Sepsis is a life-threatening condition characterized by overwhelming inflammation, resulting in organ system damage, leading to a high mortality rate. Care in the clinical setting is supportive, and there are no approved sepsis-specific treatments. In septic mice, activation of the cholinergic anti-inflammatory pathway decreases cytokine secretion by leukocytes and improves survival. The cholinergic anti-inflammatory pathway is a reflex of the parasympathetic nervous system, converging on the α7 nicotinic acetylcholine (α7nAChR) at the surface of macrophages. Signaling through the receptor blocks NF-kB activation, thus cytokine secretion. Receptor activation has other effects on macrophages, including modulating their migration to …

Histological analysis of long bone thin sections is commonly used to infer growth rates and ecology of extinct vertebrates, particularly within Archosauria. However, most comparative neontological studies have used small samples of captive individuals, limiting the scope of variation. To fill this gap, 44 femoral thin sections of wild Alligator mississippiensis were prepared and analyzed. Comparison of slides revealed that larger individuals from cooler climates tend to show more LAGs compared to southerly A. mississippiensis of similar size, however, there is considerable variation. Reactive oxygen species (ROS) such as H2O2 can damage cellular components and are formed as a byproduct of mitochondrial oxidative metabolism. Studies using the nematode C. elegans have found that increasing ROS during development or early adulthood can extend lifespan, while increasing ROS during later adulthood normally decreases lifespan. NADPH provides the reducing power for several cellular antioxidants and is synthesized in a two-step reaction from NAD+ with the first step being catalyzed by NAD kinase (NADK). The hyoid apparatus in laryngeally echolocating bats forms a mechanical connection between the larynx and auditory bullae and has been hypothesized to transfer the outgoing echolocation call to the middle ear during echolocation call emission. We used µCT data to build models of the hyoid apparatus and middle ear from six species of bats and used finite element modeling (FEM) to measure the vibroacoustic response of the tympanic membrane due to hyoid-borne sound generated during echolocation. We found that hyoid-borne sound in all six species stimulated the eardrum within a range likely heard by bats. Although there were minor differences …

The southern Appalachian Mountains have among the highest salamander diversity in the world, largely due to local speciation in the family Plethodontidae. Plethodontid salamanders (i.e., lungless salamanders) are particularly sensitive to habitat climate conditions due to their reliance on cutaneous respiration, and their immediate environmental conditions (microhabitat) likely influence their dispersion and activity more than the large-scale atmospheric conditions. Alcohol use disorder (AUD) costs the U.S. billions of dollars each year and is a leading cause of preventable death. AUD leads to many health complications, and those who suffer from AUD will often have stress and anxiety disorder comorbidities. To better understand this connection between AUD and stress and anxiety disorders, restraint stress (RS) and chronic-intermittent ethanol exposure (CIE) procedures were used on rats to analyze neuroinflammation via ELISA in the infralimbic and prelimbic cortices, and the bed nucleus of the stria terminalis (BNST). Previous work found significant differences in growth rates of native mussels at locations downstream from the regulated Walter’s Dam and the out-of-service, free-flowing Davy Crockett Dam. The purpose of this study is to investigate differences within the macroinvertebrate communities related to factors driving the differences in mussel growth between rivers. Macroinvertebrate samples were collected following the Tennessee Department of Environment and Conservation protocol for SQKICK collection and analyzed using the Tennessee Macroinvertebrate Index (TMI). European Paper Wasps (Polistes dominula) are social insects that build round, symmetrical nests. Current models indicate that these wasps develop colonies by following simple heuristics based on nest stimuli. Computer simulations can model wasp behavior to imitate natural nest building. This research investigated various building heuristics through a novel Markov-based simulation. The simulation used a hexagonal grid to build cells based on the building rule supplied to the agent. Nest data was compared with natural data and through visual inspection. Larger nests were found to be less compact for the rules simulated.

Fungal phytopathogens can cause disease epidemics in crops, weeds, and
populations of native plants. To investigate the impact of a foliar phytopathogen on the native herbaceous species, Veratrum viride, a demographic and disease assessment was carried out on two high elevation grassy bald populations on Roan Mountain, Tennessee. A leaf spot disease impacted all plants in both populations, causing widespread premature senescence of leaves and stems. Disease severity increased over the course of the growing season. Based on host disease symptoms and fungal conidia morphology, Pseudocercosporella sublineolata was shown to be the causal pathogen. A study of herbarium specimens …

