A deletion mutant of the colicin E1-derived plasmid, pDMS6642, exhibited an approximately fourfold increase in copy number. We subsequently isolated hydroxylamine-induced mutants of that plasmid that had a further increase in copy number. Analysis of them suggests that the increased copy number of pDMS6642 is associated with transcriptional readthrough from a Tn3 transposon into the region of ColE1 containing information that influences plasmid replication. The hydroxylamine mutation in one copy number mutant appeared to increase the plasmid copy number by stimulating readthrough transcription from the Tn3 transposon into the ColE1 replication control region, whereas the other hydroxylamine mutation acts by …

Comparison And Correlation Of Four Methods Of Measuring Immunity To Human Cytomegalovirus, James A. Brandt

The complement fixation test (CF) is currently the test most often used to determine the presence of antibody to human Cytomegalovirus (CMV). Several more recent techniques have now been adapted for this purpose. A comparison of CMV antibody titers was made between the CF test, a commercially available enzyme-linked immunosorbent assay (ELISA), an indirect immunofluorescent technique (IFA), and a modified indirect hemagglutination test (IHA). Forty three serum samples were tested for antibodies to CMV by each of the above procedures. The ELISA, IFA, and IHA titers were in close agreement on all samples tested. Richet and Portier studied the immunization of dogs against attenuated toxic extracts of the sea anemone and showed that, when given secondary injections of sublethal amounts of native toxic extracts, the dogs experienced severe pruritus and insensitivity followed by death. They called this increased sensitivity to the toxins, anaphylaxis. Richet and Portier, and later Jacques and Schachter, and Uvnäs also studied the direct pruritic and histamine releasing effects of cnidarian (e.g. Physalia, Actinia, Cyanea), toxic extracts. Fractionation of fully sporulated cultures of Bacillus thuringiensis by density gradient centrifugation in NaBr produced two bands which were identified as poly- β -hydroxybutyrate. This technique generated high yields of membranebound and unbound granules of exceptional purity and degree of polymerization.

Several recent reports have established the biological and molecular genetic similarity between the endogenous AKV virus of strain AKR, and an N-ecotropic endogenous virus found in the genome of feral Japanese mice, Mus musculus molossinus. The similarities are so striking as to suggest a common origin of these viruses, which are present in some, but not all, inbred mouse strains. The virogenes of AKR mice may have been acquired by either: (a) common descent of AKR (and other AKV+ strains) from a common ancestor of AKR and molossinus animals, or (b) horizontal germ line infection of the AKR …

Previous studies have shown the planktonic organisms of various natural water bodies to be quite sensitive to the levels of solar ultraviolet radiation incident at the water surface in summer. It has been assumed that the majority of the phytoplankton are simply carried about in the mass of the water which they inhabit with little control of their position or sunlight exposure. Photosynthetic microorganisms must strike a delicate balance with regard to solar radiation; they must obtain enough visible light for adequate photosynthesis while avoiding an excessive exposure to the injurious solar UV-B radiation.

Organisms of the genus Naegleria differentiate from feeding, dividing amebae into temporary swimming flagellates when deprived of nutrients. Factors critical for enflagellation of the pathogenic species, N. fowleri, were delineated and optimal conditions established for evoking reproducible conversion of N.fowleri populations. N. fowleri enflagellation differed from that of N. gruberi in its dependence upon growth phase, population density during enflagellation and in the timing and extent of the conversion. N. fowleri amebae from stationary phase cultures grown at 37°C, washed free of medium and suspended in nutrient~free ameba saline, acquired flagella and converted to mature flagellates in a synchronous manner …

The effects of pH upon methane production from anaerobic digestion of dairy cattle manure were investigated. One liter digesters were maintained by daily adjustment at the following pH levels: 7.6, 7.0, 6.0, 5.5, and 5.0. After 33 weeks of incubation the working volume of the digesters was increased to 3 liters. Digesters were incubated on a rotary shaker at 37°C. Digesters were loaded at the desired volatile solids concentrations, without an inoculum, and maintained from day one by daily additions and withdrawals to achieve a 3 day retention time. Inhibition of reaerated activated sludge was assessed using reduction of dissolved oxygen uptake by the microbial population as a criterion. Oxygen uptake is essential to biodegradation of the organic components in the activated sludge process; therefore, a decrease in dissolved oxygen uptake is an indication of reduced efficiency in wastewater treatment.

The irihibitors tested were cadmium, copper, lead, mercury, silver, 2,4-Dichlorophenoxyacetic acid (2,4-D), Terbacil and Zectran. The IIC50, determined by extrapolation of plotted data, for each inhibitor in mg/l was Hg (20), Cu (50), Ag (43), Cd (115), Pb (550), and 2,4-D (430). The IIC50 was determined …
