At this time there is no NIOSH approved means for bag sampling 1,3-butadiene. An attempt was made to use the application of cold (0°F) to inhibit the reactivity of the gas prior to analysis. The State of California Air Resources Board has an approved method for the gas chromatographic analysis of 1,3-butadiene from stationary sources. The method was utilized in the application of industrial hygiene monitoring of affected workers. The application of cold was found to be ineffective at inhibiting the highly reactive nature of 1,3-butadiene. Analyte losses were too great to classify the method as an accurate measure of …

Soil microorganisms are important sources of the nitrogen trace gases NO and N2O for the atmosphere. Present evidence suggests that autotrophic nitrifiers such as Nitrosomonas europaea are the primary producers of NO and N2O in aerobic soils, whereas denitrifiers such as Pseudomonas spp. or Alcaligenes spp. are responsible for most of the NO and N2O emissions from anaerobic soils. It has been shown that Alcaligenes faecalis, a bacterium common in both soil and water, is capable of concomitant heterotrophic nitrification and denitrification. Description, Distribution, And Abundance Of The Blue Crab, Callinectes Sapidus (Rathbun), Spawning Stock Of The Lower Chesapeake Bay, Patrick John Geer

The lower Chesapeake Bay spawning stock of the blue crab, Callinectes sapidus, (Rathbun), is examined for seven years (1986 - 1992) in an attempt to better understand stock behavior. Three methods of post-stratification are used to describe the distribution and movement of the population over time. The three methods, density strata, geographic zones, and depth strata, did well in explaining movements of the population, indicating a trend of increased concentration of blue crabs near the eastern Bay late in the spawning season - October. The data suggest a bimodal period of spawning and a trimodal period of abundance. The …

Age, Growth, And Mortality Of Atlantic Croaker, Micropogonias Undulatus, In The Chesapeake Bay Region, With A Discussion Of Apparent Geographic Changes In Population Dynamics, Luiz R. Barbieri, Mark E. Chittenden Jr., Cynthia M. Jones

Atlantic croaker, Micropogonias undulatus, collected from commercial catches in Chesapeake Bay and in Virginia and North Carolina coastal waters during 1988-1991 (n=1,967) were aged from transverse otolith sections. Ages 1-8 were recorded, but eight-year-old fish were rare. Marginal increment analysis showed that for ages 1-7, annuli are formed once a year during the period April-May. Otolith age readings were precise: >99% agreement within and between readers. Observed lengths-at-age were highly variable and growth rate decreased after the first year. Despite the high variability in sizes-at-age, observed lengths for ages 1-7 fit the von Bertalanffy growth model (r2=0.99; n=753) well. …

Florida Bay is a major nursery for the Caribbean spiny lobster, Panulirus argus. The Bay contains a series of hardbottom or seagrass-covered basins separated by mudbanks, which serve as barriers to water circulation and presumably to transport of postlarvae. Temperatures can fluctuate dramatically in the basins and salinities range from 35 ppt to > 50 ppt. In this study I: i) investigated the physiological tolerance of spiny lobster postlarvae to combinations of temperature and salinity representative of conditions in Florida Bay, and ii) examined how postlarval availability and habitat structure influence spiny lobster recruitment into basins. Mammalian mitochondria contain a unique DNA topoisomerase I with characteristics of both nuclear and bacterial type I DNA topoisomerases. This enzyme, purified from bovine liver, requires cations as cofactors and possesses optimal activity at pH 7.0, 37°C and either 150 mM K+ or Na+ ions. Similar to nuclear enzymes and different from the bacterial enzyme, divalent ions such as Ca2+ or Mg2+ will stimulate but are not essential for the enzymatic activity.

