Mercury concentrations and determinants of mercury accumulation were examined for ten finfish species from the lower Chesapeake Bay and its tributaries. None of the sampled fish had total mercury concentrations approaching the U.S. EPA human health screening value. Mercury concentrations in different fish species generally increased with increasing delta 15N, but not delta 13C, suggesting that trophic position, but not dietary carbon source was a dominant determinant. A methylmercury biomagnification model was built to estimate a food web magnification factor of approximately 10-fold increase per trophic level in Chesapeake Bay. Breathing in mammals depends on an inspiratory-related rhythm that is generated by glutamatergic neurons in the preBotzinger complex (preBotC), a specialized site of the lower brainstem. Rhythm-generating preBotC neurons are derived from a single lineage that expresses the transcription factor (TF) Dbx1, but the cellular mechanisms of rhythmogenesis remain incompletely understood. to elucidate these mechanisms we comparatively analyzed Dbx1-expressing neurons (Dbx1 +) and Dbxl- neurons in the preBotC in knock-in transgenic mice. Whole-cell recordings in rhythmically active newborn mouse slice preparations showed that Dbx1 + neurons activate earlier in the respiratory cycle and discharge greater magnitude inspiratory bursts compared to …

In vivo small animal imaging has become an essential technique for molecular biology studies. However, requirements of spatial resolution, sensitivity and image quality are quite challenging for the development of small-animal imaging systems. The capabilities of the system are also significant for carrying out small animal imaging in a wide range of biological studies. The goal of this dissertation is to develop a high-performance imaging system that can readily meet a wide range of requirements for a variety of small animal imaging applications. Breathing is essential for mammalian life. Although there is an emerging consensus that the inspiratory respiratory rhythm is generated in a lower brainstem region known as the preBotzinger Complex (preBotC), the mechanism of rhythmogenesis is still unclear. Additionally, the modulation of intrinsic currents within preBotC neurons has yet to be fully elucidated. This dissertation addresses both of these issues and relies on imaging, electrophysiological, and modeling techniques. The first chapter examines the size and composition of the preBotC. The diseases Dermo and MSX have devastated Chesapeake Bay populations of the eastern oyster, Crassostrea virginica. The protozoan Perkinsus marinus, which causes Dermo, is particularly problematic since it persists over a wide range of salinities and temperatures. An objective of this dissertation was to determine whether specific wild oyster stocks had developed natural resistance to Dermo and if several parameters (survival, growth, condition and energy reserves) were associated with resistance. Another objective was to characterize heat shock protein (hsp70) expression in the eastern oyster. Heat shock proteins such as hsp70 protect organisms from thermal stress and other stressors, and this …

Striped bass (Morone saxatilis) in Chesapeake Bay, USA, are experiencing an epizootic of mycobacteriosis. This disease, caused by bacteria in the genus Mycobacterium, causes granulomatous lesions of the skin and viscera. Diseased fish are often emaciated, and fish with skin lesions may be significantly disfigured. The overall goal of this work was to examine aspects of the pathobiology of mycobacteria in striped bass via laboratory exposure studies and cellular assays. Striped bass were injected intraperitoneally with a sublethal dose of Mycobacterium marinum, M. shottsii, or M. gordonae and sampled for histology and bacteriology at regular intervals to 45 weeks post-injection …

American oyster (Crassostrea virginica) stocks on the East Coast in general, and in Chesapeake Bay in particular, have been in decline over the past half-century in large part due to Dermo disease, caused by the protist parasite Perkinsus marinus. Efforts to restore oysters for their ecological and economic value have centered on the development and use of hatchery-based seed selectively bred for disease resistance. Selective breeding could benefit from the incorporation of oysters from wild stocks that have developed "natural Dermo resistance", but few such stocks have been identified and verified. Perkinsus marinus is an alveolate protozoan parasite of the eastern oyster (Crassostrea virginica) which is responsible for much of the decline in United States oyster populations. Perkinsus marinus can be cultured in vitro, but is rapidly attenuated in the process. Supplementation of a protein-free medium with oyster products altered proliferation, changed protease expression in the parasite extracellular products (ECP), induced morphological forms typically seen in vivo, and partially reversed parasite attenuation. Supplements derived from dissected oyster tissues were used to determine if these changes could be differentially elicited. These supplements, with the exception of adductor muscle, reduced proliferation. Whole oyster …

To investigate the potential of chemotherapeutants to control the oyster pathogen Perkinsus marinus, anticoccidial and antifungal compounds were tested in vitro on infected hemolymph and cultured P. marinus cells and in vivo on infected oysters. In addition, acute toxicity to oysters was determined for six anticoccidials. In vitro experiments with infected hemolymph consisted of 24 h exposure of 0.2 mL hemolymph aliquots to concentrations ranging from 100 mg/L to 0.01 mg/L of amphotericin-B, amprolium, arprinocid, cycloheximide, lasalocid, malachite green, monensin, sulfadimethoxine, and a potentiated sulfadimethoxine, followed by incubation in fluid thioglycollate medium (FTM) to determine prezoosporangia abundance. Cataract formation in the Elizabeth River (ER) has been reported since 1986. Previous studies have concentrated on describing cataract as a function of exposure to polluted sediments, especially polycyclic aromatic hydrocarbons (PAH). These studies, however, have not examined what processes might be involved in fish cataractogenesis. The overall objective of this research was to identify important processes associated with the early development of cataract in feral fish populations. Field studies were used to systematically examine cataract development. The disease caused by the protozoan Perkinsus marinus has been a major source of mortality in the eastern oyster, Crassostrea virginica. Variations in susceptibility to P. marinus infection among eastern oysters collected from the Chesapeake Bay and Gulf of Mexico, as well as between eastern and Pacific (Crassostrea gigas) oysters were determined. Since oyster host defense may play a role in determining susceptibility to pathogen infection, cellular and humoral defense activities of the oyster and their interactions with P. marinus were investigated.
