A number of factors have been proposed to influence within and between species variation in handedness in non-human primates. In the initial study, we assessed the influence of grip morphology on hand use for simple reaching in a sample of 564 great apes including 49 orangutans Pongo pygmaeus, 66 gorillas Gorilla gorilla, 354 chimpanzees Pan troglodytes and 95 bonobos Pan paniscus. Overall, we found a significant right hand bias for reaching. We also found a significant effect of the grip morphology of hand use. Tryptophan 207 Is Crucial To The Unique Properties Of The Human Voltage-Gated Proton Channel, Hhv1, Vladimir V. Cherny, Deri Morgan, Boris Musset, Susan M. E. Smith

Part of the “signature sequence” that defines the voltage-gated proton channel (HV1) is a tryptophan residue adjacent to the second Arg in the S4 transmembrane helix: RxWRxxR, which is perfectly conserved in all high confidence HV1 genes. Replacing Trp207 in human HV1 (hHV1) with Ala, Ser, or Phe facilitated gating, accelerating channel opening by 100-fold, and closing by 30-fold. Mutant channels opened at more negative voltages than wild-type (WT) channels, indicating that in WT channels, Trp favors a closed state. The Arrhenius activation energy, Ea, for channel opening decreased to 22 kcal/mol from 30–38 kcal/mol for WT, confirming that Trp207 …

CrossFit™, a popular high-intensity training modality, has been the subject of scrutiny, with concerns of elevated risk of injury and health. Despite these concerns empirical evidence regarding physiologic stresses including acute oxidative stress is lacking. Therefore, the purpose of this investigation was to examine the acute redox response to a CrossFit™ bout. Furthermore, these findings were compared to a high-intensity treadmill bout as a point of reference. Ten males 26.4 ± 2.7 yrs having three or more months of CrossFit™ experience participated in the present study. The bacterial flagellum contains its own type III secretion apparatus that coordinates protein export with assembly at the distal end. While many interactions among export apparatus proteins have been reported, few have been examined with respect to the differential affinities and dynamic relationships that must govern the mechanism of export. FlhB, an integral membrane protein, plays critical roles in both export and the substrate specificity switching that occurs upon hook completion. Chimpanzees and gorillas are sympatric in most of their range in Central Africa, however, bonobos do not overlap with either of the other two species. This difference has been hypothesized to be the main factor leading to the socio-ecological traits of each species. We have modified the combinatorial selection method Restriction Endonuclease Protection and Selection Assay (REPSA) to work in high temperature conditions for the discovery of new DNA-binding proteins in thermophiles (HT-REPSA). We utilized Thermus thermophilus (HB-8/ATCC 27634/DSM 579) as a test organism due to its amenable nature in a laboratory setting and current status as a model thermophilic organism. We used a TetR Family (TFR) transcription factor SbtR as the model protein for optimization of HT-REPSA protocols, as data had previously been obtained regarding SbtR physical characteristics and DNA-binding properties. Mesograzers have the ability to greatly mitigate the effects of eutrophication in seagrass systems. In this study we look at pinfish (Lagodon rhomboides Linnaeus) as a potential epiphytic grazer and assess feeding preferences during a transitional stage in the ontogenetic diet shift exhibited by these fish. Since pinfish are abundant in seagrass meadows in the northern Gulf of Mexico, their dietary preferences have the potential to greatly impact seagrasses in this system. The highly conserved nuclear protein Akirin was previously identified as a cofactor that modulates Twist transcription factor activity during muscle development in Drosophila melanogaster. Akirin mediates an interaction between the Twist transcription factor and the multisubunit Brahma SWI/SNF-class chromatin remodeling complex at control elements of the Dmef2 locus to maintain optimal myogenic gene expression levels. Therefore, Akirin represents a class of novel secondary cofactors that work with transcription machinery to link transcription factor output with chromatin remodeling machinery to facilitate gene expression. Eph receptor tyrosine kinases and their ephrin ligands are required for multiple aspects of nervous system development including axon outgrowth, synaptic plasticity, and the formation of topographic maps in the visual system. The Caenorhabditis elegans ephrin-A, efn-4, has a defined role in hypodermal patterning but its role in nervous system development is not well understood. We find that loss-of-function mutations in efn-4 lead to suppression of axon branching in C. elegans model of X-linked Kallmann syndrome, a human genetic disorder that presents with loss of sense of smell and failure to undergo spontaneous puberty. Using the tools of environmental history and the history of science, it examines the station’s founding, its fieldwork, and its place within the broader history of malaria control, eradication, and research. Background The C. elegans proteins PTP-3/LAR-RPTP and SDN-1/Syndecan are conserved cell adhesion molecules. Loss-of-function (LOF) mutations in either ptp-3 or sdn-1 result in low penetrance embryonic developmental defects. Work from other systems has shown that syndecans can function as ligands for LAR receptors in vivo. We used double mutant analysis to test whether ptp-3 and sdn-1 function in a linear genetic pathway during C. elegans embryogenesis. Results We found animals with LOF in both sdn-1 and ptp-3 exhibited a highly penetrant synthetic lethality (SynLet), with only a small percentage of animals surviving to adulthood. Lead (Pb) contamination in soil represents a threat to human health and cleanup efforts often requires expensive or environmentally disruptive techniques. Phytoextraction has gained attention as a possible alternative to traditional remediation methods because of lower cost and minimal soil disruption. Panicum virgatum L. Introduction: Abdominal obesity is associated with chronic diseases such as obesity and coronary artery disease (1). Abdominal (or visceral) fat is strongly correlated with an inactive and sedentary lifestyle. Recent research shows that pregnancy is associated with high levels of visceral fat beyond appropriate levels; however, no studies have linked them with physical inactivity. Given the sequence-specific DNA-binding properties of transcription factor proteins, it is possible to purify them, identify the responsible polypeptide(s), determine their consensus binding sequences, and identify their genomic binding sites. A novel phage, Φ241, specific for Escherichia coli O157:H7 was isolated from an industrial cucumber fermentation where both acidity (pH ≤ 3.7) and salinity (≥5% NaCl) were high. The phage belongs to the Myoviridae family. Its latent period was 15 min and average burst size was 53 phage particles per infected cell. The phage was able to lyse 48 E. coli O157:H7 strains, but none of the 18 non-O157 strains (including E. coli O104:H7) or the 2 O antigen-negative mutants of O157:H7 strain, 43895Δper (also lacking H7 antigen) and F12 (still expressing H7 antigen). A popular children's arts and crafts toy made international headlines when it was discovered that 1,4-butanediol had been substituted into some of its formulations. The chemical 1,4-butanediol rapidly converts to the date-rape drug g-hydroxybutyric acid (GHB) upon ingestion and resulted in the accidental poisoning of some children. In this experiment, students extract 1,4-butanediol from commercially available toy beads (or prepared simulations) using their knowledge of pharmaceutical chemistry, solubility, etc. Using case studies available in the literature, students develop a quantitative analysis experiment for 1,4-butanediol extracted from the toy beads and, through approximation, relate the extractable amount of 1,4-butanediol to dose-response …