Like the nuclear DNA topoisomerase I, the mitochondrial enzyme is capable of relaxing both negatively and positively supercoiled DNA. The relaxation of negatively supercoiled DNA is …

A non-estrogenic hormone, 6-methoxybenzoxazolinone (6-MBOA), found in young growing grasses, has been shown to be a reproductive initiator in several herbivorous rodents. However, the mechanism by which 6-MBOA initiates reproduction has not been determined. Many believe that 6-MBOA represents a reliable cue that the vegetative growing season has begun, but it is unknown whether this hormone is a primary cueing mechanism or if it works in conjunction with other environmental factors. Two species of bont ticks, Amblyomma hebraeum and Amblyomma variegatum, common to Africa have been studied to compare types and quantities of compounds known or believed to serve as components of the attraction-aggregation-attachment pheromone (AAAP). A complex of attraction, aggregation and attachment pheromones are used by these ticks to detect hosts, mates and perhaps minimize interspecific breeding. Solvent extraction of pheromone emitting ticks followed by Gas Chromatography and Mass Spectrometry revealed little difference in the composition of the AAAP in these two species. A bloom of the dinoflagellate Ptychodiscus brevis occurred off the North Carolina coast in late 1987, and samples were forwarded by NOAA to the Old Dominion University Phytoplankton Laboratory for total phytoplankton analysis. Six sampling stations were chosen to represent different bloom scenarios and these six were clustered into three site groups based on the concentrations of statistically significant taxa. Abundances of phytoplankton taxonomic groups over time were examined in relation to P. brevis concentrations to identify any general tendencies. Grape seed extract is being marketed in France for cardiovascular use and skin care. The active constituents of both these drugs are certain polyphenols called procyanidins. A similar seed extract has now been obtained from Virginia Chardonnay grape seeds. Ten of the major components in this extract have been isolated in their pure form by the semi-preparative HPLC separation on a YMC ODS-AQ (Octadecylsilane-AQ) column. The ODS-Aq column has been shown to have a better resolving capacity compared to the Spherisorb ODS (Octadecylsilane) column previously described in the literature. The perinea! gland in the hispid cotton rat, Sigmodon hispidus, is a seasonally cyclic, subcutaneously located organ which is associated with the reproductive system. On gross dissection, the gland possesses a strong attachment to the penis with only loose fascia! connections to the rectum and surrounding muscle and skin. Histologic exam confirms this connection to the reproductive tract and shows the organ to be a compound tubuloacinar gland with a projection leading into the penis. The purpose of this research was to study the effects of cryopreservation on the proteoglycan (PG) and mineral content of aorta conduit tissue. Proteoglycans from fresh and cryopreserved porcine aorta conduit tissues were isolated by extraction with 4 M guanidine (Gdn) - hydrochloride (HCl) for 48 hours. Quantitative analysis of extracted proteoglycans revealed that the content of proteoglycan material from cryopreserved tissue, measured as uronate and protein per unit weight of wet tissue, was similar to that from fresh tissue. Gel permeation column chromatography studies suggested that proteoglycans present in three CsCl fractions from cryopreserved tissues had molecular weights similar …

Two time- and space-dependent, physical-bio-optical models have been developed for the California Coastal Transition Zone (CTZ) region with the overall objective of understanding and quantifying the processes th at contribute to the spatial and temporal development of nutrient and plankton distributions in the CTZ. The first of these models considers only time- and vertical processes at specific locations in the CTZ. The model food web components include: silicate, nitrate, ammonium, two phytoplankton size fractions, copepods, doliolids, euphausiids and a detritus pool. Permanent plots in the Zuni Pine Barrens (Isle of Wight Co., Virginia) were sampled in order to: quantify plant population dynamics in response to fire, compare these dynamics among different moisture regimes, compare the effect of mechanical understory removal to that of fire on tree seedling and herbaceous dynamics, and determine the effect of dense lichen (Cladonia spp.) coverage on pine seedling establishment and survival. Fire resulted in 40% aboveground mortality in the overstory, 80% in the sapling/large shrub layer, and nearly 100% in the understory, followed by a 3.3 to 10.6 fold increase in understory density. Long-term changes in the macrobenthos of the Southern Branch of the Elizabeth River, Virginia were investigated by comparing the present study of community structure with that described ten years earlier by Hawthorne (1980). The results indicated that dominant organisms remained relatively constant in composition but exhibited considerable fluctuation in density. Differences in diversity and spatial distribution of fauna were also observed relative to the previous decade.

