple in Native American diets. The tuber is capable of storing up to 20 percent protein by weight. P. esculentum is a legume, but not a prominent nodule former; instead, it grows in nitrogen-limited soils and produces large amounts of protein. This suggests the involvement of biological nitrogen fixation. We have investigated the presence of diazotrophic endophytes in P. esculentum. Heart disease is the leading cause of death in the United States. Understanding heart function at the molecular level is critical for developing of more effective treatments. In the cardiac muscle, the thin filament is composed by troponin (Tn), tropomyosin (Tm), and F-actin. It provides Ca2+-dependent regulation of contraction by modulating myosin attachment and force generation in a cooperative scheme. However, this mechanism remains unclear. In the concentration of cheese whey by membrane filtration processes, fouling is a major limiting factor that leads to flux decline and affects product quality. The growth of biofilms on membranes contributes to biofouling. Biofilms are very difficult to eliminate, and will serve as the seed for additional biofilm development and product contamination. The cropping patterns, grain marketing flows, and transportation needs have changed in South Dakota as increased ethanol production has led to a significant increase in corn production and the production of distiller’s dried grains with solubles (DDGS) as a co-product. Cardiomyopathies (CM) are the leading cause of death in America, and can develop from mutations in sarcomeric proteins, leading to altered protein structure and function. Current therapies target upstream signaling pathways to treat the symptoms of heart failure, but are associated with increased mortality by affecting downstream signaling pathways and other muscle types. Rational drug design can develop therapies to treat CM at the protein level. However, a detailed knowledge of how sarcomeric proteins regulate muscle contraction is required. Two studies were conducted to evaluate the effect of limit-feeding heifers distillers grains with solubles (DDGS) with varying forage to concentrate ratios. The effects on growth, feed efficiency, rumen fermentation, nutrient digestibility, metabolic profile and onset of puberty, as well as post trial performance were investigated. First, a 16-wk feeding trial was conducted using 48 Holstein heifers to evaluate effects of dietary treatment on dry matter intake (DMI), average daily gain (ADG), growth performance, rumen fermentation, and nutrient digestibility. To assess accelerometer-derived physical activity and sedentary time from 0 to 24- months in youth in the iCook 4-H program. The iCook 4-H Program was a 5-state, randomized, control-treatment, family-based childhood obesity prevention intervention promoting cooking, eating and playing together. Youth, 9-10 years old, and their main adult meal preparer, participated in the 12-week program followed by monthly newsletters and bi-yearly booster sessions until 24-months. Physical activity and sedentary time were determined for youth who wore an Actigraph GT3X+ accelerometer for 7 days at 0, 4, 12, and 24-months and met defined accelerometer compliance standards. Ferruginous hawks (Buteo regalis) are a grassland and shrubland obligate nesting raptor and prefer lightly grazed pasture or idle areas for nesting. Their population reportedly declines in number if more than 30% of an area is cultivated and they rarely nest in areas dominated by croplands. Golden eagles (Aquila chrysaetos) are long-lived raptors with high nest-site fidelity and relatively low reproductive success. Population trends of golden eagles in western United States are unclear although long-term monitoring of populations shows declines in occupancy and breeding performance. Three studies were conducted to determine production parameters and study specific hypothesis in regard to improving feed efficiency through various forage utilization strategies with or without the inclusion of various supplemented products. The first study evaluated the supplementation of a cobalt-lactate product and its effects on fiber digestibility and milk production parameters when fed to cows consuming a 70% forage diet. The purpose of this research was to study the differences in the quality of 100% whole wheat flour (WWF) when the bran fraction was subjected to mild solvent treatment before reconstitution with the endosperm. The bran treatments included water washing and washing with ethanol solutions (50% and 100%). WWF prepared with untreated bran served as the control. Effects of these treatments on particle size, chemical composition, color, dough rheology, product formulation, and sensory characteristics of 100% whole wheat flour were studied. Census of Agriculture data are evaluated to test for economies