With an increasing human population, there has been increased production of fish to meet nutritional needs. Commercial aquaculture accounts for a significant portion of seafood production with salmonids being the major farmed finfish in Canada. To obtain greater biomass from aquaculture with minimal drawbacks (e.g., compromised flesh quality), triploidization has been implemented, altering ploidy from 2N to 3N, to induce sterility and promote energy investment towards somatic growth. Triploid individuals experience transcriptional and behavioural changes resulting in disease, mortalities, and reduced growth. Triploid salmon – fish with three sets of chromosomes – are regularly used in aquaculture as they are functionally sterile and therefore avoid the decline in flesh quality associated with sexual maturation. However, triploid fish exhibit many immune, behavioural, and physiological drawbacks which reduce their condition and survival in aquaculture environments. My research examines the effects of probiotic treatment to promote behavioural and stress-related (termed ‘coping styles’) traits more suited for aquaculture environments in triploid fish with the ultimate goal of increasing fish yield. The microbiome is an important determinant of plant health, growth and resistance to stress. This study was conducted to determine the efficacy of the phyllosphere-microbiome on managing Pseudomonas syringae pv. Glycinea (Psg), gas exchange and growth of Glycine max L. Merrill. A greenhouse study was conducted in the summer of 2019. A field isolated and cultured soybean phyllosphere-microbiome was applied to purposely infected and uninfected soybean. Gas exchange, Psg density, chlorophyll concentrations, and percent nitrogen and carbon in the leaves were measured during the V3-V6 stages of soybean. The prevalence of non-alcoholic fatty liver disease (NAFLD) in children has increased over the past decades, creating a need for animal models that recapitulate the features of the pediatric disease. Iberian pigs have a leptin-resistant phenotype characterized by hyperleptinemia, hyperphagia, and extreme adipogenesis. We hypothesized that neonatal Iberian pigs fed a high fat high-fructose (HFF) diet will develop a pattern of liver injury resembling pediatric NAFLD. Bacterial pathogens have developed exquisite virulence mechanisms to survive in the host cells. These virulence mechanisms help them bind and internalize into host cells, replicate, and evade the host immune response. The ability of probiotics to adhere to other microorganisms and the intestinal epithelium is thought to play a major role in their protective functions. Research on combining pro- and prebiotics as synbiotics to enhance human and animal health has accelerated in the past 10 years, including many clinical trials that have assessed a diverse range of synbiotic formulations. In this review, we summarize these studies as well as the commercial applications of synbiotics that are available. In particular, we critically assess the claimed health benefits of synbiotic applications and the ecological and therapeutic factors to consider when designing synbiotics and discuss the implications of these concepts for future research in this field.

Antibiotic growth promoters that have been historically employed to control pathogens and increase the rate of animal development for human consumption are currently banned in many countries. Probiotics have been proposed as an alternative to control pathogenic bacteria. This study was performed to test the in vitro effects of a Lactobacillus rhamnosus isolate from “amabere amaruranu”, a traditional Kenyan cultured milk, on 3T3-L1 and Caco-2 cell lines. Cultures of fully mature 3T3-L1 adipocytes were treated with bacterial isolate cell extract (CE), filtered spent broth (FSB) from overnight bacterial culture, or with a PBS control. Expression levels of PPAR³1 and 2, C/EBP±, and ATGL proteins in 3T3-L1 cells were upregulated by FSB treatment. CE treatment did not affect protein expression levels. Expression of MTTP and SREBP-1c proteins in Caco-2 cells showed no change with either treatment. Optical density measurements …

The vaginal microbiota, in particular Lactobacillus species, play an important role in female health through modulation of immunity, countering pathogens and maintaining a pH below 4.7. We report the isolation and genome sequence of Lactobacillus pentosus strain KCA1 (formally known as L. plantarum) from the vagina of a healthy Nigerian woman. The genome was sequenced using Illumina GA II technology. The resulting 16,920,226 paired-end reads were assembled with the Velvet tool. Contigs were annotated using the RAST server, and manually curated. A comparative analysis with the available genomes of L. pentosus IG1 and L. plantarum WCFS1 showed that over 15% …

Symbiosis is defined as two species living together. This association between organisms is present at all taxonomic levels making it a ubiquitious phenomenon in ecology and evolution. I studied symbiosis among three species: a mutualistic bacterium, a parasitic fungus and an amphibian host. The first goal of my research was to examine how the mutualistic bacteria of amphibians’ skin are acquired. I demonstrated that a cutaneous mutualistic bacterial species, Janthinobacterium lividum, was transmitted environmentally, via soil, to the skin of an amphibian species, Plethodon cinereus. A continuous-flow porcine cecal bacterial culture has been used experimentally as treatment against enterotoxigenic Escherichia coli in weanling pigs. Periodically, the cultures must be started from frozen stock. Our results indicate that denaturing gradient gel electrophoresis can be applied as an indirect indication of culture similarity for each new batch generated from frozen stock.

Products containing probiotic bacteria are gaining popularity, increasing the importance of their accurate speciation. Unfortunately, studies have suggested that improper labeling of probiotic species is common in commercial products. Species identification of a bank of commercial probiotic strains was attempted using partial 16S rDNA sequencing, carbohydrate fermentation analysis, and cellular fatty acid methyl ester analysis. Results from partial 16S rDNA sequencing indicated discrepancies between species designations for 26 out of 58 strains tested, including two ATCC Lactobacillus strains. When considering only the commercial strains obtained directly from the manufacturers, 14 of 29 strains carried species designations different from those obtained …