Previous work demonstrated that WD in male ATM deficient mice induces accelerated body weight gain and heart dysfunction (Am J Physiol Heart Circ Physiol. 2021;320:H2324-H2338). The flexible shell of some oviparous reptiles has led to differences in nutrient mobilization and water relations when compared to their rigid-shelled counterparts. Flexible-shelled eggs gain more water during development, and because of the shell structure, yolk calcium content is higher than that of eggshells. When water availability is altered for reptiles with flexible-shelled eggs, differences in both energy and nutrient utilization are observed. Autism spectrum disorder (ASD) is a neurodevelopmental disorder marked by social deficits and repetitive actions. A communication pathway exists between the brain and gut called the gut-brain axis. It is thought that gut bacteria can secrete signaling molecules, triggering inflammation across the body. These studies attempt to determine if markers are expressed in two mouse models of ASD behaviors, BTBR and a valproic acid model. Immunohistochemistry of ionized calcium binding adaptor molecule 1 from male mouse brain tissue showed no microglial activation in any group. There are limited studies on bat activity in higher elevations in the Appalachian region. Levels of bat activity were compared in south central Appalachia at low (< 914.4 m) and high (> 1, 524 m) elevations in open, forest edge, and riparian habitats. Additionally, habitat suitability was modeled for a common species, big brown bat (Eptesicus fuscus). The study started May 27th 2019, and sites were monitored biweekly until October 2nd 2019. Autism spectrum disorder (ASD) is a neurodevelopmental disorder affecting social behaviors. ASD affects 1 in 59 children with males affected more frequently. ASD is postulated to result from excitatory and inhibitory neurotransmission imbalances. Brain-derived neurotrophic factor (BDNF) signaling affects ASD by influencing synaptogenesis, plasticity, and survival. Studying early in-utero neuropathological changes within ASD requires the use of animal models. Expression of BDNF-associated genes were analyzed within laser capture microdissected pyramidal neurons from the anterior cingulate cortex of male and female BTBR and valproic acid mouse models. Up to 87% of the world’s wetlands have been destroyed, considerably reducing ecosystem services these wetlands once provided. More recently, many wetlands are being restored in an attempt to regain their ecosystem service. This study seeks to determine the effects of restored wetlands on local bat habitat use. Bat activity was found to be significantly higher around the wetlands when compared to distant grassy fields; however, no significant difference was found among the restored wetlands and a remote cattle farm containing multiple water features. Apixaban is a novel oral anticoagulant that prevents clotting by directly inhibiting Factor Xa in the coagulation cascade. Due to its different pharmacokinetics, previous standards for testing anticoagulant concentrations are ineffective at measuring apixaban. In this study, Hyphen Biomed Biophen Direct Xa Inhibitor and Biophen Heparin chromogenic kits from Aniara Diagnostica were used along with a NanoDrop™ One/OneC Microvolume UV-Vis Spectrophotometer to see if either of these kits provide acceptable precision and accuracy for the quantification of apixaban in plasma samples, as well as if there is a significant difference in these two kits at varying concentrations of apixaban. …

Circadian clocks enable coordination of essential biological and metabolic processes in relation to the 24-hour light cycle. However, there are many habitats that are not subject to this light cycle, such as the deep sea, arctic regions, and cave systems. This study analyzes the circadian pattern of isolated populations of a subterranean spider, Meta ovalis from two Tennessee caves and five Indiana caves. The Pocket-book mussel, Lampsilis ovata, is a native freshwater bivalve species that is endemic to North America. The salinity tolerance of this species is of interest because anthropogenic salinization events and climate change factors threaten their natural freshwater habitats. Furthermore, the invasive freshwater bivalve species Corbicula fluminea has been shown to display significant salinity tolerance, which may lead to negative competitive interactions with native freshwater bivalve species if the salinization of freshwater habitats exceeds thresholds beyond which native species can effectively cope. Endocannabinoids were known to exist only among Animalia but recent report of their occurrence in early land plants prompted us to study its function and metabolism. In mammals, anandamide, as an endocannabinoid ligand, mediates several neurological and physiological processes, which are terminated by fatty acid amide hydrolase (FAAH). We identified nine orthologs of FAAH in the moss Physcomitrella patens (PpFAAH1 to PpFAAH9) with amidase signature and catalytic triad. The optimal amidase activity for PpFAAH1 was at 37 °C and pH 8.0, with higher specificity to anandamide. This study examines locomotor activity in samples of Frontinella pyramitela collected over its active season (April-October) to investigate whether seasonality and/or age may help explain the exceptional variability typically found in spider clock systems. Despite its noteworthy variability (%CV= 7.7), we have found that Frontinella has a mean free-running period of 28.4±2.18 hours that does not significantly vary over time. There is no correlation between day length and free-running period, indicating that varying length of FRP is not a function of photoperiod length. The clearance of viruses is largely dependent upon the activation of T cells to generate a robust immune response. However, host responses are suppressed during chronic viral infections. In this thesis, we explored the role of Top2α in DNA topology in individuals with chronic HBV, HCV, and HIV infections. We found that Top2α protein expression and activity were low in T cells derived from chronically virus-infected individuals compared to healthy subjects. Urbanization is increasing at a dramatic rate as the human population increases. While it is well-known that urbanization tends to decrease species diversity (i.e., biotic homogenization), it is not known how urbanization affects the frequency and efficiency of species interactions. Seed dispersal is a plant-animal interaction that depends on disperser feeding rate, disperser diversity, probability of seed dispersal and germination. Thermotolerance limit in aquatic organism is set by the ability to sustain aerobic scope to sudden temperature shifts. This study tested the genetic and plastic differences in thermotolerance of Daphnia that can be explained by the differences in the ability to retain mitochondrial integrity at high temperatures. Five genotypes with different biogeographic origins were acclimated to 18C and 25C. We developed a rhodamine 123 in-vivo assay to measure mitochondrial membrane potential and observed higher fluorescent in heat damaged tissues as the disruption of the mitochondrial membrane potential. Upon exposure to environmental stressors, amphibians such as Plethodon montanus will release corticosterone (CORT) thus causing a behavioral and physiological response to cope with the stress. Currently, there are several invasive ways of collecting CORT in salamanders. However, these techniques typically require euthanasia of the organism. We hypothesized that exposure of P. montanus to stressful handling conditions will result in elevations of CORT that can be detected through dermal swabbing.
