Insects have the ability to reflect the chemical environment that they have developed in. Thus, necrogenous insects can reflect the chemical environment of cadavers they have fed on. Forensic scientists can potentially utilize insect larvae as an alternate biological matrix to detect drugs in cadavers that are skeletonized, burned, or otherwise badly damaged. Analysis of insects using paper spray ionization mass spectrometry (PSI-MS), an analytical method requiring little to no sample preparation, could prove a rapid, cost-effective, and non-destructive alternative form of toxicological analysis. Juvenile Hormone Mediation In An Insect With Parental Care Behavior, Jessica M. Rodino

Juvenile hormone (JH) is a well-known catalyst for hormonal processes in insects. However, the role of JH in insects that exhibit parental behavior is unknown. We investigated the influence of JH on parental behavior in the burying beetle (Nicrophorus orbicollis). In the first experiment, we manipulated the JH production of females via the administration of varying doses of fluvastatin sodium immediately following oviposition. We found that with increasing fluvastatin dosage, the total mass of offspring and number of offspring decreased while at the same time less of the food source was consumed. These results suggest a link between …

Scorpions are well known, venomous arthropods (Class: Arachnida, Order: Scorpiones). Their venom is composed of neurotoxins, proteases and cytotoxic peptides which plays a major role in toxic components such as inflammatory, antimicrobial and hemolytic activity. Venom is key for the survival of scorpions as it is involved in defense against prey and in feeding on predators. Centruroides vittatus (bark scorpion) is commonly found in North America, with habitat ranges from Nebraska to southern Texas. The recent decline in native bee populations across the Midwest has been linked to the drastic decrease of prairie habitats and pollinator floral resources. Since the agricultural revolution, grassland areas have been removed and replaced with row crops to meet the needs of expanding markets, including the production of ethanol. In response to the loss of these (and other) essential pollinators, the Conservation Reserve Program administered by the USDA initiated a pollinator habitat initiative called CP-42. The conversion of grassland habitats and intensification of row crop agriculture over the last several decades have driven declines in native pollinator populations throughout the Midwestern USA. Using stable isotope measurements of insect tissues to determine origin and migratory patterns is well established. However, isotopically determining nutritional origins of lipids, the primary fuel of migration, has not been as thoroughly researched. We explored isotopic links between diet and stored lipids in laboratory raised True armyworm moths (Mythimna unipuncta) using δ13C and δ2H measurements. Female Dolomedes scriptus, the striped fishing spider, serve as model organisms exhibiting pheromone production as a means of chemical signaling. Previous work has shown that male fishing spiders are responsive to female silk, and preferentially court in the presence of silk from mature virgins. In our research, we hypothesized that mature females produced larger quantities of silk than penultimate females in an attempt to elicit greater male mating response. Female spiders of both age statuses were kept for two days in a gridded tank. Sub-sampling of silk lines that crossed a random portion of the grid produced results signifying …

A Chemical In Filter Paper Interferes With Trail Following Behavior In The Termite Reticulitermes Hesperus (Banks), Kathryn Vest

Termites follow chemical trails. In previous experiments we dissolved and diluted chemicals in acetone to test if they elicited trail following. Each trail was a straight line across the arena drawn with 10 µl using a glass capillary (Microcaps). We noticed a minor but stable response to trails laid with pure acetone. The response could be caused by contaminated acetone or by a chemical in the filter paper (Watman #5) that lined our arena (petri dish, 10cm diameter). We could confirm that the termites respond to acetone alone. Soxkhlet extraction for 8 hours removed the signal from the filter paper. …

