As Americans are spending greater portions of their dollar on food consumed outside the home, the foodservice industry plays more of an integral part of daily existence compared to previous generations. Given the numerous annual foodborne illness outbreaks that threaten human lives while undermining confidence in the food supply, food safety is a pertinent issue for industry stakeholders, government regulators, and consumers. Food worker handwashing reduces the risk of foodborne illness transmission, yet compliance with this simple behavior is a complex problem. Histomoniasis, commonly known as blackhead disease, has increased in prevalence due to the regulatory ban of prophylactics and therapeutics within the past 30 years. The objective of this thesis was to evaluate the efficacy of selected dietary chemoprophylaxis candidates as well as an in vitro live-attenuated vaccine candidate Histomonas meleagridis for prevention of histomoniasis. Chapter one addresses deoxycholic acid and a biliogenic diet intended to endogenously increase production of this secondary bile acid. Deoxycholic acid was effective in vitro but failed to prevent histomoniasis when evaluated during the in vivo experimental disease trial with turkeys. The biliogenic diet did not …

Samples collected at five different large bird poultry processing facilities over a period of 7 months from prescald to post debone locations were enumerated for Enterobacteriaceae, Salmonella spp., and Campylobacter spp. and the results were used to create Quantitative Microbial Risk Analyses (QMRA) models for parts, ground, and mechanically separated chicken (MSC) products. Sensitivity analyses indicated the points in the process at which reductions would be most advantageous to the endpoint and simulation models were run to test reductions required to meet the current USDA performance standards.

Campylobacter jejuni is a prevalent infectious enteritis mainly foodborne from chickens. Despite of reducing C. jejuni food contamination dramatically decreases campylobacteriosis, few effective approaches are available for the bacterial reduction in chickens. The aim of this study was to use microbial metabolic product deoxycholic acid (DCA) to reduce C. jejuni chicken colonization. Broiler chicks were fed 0 or 1.5 g/kg DCA, lithocholic acid (LCA), or urodeoxycholic acid (UDCA) in diets or orally gavaged with cholic acid (CA, 1.5g/kg body weight). No one cause has been attributed to colony fluctuations due to the theory that multiple stressors interact with one another to impact colony health. Alpha-toxigenic Clostridium septicum (CS), the cause of turkey cellulitis, results in devastating mortality with high costs for the industry. Various vaccinations have been evaluated to prevent this disease with moderate success. Ability of a CS bacterin-toxoid, in conjunction with adjuvants such as aluminum hydroxide, mannoslyated chitosan, or a water-in-oil Seppic Montanide 71 R VG adjuvant (OE) to induce immunity was evaluated in a 7-week study (Experiment 1). Poults (20/group) were vaccinated day-of-hatch, boosted at 5 weeks-of-age and compared to unvaccinated controls. Antibody titers were determined by ELISA for all experiments. African wild dogs (Lycaon pictus) are endangered carnivores whose population is decreasing from habitat loss and fragmentation, interspecific competition, and disease. Survival rates are especially low in Kruger National Park (KNP), though it is unclear why. I estimated the abundance in KNP and survival rates over different time spans, six years and nine months, using public photographic survey data. In 2015, there were 298 (SE=12.1) individuals. Using a mark-recapture analysis in program R, I found that the survival rate between 2009-2015 was only 3.2%, and within the 9-month survey period (September 2014 – June 2015), monthly survival rates for the …

Formaldehyde has been used as a disinfectant in poultry hatching cabinets to aid in controlling key pathogenic organisms, such as Salmonella and Escherichia coli (E.coli). There is some evidence that prolonged exposure of chicks to formaldehyde can reduce tracheal ciliary function and thus reasons to believe that exposure to formaldehyde, in the absence of pathogen challenge, may reduce chick vitality. It has been found that elevated temperatures during incubation may adversely affect body weights of broiler chicks, as well as post-hatch environmental heat stress on performance in poultry. Histone-like nucleoid structuring proteins (HNS) play significant roles in shaping the chromosomal DNA, regulation of transcriptional networks in microbes, as well as bacterial responses to environmental changes such as temperature fluctuations. In this work, the intracellular organization of HNS proteins in E. coli bacteria was investigated utilizing super-resolution fluorescence microscopy, which surpasses conventional microscopy by 10–20 fold in spatial resolution. Highly Pathogenic Avian Influenza (HPAI) presents a substantial economic risk to the poultry industry. Domesticated fowl contract HPAI initially through exposure, direct or indirect with migratory waterfowl and outbreaks can result in significant economic losses to growers and the poultry industry at large. A HPAI outbreak occurred in Minnesota and Iowa and spread across over 13 other states in 2014 and 2015. The objective of this study was to determine if using an intranasal zinc (Zn) solution would impact health and growth performance of high-risk stocker cattle. Male beef calves (n = 239; 3 arrival dates [block]; initial BW = 276 ± 2.4 kg) were stratified by arrival gender and BW and assigned to 1 of 2 treatments: 1) treated with 3 ml of a Zn solution containing 36.24 mg of Zn administered intranasally, or 2) control, in which calves were not treated. Poultry is one of the main sources of protein in the United States and in 2014 Americans ate approximately 100 lbs per person. However, consumption of poultry products is strongly associated with foodborne illness from Campylobacter. In the first study, chitosan, caprylic acid and their combination were evaluated as a coating treatment for the reduction of Campylobacter jejuni on poultry products. It has become a problem of worldly stature, especially when considering sheep nematodes. Recommendations have been modified from chemical-based control to multipart, managerial intervention. Certain pathogenic Escherichia coli known as Shiga toxin (Stx)-producing Escherichia coli (STEC) are commensals in cattle, and typically cause bloody diarrhea in humans once the Stx toxin is secreted in invaded intestinal epithelial cells. Infections with STEC cells can lead to hemolytic uremic syndrome, which is commonly associated with kidney failure. Several STEC serogroups have been declared adulterants in raw, non-intact ground meat, and future regulations could potentially lead to a higher number of STEC serogroup detection strategies for these pathogenic microorganisms. Parasite control is an important aspect of health management of horses, particularly the control of gastrointestinal (GI) parasites. Recently, treatment recommendations have involved selective use of anthelmintics in horses with fecal egg counts (FEC) greater than a specified threshold. Due to increasing restrictions regarding the use of AGP, precise and high throughput enteric inflammation models and markers to search for effective alternatives are urgently needed. Oral administration of fluorescein isothiocyanate dextran (FITC-d) and its passage into blood can be used as a marker for tight junction permeability. This thesis consists of four research parts: a literature review that covers Salmonella spp., one of the more prominent foodborne pathogens that represents a major risk to humans (chapter 1). Understanding the growth of Salmonella serovars and strains is an important basis for more in depth research. The safety of fresh produce is often challenged by the sporadic incidences of outbreaks owing to Salmonella Typhimurium (S.T.) and Escherichia coli O157:H7 (E.c.). Washing and current disinfection strategies do not completely decontaminate leafy greens such as spinach and iceberg lettuce. Both consumers and suppliers have been negatively affected by an increase in foodborne pathogens contaminating fruits. Consequently, there is a need for the development of more efficient antimicrobials and application techniques to decrease contamination. Natural preservatives such as organic acids and plant extracts have demonstrated promising results in decontaminating produce. Poultry is key in genetic research due to breeding feasibility, relatively short generation interval, and distinct phenotypes. It is estimated that 8% of broiler deaths annually can be attributed to ascites, an economically important disease that has been challenging the industry for the past 2 decades. Genetically selected ascites resistant (RES) and susceptible (SUS) chicken lines have been established and maintained by the University of Arkansas, Fayetteville.
