Micropropagation Of Grammatophyllum Multiflorum Var. 'Tigrinum' Lindley And Evaluation Of Its Antimicrobial Activities, Eufrocinio Marfori
The Philippine Agricultural Scientist
Grammatophyllum multiflorum var. ‘tigrinum’ Lindley is an endangered orchid species endemic to the Philippines. To ensure its sustainable utilization as a source of phytochemicals, the types and concentrations of cytokinin, and auxin for shoot and root induction using nodal explants from in vitro germinated seedlings for its mass propagation were established. Seeds were germinated in vitro using MS medium without growth regulator. The nodal explants from the in vitro germinated seedlings were placed in MS medium containing different cytokinins and auxins to optimize multiple shoots and root formation, respectively. Thidiazuron (TDZ) at 2.5 μM was found to be the best …

Aegle marmelos Corr. is better known for its medicinal values rather than its edible quality. Every part of the plant viz. leaves, fruits, roots, and the bark are used for treating various diseases related to heart, stomach, bacterial, fungal, viral, worms, fertility, fever and cosmetics. Marmelosin is one of the major compounds (coumarin), used to treat various diseases. Till date, there are no varieties developed for commercial orchards of Bael and hence, regarded as one among the underutilized fruits of India. Therefore, there is an urgent need for breeding new and cultivable varieties in Aegle marmelos. But the problem for …

Bael (Aegle marmelos Corr.) is an important medicinal fruit tree. The fruit pulp contains marmelosin, which is a laxative, diuretic, is being used in many patented drugs in India. Micropropagation technology can be gainfully employed in mass multiplication of improved bael varieties. We have developed micropropagation protocol of bael through shoot bud culture. It was imperative to test genetic fidelity of micropropagated plants using molecular markers (RAPD, DAMD and ISSR).

The present study describes the plant regeneration via somatic embryogenesis in suspension culture derived from the leaf and stem explants of Phyla nodiflora. The medium type, plant growth regulators, complex extract (coconut milk and malt extract) and anti-oxidant (activated charcoal, ascorbic acid, Polyvinylpyrrolidone and citric acid) markedly influenced the embryo regeneration of P. nodiflora. MS with 2,4-D and activated charcoal (10 mg/L) gave the highest stimulation of embryogenic callus growth. Optimized callus was transfered into suspension culture, which showed the globular, heart shaped embryos in MS with 2,4-D + BA + picloram (0.1 mg/L), coconut milk (10 ml/L), citric acid …

Rapid clonal micropropagation protocol of Aegle marmelos (L.) Corr. cv. CISH-B1 was achieved by nodal stem segment of mature bearing tree. Three centimeter long shoots having one axillary bud excised from 10-15th nodal region of shoots during September gave quick in vitro bud burst (5.33 days) when cultured on MS medium supplemented with BAP, 8.84 μM + IAA 5.7 μM. The maximum number of proliferated shoots (9.0/explant) were obtained on same medium supplemented with BAP 8.84 μM + IAA 5.7 μM. The micro shoots were rooted (100 %) on ½ strength MS medium supplemented with IBA 49.0 + IAA 5.7 …