Cattle typically avoid consuming milkweed plants that contain high levels of toxic defense compounds, and therefore many people assume cattle avoid all milkweeds. However, common milkweed (Asclepias syriaca) contains only moderate levels of toxic compounds, and observations suggest that cattle regularly consume common milkweed and may even preferentially graze flowers and leaves of this species. These observations directly relate to efforts to add over one billion stems of milkweed to the central USA for monarch butterfly (Danaus plexippus) conservation. Monarch butterflies (Danaus plexippus) are suffering from declining populations due to habitat loss. To help offset this habitat loss conservationists have encouraged planting milkweed gardens to increase the availability of host plants for larvae. A tachinid fly (Lespesia archippivora) that causes mortality parasitizes monarchs. Understanding the influence these gardens can have on parasitism rates and monarch recruitment is vital to evaluating their effectiveness. I am quantifying egg/larval abundance and demography at conservation areas and residential gardens. Additionally, I am collecting and rearing monarch larvae to compare parasitism rates in both areas. Environmental challenges shape the evolution of animal behavior and morphology. For wingless terrestrial invertebrates like ants, pools of water on the forest floor are particularly dangerous. Here we show that ants can overcome this obstacle using a modified gait to transverse the water surface. The Emerald Ash Borer (EAB) Agrilus planipennis Fairmaire (Copleoptera: Buprestidae) is a destructive colonizer of ash trees that arrived in the United States in 2002. Since then, EAB has been detected in 22 states and two Canadian provinces, and has proven to be a complicated management issue for many cities including Rock Island. We are developing a prototype entomological data-collection application called "iFly," which runs on a field-capable iPad device. In this phase, we tackled refining screens and introducing a database manager to streamline operations as info is entered, stored, retrieved and delivered. We used SQLite3 database in Apple's Xcode Integrated Development Environment (IDE). Xcode gives mixed programming results. Apple's iOS environment ensures functional and fairly error-free apps can be built. But the sophisticated Xcode IDE requires specialist developers and valuable project time is spent as new programmers learn key techniques. Minimum postmortem interval (mPMI) estimations are critical to solving cases of equivocal death and the accuracy of these predictions can depend on the understanding of necrophagous fly successional patterns. Temperature ranges between day and night in southern California can be as great as 30°C and may influence vital functioning of ectothermic organisms. Arachnids produce silks from a pair of spinnerets in their abdomen and rely on variance in protein composition to make different types of silks. Temperature may influence tensile strength of filaments. We tested the effects of temperature on tensile strength of dragline silk of five specimens of Araneus gemmoides (orb-weaver) which were collected from Malibu Creek State Park in the Santa Monica Mountains of southern California, under five temperature conditions. This research addresses some of the possible factors that affect male Acheta domesticus phonotactic responses to calls of males of the same species. The study examines cricket age and social interaction with females as possible factors affecting this behavior. Through logistic regression analysis, the study attempts to demonstrate a link between these factors and the responsiveness of the male crickets in the study. As the crickets aged, they responded to a wider range of syllable periods, which may imply that they follow a similar pattern of decreasing selectivity in their response with age as their female counterparts. National Cave and Karst Management Symposium 2013

A team developed the Cave Ecology Inventory and Monitoring Framework for National Park Service (NPS) units. It contains information for NPS cave managers across the United States to determine how to inventory and monitor cave ecology. Due to the wide geographical scope of NPS caves and their many different types, the document does not prescribe exact protocols. Instead, it provides guidance for what types of inventory and monitoring are possible, a framework for deciding how to prioritize inventory and monitoring activities, and references to specific protocols that are already in place at NPS cave parks.

Ipsdienol is an important pheromone component for pine engraver beetle, Ips pini. Ipsdienol is a ten carbon monoterpenoid secondary alcohol and ipsdienone is the corresponding ketone. We are characterizing the activity of recombinant IDOL DH produced in Sf9 (insect) cells. The enzyme has a high stereospecificity: (-) ipsdienol was found to be a substrate while (+)-ipsdienol was neither a substrate nor inhibitor. Closely related monoterpenoids, such as nerol, geraniol, and citral, were neither substrates nor inhibitors. Smaller compounds, such as 2-propanol, also failed to act as an inhibitor or substrate.
