Staufen Negatively Modulates Microrna Activity In Caenorhabditis Elegans, Zhiji Ren, Isana Veksler-Lublinsky, David Morrissey, Victor Ambros

The double-stranded RNA-binding protein Staufen has been implicated in various post-transcriptional gene regulatory processes. Here we demonstrate that the Caenorhabditis elegans homolog of Staufen, STAU-1, functionally interacts with microRNAs. Loss-of-function mutations of stau-1 significantly suppress phenotypes of let-7 family microRNA mutants, a hypomorphic allele of dicer and a lsy-6 microRNA partial loss-of-function mutant. Furthermore, STAU-1 modulates the activity of lin-14, a target of lin-4 and let-7 family microRNAs, and this modulation is abolished when the 3' untranslated region of lin-14 is removed. In this presentation, Schmitt and Doan examined data from student projects completed in their HONR 1000N Genetics and Genealogy course, taught during winter 2010. Students traced their deep ancestry through mitochondrial and Y-chromosome DNA analysis, as well as more recent ancestry, using genealogical databases and family histories. Frequently, students were challenged or pleasantly surprised by the information they gleaned. Genetic and genealogical research can provide clues to one’s identity and ethnic background, as well as potential medical conditions.

Background: Alzheimer's disease (AD) is a debilitating age related progressive neurodegenerative disorder characterized by the loss of cognition, and eventual death of the affected individual. One of the major causes of AD is the accumulation of Amyloid-beta 42 (Aβ42) polypeptides formed by the improper cleavage of amyloid precursor protein (APP) in the brain. These plaques disrupt normal cellular processes through oxidative stress and aberrant signaling resulting in the loss of synaptic activity and death of the neurons. Historically, attempts to elucidate evolutionary relationships among members of the genus Epinephelus (Teleostei: Serranidae), commonly known as groupers, have been hindered by the overwhelming number of species (98, sensu stricto), a pan global distribution, and the lack of morphological specializations traditionally used in ichthyological classification. To date, no comprehensive phylogenetic study, morphological or molecular, to evaluate the monophyly of this genus has been presented. In this study, previous hypotheses regarding the relationships among the American grouper species and the allied genera were evaluated by examination of mitochondrial DNA sequences of the 16S ribosomal DNA region.