Multivariate statistical analyses indicated the macrobenthos exhibited significant spatial and seasonal patterns. Sediments characterized as silts and clays predominated down river while sandier sediment types were most common farther upriver. The spatial distributions of …

Copia, a Drosophila retrotransposon, is constitutively expressed in all developmental stages, except the embryo in Drosophila melanogaster. The effect of random integration of the copia element results in phenotypic change in Drosophila. The regulatory sequences, controlling copia expression, are located within the 5'LTR. The DNA sequence in the 5'LTR and in the location between downstream of entire the 5' LTR and the initial translation site have been identified by mobilityshift binding assays and DNase I footprinting assays. The effects of temperature, food concentration, salinity and turbidity on the growth and development of Crassostrea virginica larvae were investigated with a time-dependent mathematical model. Formulations used in the model for larval growth are based upon laboratory data. Simulations were done using temperature conditions characteristic of Laguna Madre, Galveston Bay, Apalachicola Bay, North Inlet and Chesapeake Bay. These simulations show that the duration of the planktonic larval phase, which is determined by larval growth rate, decreases at lower latitudes in response to warmer water temperatures. The pearl oyster fishery of Kuwait was monitored daily from January 1989 to May 1990. Landings of pearl oysters in 1989 totaled 287 tons with a market value of U.S. $1.0 million. Commercial pearls (>3 mm) were estimated to be present in one of every 4200 oysters. Most of the pearl oysters landed were new recruits with hinge lengths between 40-56 mm. There was a curvilinear relationship between total weight and size of oysters (length) and the sex ratio approached 1:1. Spawning occurs throughout the year, with a spat settlement peak in early fall. Over the size range examined …

To test the hypothesis that epidermal rather than dermal mosaicism determines Blaschko's lines in hypomelanosis of Ito (HI), we studied the distribution of chromosomal mosaicism in four patients. In two, mosaicism had not been detected in lymphocytes or dermal fibroblasts, but was clearly shown in epidermal keratinocytes; furthermore, the abnormal cell line was confirmed to the hypopigmented epidermis and the normal epidermis contained only normal cells. Negative findings in the other two patients might be because of mosaicism which was undetected either because it was submicroscopic or because it was present in melanocytes, which have not yet been studied. We have studied an inbred family in which two cousins presented with the same clinical features of ataxia telangiectasia (AT). Both patients are still ambulatory at ages 25 and 20. Cellular features of both patients are typical of AT and include increased radiosensitivity and an increased level of spontaneously occurring chromosome aberrations in peripheral blood lymphocytes. Linkage studies and haplotype analysis show no clear evidence that the gene for AT in this family is on chromosome 11q22-23. As previously reported AT families from complementation groups AB, C, and D have all shown linkage to this region of 11q22-23. Our study …

The land-fast sea-ice brine contains a diverse phagotrophic protist assemblage consisting of < 5 mum heterotrophic flagellates, Cryothecomonas spp., heterotrophic dinoflagellates, and heterotrophic and mixotrophic ciliates. Fine-scale horizontal spatial variability is a feature of this assemblage; samples taken within 1 m of each other can be dominated by different heterotrophic protists. Many of the larger heterotrophic protists found in the brine are also found in the water column. The photosynthetic ciliate Mesodinium rubrum is also common. In mid to late austral spring, the heterotrophic assemblage accounts for ca 10% of the total protist biomass in the brine and is dominated by Cryothecomonas spp. This flagellate …

Benthic microalgal chlorophyll a and production were measured at 3 sites at Stellwagen Bank, a cold temperate continental shelf habitat in Massachusetts Bay, USA, during August 1991. Benthic microalgal chlorophyll a averaged 39.8 Mg M-2, vs average integrated phytoplankton chlorophyll a of 25.9 mg m-2. Gross benthic microalgal production, measured by oxygen exchange in clear and opaque benthic chambers, averaged 20.9 mg C m-2 h-1. The seasonal abundance and successional patterns of phytoplankton, including autotrophic picoplankton, are compared to spring, summer, and fall primary production maxima that occurred in three tidal rivers. The tidal freshwaters were dominated by diatoms, chlorophytes, and cyanobacteria during a late spring through early fall period of maximum growth. In contrast, downstream assemblages were dominated by estuarine diatoms, dinoflagellates, and cryptomonads which developed spring pulses and a protracted summer-fall maximum. Autotrophic picoplankton produced a major summer pulse at all river stations with reduced abundance during other seasons. Microzooplankton In The Lower Chesapeake Bay, And The Tidal Elizabeth, James, And York Rivers, Gyung-Soo Park, Harold G. Marshall

