Preeclampsia (PE), intrauterine growth restriction (IUGR) and gestational diabetes (GDM) increase the risk of maternal and fetal morbidity and mortality. The roles of Advanced Glycation End-products (AGEs) are already well documented concerning inflammation, hypoxia and oxidative stress. AGEs bind to its receptor, Receptor for Advanced Glycation End-products (RAGE), and activate an inflammatory pathway. This pathway alters the efficacy of invasive trophoblast cells and in the placenta and can result in placental dysfunction. We hypothesized that the placental dysfunction found in PE, IUGR, and GDM resulted from an over activation of the RAGE-mediated inflammatory pathway. Using human placental samples, we found …

Pregnancy necessitates interactions between the mother and the fetus, and the placenta is the medium through which this is accomplished. Many complications during pregnancy, such as intrauterine growth restriction (IUGR) and preeclampsia (PE) arise from abnormalities in trophoblast cells of the human placenta. Trophoblast cells form the connection between the fetus and the uterine wall. We see placental hyperosmolarity in pregnant patients with diabetes, and there are some reports of aberrant trophoblast functioning during IUGR and PE at term, but more needs to be studied regarding the hyperosmolar stress associated with these pathologies and its role in embryo development. Obstructive sleep apnea (OSA) is a chronic disorder involving repetitive interruptions in breathing during sleep. Sufferers of OSA are exposed to chronic intermittent hypoxia (CIH), characterized by cyclical reductions in oxygen availability. A number of studies have established a link between OSA and various cardiovascular and metabolic comorbidities in adulthood, including hypertension, obesity, and type II diabetes. While the consequences of OSA in adults have been well described, the cross-generational impact of this condition and potential effects on fetal development are not known. Chronic diseases such as type 2 diabetes and the Metabolic Syndrome create enormous burdens on society. Epidemiological studies now strongly implicate intrauterine growth restriction (IUGR) for increasing the risk of developing chronic diseases later on in life. However, the molecular mechanisms underlying how IUGR leads to the increased susceptibility to these metabolic diseases in adulthood is not well understood. The Liver-X-Receptor (LXR) is a nuclear receptor involved in cholesterol, glucose, and lipid metabolism. LXR acts to decrease gluconeogenesis through repression of glucose-6-phosphatase(G6Pase), phosphoenolpyruvate carboxykinase(PEPCK), and 11β-hydroxysteroid dehydrogenase type-1(11β-HSD1). Using the C2C12 mouse muscle myoblast cell line, I sought to investigate the hypothesis that differentiation under hypoxia impairs muscle mitochondrial respiratory function irreversibly. Resveratrol (RSV) will increase expression of markers of mitochondrial biogenesis and reverse the hypoxia-induced depression of O2 consumption rates. Hypoxia decreased oxygen consumption rates after five days of differentiation and after two days of normoxic recovery. This coincided with a hypoxia-induced decrease in protein levels of the NDUFB8 subunit of complex I of the electron transport chain and decreases in mRNA levels of mitochondrial biogenesis transcription factors, both of which could contribute …
