The biosynthesis of individual branched- and normal-chain fatty acids during Bacillus thuringiensis spore germination and outgrowth was studied by comparing pulsed and continuous labeling of these fatty acids with [U- 14C ]acetate. The relative specific activity of each fatty acid varies with time as the cell progresses through outgrowth. However, fatty acid synthesis does occur in two distinct phases. Upon germination, acetate is incorporated only into the iso-isomers i-C13, i-C14, and i-C16; no normal or anteiso synthesis occurs. The regulation of fatty acid biosynthesis by compounds that are required for growth of Bacillus thuringiensis was investigated using an in vivo assay developed to measure fatty acid synthesis in germinating spores. A minimal glucoseammonium- salts medium does not support growth even though previous radiorespirometric studies have shown B. thuringiensis to possess intact tricarboxylic acid and Embden-Meyerhof-Parnas pathways. Abundant growth does occur, however, when this medium is supplemented with either glutamate, aspartate, citrate, thiosulfate, cystine, or ethylenediaminetetraacetic acid. Cells held under non-growing conditions incorporate acetate into fatty acids; fatty acid synthesis is stimulated by the compounds that permit growth. These …

The success of the fetus as a transplant has been attributed to an immune depression in the mother. It has not been shown whether this purported depression was due to fewer T-lymphocytes or a dysfunction of these cells. T cells were measured by the rosette technique using neuraminidase treated sheep red blood cells (SRBC). B cells were measured using a SRBC-hemolysin-guinea pig complement indicator system. The total leucocyte count in the pregnant group was significantly higher than the control group but this was not reflected in the absolute number of lymphocytes. The purpose of this study was to develop a diagnostic test and conduct a survey for the neonatal calf diarrhea virus (NCDV) and human infant reolike diarrhea virus. Two immunologic methods were developed in this investigation.
Immune electron microscopy (IEM) and the fluorescent viral precipitin test (FVPT) are the methods used to detect NCDV and the human virus. Both methods are based upon the principle that viral aggregates form when the virus is reacted with anti-NCDV antibody. The timing and kinetics of fatty acid synthesis are delineated for Bacillus thuringiensis spore germination and outgrowth by analyzing [U-14C ]acetate and [2-3H ]glycerol incorporation into chloroform-methanol-extractable and trichloroacetic acid-precipitable lipids. In addition to measurement of pulsed and continuous labeling of fatty acids, monitoring the incorporation of radioactive phenylalanine, thymidine, and uridine from the onset of germination through first cell division provides a profile of biochemical activities related to membrane differentiation and cellular development. In spite of many years of intensive investigation lupinosis remains a remarkably complex and baffling problem. Current research is throwing new light on the disease and it is now much better understood— although far from beaten.
Lupinosis research is co-ordinated by the Department of Agriculture's Lupinosis Committee, on which are represented all groups working on the disease. These include veterinary surgeons and pathologists, chemists, plant pathologists and animal husbandry specialists.
Chairman of the Committee is Dr M. R. Gardiner, Chief of the Department's Animal Division and a veteran of lupinosis research.

Selected recreational waters of Arkansas were sampled for pathogenic free-living limax amoebae. Water quality parameters were determined for correlation with amoebic population densities and species diversity. Cultural criteria and animal inoculation revealed no pathogenic strains. The possibility of introduction and/or induction of pathogenic amoebic strains by environmental factors requires further ecological investigations.

Summary. Four of the five subspecies of Geomys pinetis were karyotyped. All specimens examined had a diploid number of 42 and a fundamental number of 80. This karyotype was compared with the described karyotypes of other species of Geomys and was considered to be derived from an ancestoral form having a karyotype of about 70 acrocentric elements.

The fine structure of the haplosporidan mitotic apparatus is described from observations of plasmodial nuclei of Minchinia nelsoni, M. costalis, Minchinia sp., and Urosporidium crescens. The apparatus, which is the Kernstab of light-microscope studies, consists of a bundle of microtubules terminating in a spindle pole body (SPB) at each end of the bundle. A few microtubules extend from SPB to SPB, but most either extend from an SPB and terminate in the nucleoplasm or lie in the nucleoplasm, free of either SPB. The bundle lengthens during mitosis, increasing the SPB-to-SPB distance by a factor of 2 to 3 as …

It is now well known that lupinosis of sheep and cattle follows ingestion of lupin stubble colonised by the fungus Phomopsis rossiana. The fungus infects green lupins but persists on the stubble and under certain conditions apparently produces a toxin which causes the disease.
As Phomopsis is now widespread on lupins in Western Australia its ecology has been studied in detail as a basis for possible control measures.
This article outlines what is known so far of the disease and its control.

These fungi, commonly called "water molds", are of particular interest as many are potential parasites of fish, crustaceans, and other aquatic plant and animal life. Gross primary productivity, heterotrophic bacterial numbers, and net phytoplankton densities of seven ponds in Coles County, Illinois, were studied in relation to physical, chemical, and biological habitat variables (light intensity and duration, turbidity, water temperature, pH, dissolved oxygen, sulfur, nitrogen, phosphorus, production, bacteria, and phytoplankton). Ten observations were made for each pond (except where otherwise noted) from 17 June through 25 August 1974. Stepwise multiple linear regression analyses of the data were used in order to determine those environmental factors which were important in predicting (i.e., significantly correlated with) bacterial and phytoplankton densities, and production.