of size of South Dakota farms. Economies of size occur when per-unit costs decrease as the size of a firm increases. This research measures technical economies of size, which assume that all firms pay identical input prices and receive identical output prices. The dependent variable of the model is expenses per dollar of sales. This is a typical measure of efficiency in agricultural statistics. To examine the relationship between baseline, post, and follow-up accelerometer derived physical activity (PA) in 9-10 year old children participating in iCook 4-H, a 16-week, 5- state, randomized control family centered childhood obesity prevention program. This family focus intervention was designed to promote PA through lifestyle modifications. Between group differences in minutes-per-day and intensity of PA were determined on accelerometer compliant participants. The overall objectives of the six studies were to increase the feed efficiency in lactating dairy cows. The first study evaluated the efficacy of different water sources and treatment systems on ruminal parameters using an in vitro gas production system. Outcomes of the study suggested that the source and nutrient quality of water can affect rate of ruminal fermentation. Thus, use of real farm water being offered to the cow to conduct in vitro gas production measurements may accurately predict the ruminal fermentation, digestibility and total gas production. This research has been structured into 3 major parts. The first part evaluates a centrifugal evaporator to concentrate micellar casein concentrate (MCC). The second part evaluates a wiped film evaporator (WFE) to concentrate MCC. The third part evaluates heat stability of the high concentration MCC produced by WFE. In part 1, the primary objective was to achieve >25% total solids (TS) without a loss of water removal rate associated with high solids materials. New corn hybrids have been developed by Masters Choice (MC) that vary in energy density and starch digestibility. These MC hybrids have lower starch densities due to an altered starch structure, which allows for greater ruminal and intestinal starch digestion. The altered starch structure of these new hybrids results in reduced feed manufacturing costs (i.e., grinding). Wetlands in Stutsman County, North Dakota were revisited after 50 years to assess changes in wetland conditions and plant communities within wetland zones in the Prairie Pothole Region. In 1961-1966, Robert E. Stewart and Harold A. Kantrud conducted a study to investigate the relationship of wetland plant communities to variations of water permanence and chemistry on three study areas: Crystal Springs, Cottonwood, and Mt. Moriah. Within in this region, a severe drought occurred in 1988- 1992 followed by the longest deluge starting in 1993 with wetlands still impacted today. Atherosclerosis is a serious disease affecting a large portion of the world’s population. In atherosclerosis, macrophages become filled with lipid droplets and form fatty plaques in artery walls. The formation of these atherosclerotic plaques is dependent on the macrophage growth factor colony stimulating factor-1 (CSF-1); however, understanding the changes in CSF-1 signaling and related pathways in foam cells remain poorly investigated. Lipid-laden macrophages were formed through exposure of murine bone marrow derived macrophages to 100 μg/ml acetylated LDL for 24 h. Weed management is the main yield-limiting factor in production of organic crops. Currently, organic producers have limited resources available for in-row weed management. Flame weeding has been recognized for its potential as one part of an integrated approach at weed management in organic systems. Flame weeding uses propane fueled torches to heat plant tissue and denature proteins, thus injuring or destroying the affected tissue. Preovulatory estradiol has been reported to play a critical role in pregnancy establishment and embryonic survival, but the mechanism by which estradiol exerts its effects has not been well characterized. The objective of this thesis is to determine the effects of preovulatory estradiol on the uterine environment and conceptus survival from fertilization to maternal recognition of pregnancy. Beef cows/heifers were synchronized with the CO-Synch protocol and AIed (d 0). The objective of this research was to determine the effects of increases in energy available for gain from increased small intestinal starch digestion (SISD) and dietary lipid source and amount on the efficiency of N use in cattle. Five ruminally, duodenally, and ileally cannulated steers were placed in a 5 × 5 Latin square. Each received duodenal infusion of 1.5 ± 0.08 kg/d raw cornstarch and either 0, 30.9 ± 0.59, 62.4 ± 1.16, or 120.4 ± 3.39 g/d Glu, or 387.9 ± 17.47 g/d casein. Casein increased (P = 0.05) SISD.
