Signaling Through The Trail Receptor Dr5/Fadd Pathway Plays A Role In The Apoptosis Associated With Skeletal Myoblast Differentiation, J. O'Flaherty, Y. Mei, M. Freer, C. M. Weyman

Apoptosis rather than differentiation is a physiological process during myogenesis and muscle regeneration. When cultured myoblasts were induced to differentiate, we detected an increase in caspase 8 activity. Pharmacological inhibition of caspase 8 activity decreased apoptosis. Expression of a dominant-negative mutant of the adapter protein FADD also abrogated apoptosis, implicating a death ligand pathway. Treatment with TRAIL, but not Fas, induced apoptosis in these myoblasts. Accordingly, treatment with a soluble TRAIL decoy receptor or expression of a dominant-negative mutant of the TRAIL receptor DR5 abrogated apoptosis. alker Lake, a hydrologically closed, saline, and alkaline lake, is situated along the western margin of the Great Basin in Nevada of the western United States. Analyses of the magnetic susceptibility (χ), total inorganic carbon (TIC), and oxygen isotopic composition (δ18O) of carbonate sediments including ostracode shells (Limnocythere ceriotuberosa) from Walker Lake allow us to extend the sediment record of lake-level fluctuations back to 2700 years B.P. To demonstrate how insects may adapt to ecologically relevant levels of heat stress, we performed artificial selection on the ability of Drosophila melanogaster to fly after an exposure to a high but non-lethal thermal stress. Both tolerance and intolerance to heat stress arose very quickly, as only a few generations of selection were necessary to cause significant separation between high and low lines for heat tolerance. Estimates of heritability based on the lines artificially selected for increased flight ability ranged from 0.024 to 0.052, while estimates of heritability based on the lines selected for the inability to fly after heat …

Walker Lake, a hydrologically closed, saline, alkaline lake located along the western margin of the Great Basin of western United States, has experienced a 77% reduction in volume and commitment drop in lake level as a result of anthropogenic perturbations and climatic fluctuations over the last century. The history of lake-level change in Walker Lake has been recorded instrumentally since 1860. Reservoir storage reduces fluctuation in streamflow salinity, as inflow blends with stored water. This process is directly relevant to reservoir management, yet the methods for predicting outflow salinity from inflow and storage data are not adequately addressed. This study examined three simple methods for estimating outflow salinity on a monthly time step. The first method applies when inflow approximately equals outflow. The second method is applicable to changing reservoir storage. The third method utilizes a two-layer model in which evaporative concentration is assumed to occur in the top layer. The replication of damaged DNA is a promutagenic process that can lead to disease development. This report evaluates the dynamics of nucleotide incorporation opposite an abasic site, a commonly formed DNA lesion, by using two fluorescent nucleotide analogues, 2-aminopurine deoxyribose triphosphate (2-APTP) and 5-phenylindole deoxyribose triphosphate (5-PhITP). In both cases, the kinetics of incorporation were compared by using a 32 P-radiolabel extension assay versus a fluorescence-quenching assay. Although 2-APTP is efficiently incorporated opposite a templating nucleobase (thymine), the kinetics for incorporation opposite an abasic site are significantly slower. Lon protease, also known as protease La, is one of the simplest ATP-dependent proteases that plays vital roles in maintaining cellular functions by selectively eliminating misfolded, damaged and certain short-lived regulatory proteins. Although Lon is a homo-oligomer, each subunit of Lon contains both an ATPase and a protease active site. This relatively simple architecture compared to other hetero-oligomeric ATP-dependent proteases such as the proteasome makes Lon a useful paradigm for studying the mechanism of ATP-dependent proteolysis.