Results of a one year study in the lower Chesapeake Bay and three tidal rivers indicate an abundant microzooplankton population, with a mean concentration of 4,231.1/Liter. The most abundant components are the non-loricate ciliates (2,518.2L, 59.5% of the annual total) and tintinnids (1,400.1/L 33.1%). In lesser abundance were the rotifers (191.4/L) and nauplii larvae (121.7/L). Seasonal abundance maxima were highest in summer, followed by fall, spring and winter.

A twelve-month phytoplankton study was conducted in Lake Drummond and Washington and Jericho Ditches from December 1988 to November 1989. Four dominant phytoplankton groups were identified at these sites. These were the Bacillariophyceae, Cyanophyceae, Cryptophyceae and an autotrophic picoplankton component. Over the past 20 years there has been a decrease in the mean pH levels of Lake Drummond and the replacement of one its former major components, the Chlorophyceae, by the Cyanophyceae. Based on water quality analysis results and species diversity indices, Lake Drummond is classified as in an early eutrophic stage of development.

Tautog Tautoga onitis are gaining popularity in Virginia's coastal waters as a recreational and food fish. Adult tautog are seasonally abundant on inshore hard-bottom habitats (1-10 m) and inhabit offshore areas (10-75 m) year-round. Juveniles, especially newly-settled recruits, inhabit vegetated areas in shallow water (usually < 1 m). From March 1979 to July 1986, tautog were collected in lower Chesapeake Bay and nearby coastal waters to examine age, growth, and sexual maturation. Age estimates were determined from annular marks on opercle bones: 82% of the fish were age-10 or younger, 18% exceeded age-10, and 1% were age-20 or older. Marginal increment analysis revealed that annuli formed concurrent with a protracted spawning season (April-July). The von Bertalanffy growth equation, derived from back-calculated mean lengths-at-age, was l(t) = 742 [1-e-0.085 (t-1.816)]. Tautog are long-lived (25+ yr) and attain relatively large sizes (672 mm TL) slowly (K for sexes combined = 0.085). Growth rates of both sexes are similar, although males grow slightly faster (K = 0.090 vs. 0.085 for females). Maturity occurs at age-3 in both sexes. Growth rates for tautog from Virginia are similar to those reported nearly 25 years ago for tautog in Rhode Island. Growth rates for tautog are similar to those of other reef fishes, such as snappers and groupers. Habitat restriction, slow growth, great longevity, and increasing popularity by user groups may contribute to over-exploitation of this species in Virginia waters.

Three different estuarine microenvironments in the lower Chesapeake Bay were investigated for the occurrence and geographical distribution of antimicrobic-producing bacteria. Samples were taken from the water column, fish skin, and algal surfaces. A total of 1472 bacterial isolates were assayed for antimicrobial activity against fungal, Gram-negative and Gram-positive bacterial test organisms. Of the total bacterial isolates assayed, 5. 2% were active antimicrobic producers. Algal surf aces were shown to have a higher incidence of antimicrobic-producing bacteria than the water column or fish skin. Dormancy is an important life-history strategy which allows copepods to increase their fitness by delaying growth and reproduction until harsh environmental conditions have ameliorated. For marine species, the primary strategies identified to date include the production of dormant eggs by shallow-water species, and copepodite overwintering in deep-water species. Herein, we describe a third strategy in which fertilized adult females enter a “reproductive-resting” stage during the late fall that allows them to overwinter and provide a first source of spring naupliar recruitment. This strategy has been observed in the estuarine copepod Coullana canadensis, but may also occur in other species. …
