Phytophthora ramorum is the causal agent of Sudden Oak death (SOD), ramorum dieback and ramorum leaf blight which affect both forest environments and nurseries. This oomycete pathogen has had a huge economic impact on the nursery and lumber industry. Forests in California have experienced substantial mortality of oaks affecting the forest dynamics and diversity. Our research investigates four native species and two ornamental cultivars of plants, which belong to the genus Arctostaphylos (manzanita) and are considered to be new hosts for P. ramorum in the chaparral ecosystem of California. The presence of fungal species associated with herbaceous plants was monitored in coastal Marin County, California, USA. The research involved a combination of field sampling surveys and data collection using a stratified random design, pathogen identification through microbiological and molecular analysis, and descriptive analysis and ordination of results. A total of two years of repeated sampling (four times a year) was organized to allow for the detection of seasonal differences in pathogen presence on aerial plant parts. Cyclooxygenase-2 (COX-2) is an enzyme that helps catalyze the formation of prostaglandins, which promote inflammation, pain, and fever and maintain other normal physiological functions throughout the body. However, the overexpression of COX-2 has been found to play a role in various diseases including breast cancer. COX-2 exists as two major glycoforms— 72 kDa and 74 kDa— due to the glycosylation site Asn580 which is glycosylated 50% of the time. Past studies from our lab have shown that this glycosylation regulates COX-2 protein turnover in the cell3. Captive environments are maintained in hygienic ways that lack free-flowing microbes found in animals’ natural environments. As a result, captive animals often have depauperate host-associated microbial communities compared to conspecifics in the wild and may have increased disease susceptibility and reduced immune function. Eastern hellbenders (Cryptobranchus alleganiensis alleganiensis) have suffered precipitous population declines over the past few decades. To bolster populations, eastern hellbenders are reared in captivity before being translocated to the wild. However, the absence of natural microbial reservoirs within the captive environment diminishes the diversity of skin-associated bacteria on hellbender skin and may negatively influence their ability to …

Diabetes patients are at higher risk of contracting an age-related neurodegenerative disease such as Alzheimer’s disease (AD). However, the mechanisms which link these diseases are poorly understood. We hypothesize that glucose and elevated levels of the glycolysis by product advanced glycation end-products (AGEs), may be involved. AGEs accumulate with age and are elevated in both diabetic and AD patients. Diabetes is a metabolic disorder for which consumption of sugar-rich diets is a major risk factor and is central to etiology in the vast majority of cases.

Reactive a-dicarbonyls (a-DC’s), such as methylglyoxal (MGO), are unavoidable metabolites generated during glycolysis that accumulate with age and have been linked with chronic age-related metabolic diseases such as Diabetes Mellitus. Diabetes Mellitus is generally characterized by peripheral neuropathy and sustained hyperglycemia. Chronic hyperglycemia leads to an increase in glycolysis and a downstream increase in reactive a-DC’s. The human body has a natural method of detoxifying these a-DC’s. Glycolytic cells have enzymes which can detoxify a-DC’s, but if overwhelmed, a-DC’s can accumulate and react non-enzymatically with proteins, lipids and DNA to yield a group of molecules called advanced glycation end-products (AGEs). …

The pre-analytical phase contributes 60-70% of total error of the total testing process (TTP) (Plebani 2006). The pre-analytical phase can be further divided into two phases; the ‘pre-preanalytical’ and ‘preanalytical’ phases, which commonly includes tasks performed outside of the laboratory walls, and tasks perform within the laboratory’s walls and control, respectively. Hypophosphatemic rickets is a rare, renal phosphate wasting disorder that presents various skeletal deformities. Although there are specific clinical presentations and biochemical findings used to identify hypophosphatemic rickets, there are various underlying renal phosphate wasting disorders that can lead to hypophosphatemic rickets making diagnosis extremely difficult. A targeted literature review through electronic search engines (e.g., BMC, PubMed, Google Scholar) was conducted to: 1) consolidate and summarize currently available data regarding the various renal phosphate wasting disorders, 2) identify current obstacles of successful diagnosis and treatment, and 3) propose directions for future hypophosphatemic rickets research into new biomarkers.

von Willebrand Disease (vWD) is a bleeding disorder caused by a deficiency in von Willebrand Factor (vWF), a large glycoprotein that assists in coagulation. Specifically, large vWF multimers in the blood stream are key components in starting the coagulation cascade. vWF is cleaved by the metalloprotease ADAMTS13, regulating the multimers size, which hinders vWF’s ability to function properly. The three main types of vWD —Type I, II, and III— are not well defined, and as a result are all similarly treated with plasma-derived vWF replacement therapy. Multicellular hosts maintain complex associations with microbial communities. While microbial communities often serve important functional roles for their hosts, our understanding of the local and regional processes that structure these communities remains limited. Metacommunity analyses provide a promising tool for investigating mechanisms shaping microbiome heterogeneity, which is essential for predicting functional variation between hosts. Using a metacommunity framework, we examined heterogeneity in the skin microbiome of the eastern hellbender (Cryptobranchus alleganiensis alleganiensis). An emerging technology known as three-dimensional (3D) tissue engineering has allowed scientists to mimic tissues found in vivo. Previous studies indicate that it is possible to differentiate dissociated mouse embryonic stem cells (mESCs) into 3D retinal tissues in vitro (Bertacchi, 2015; Eiraku, 2012). The newly differentiated retinal tissues are said to encompass all of the major components found in retinal tissues. Cystinuria is an autosomal recessive genetic disorder characterized by the defect of a renal transporter involved in cystine reabsorption. When this transporter is deficient, cystine cannot be broken down and reabsorbed by the body and is excreted via urine in high concentrations. The high levels of cystine present in the urine eventually lead to recurrent cystine urolithiasis due to its inability to solubilize. Despite having various forms of treatments such as thiol pharmaceutical therapies such as tiopronin and urine alkalinizing agents like potassium citrate, only few patients with cystinuria are able to successfully decrease cystine urine concentration. As with any complex biological pathway, the splicing process has both advantages and obstacles with respect to the diversity and fidelity of protein production. The potential benefits of being able to produce multiple versions of a gene (isoforms) must be weighed against the additional complexity introduced by the noisy and mechanistically complicated process of splicing. ERBB2-positive breast cancer is an aggressive disease form that can result in rapid tumor growth and metastasis driven by the overexpression of ERBB2 growth factor receptors present on the cell surface. Currently, approved agents can target and disable ERBB2 receptor function, but as few as 30% of patients with disseminated ERBB2-positive breast cancer will respond to these targeted therapeutics when given as single agents. Even then, many initial responders will soon develop resistance to these agents. Alzheimer’s disease (AD) is the most common form of dementia, affecting over 5 million Americans. In AD patients there is a buildup of amyloid plaque in the brain, which contributes to AD symptoms. The main component of plaques is Amyloid-β (Aβ), a 40-42 length peptide formed from Amyloid-β Precursor Protein (APP) proteolysis. APP proteolysis can result in either anti-trophic or trophic peptides. Several APP polymorphisms exist that cause or protect against AD by altering APP proteolysis. In humans, the major predictor for gout and forming uric acid kidney stones is elevated uric acid in the serum and urine respectively. It is known that uric acid is an end metabolite of purine degradation in humans, but in other species from D. melanogaster to lower apes, uric acid can be further metabolized by urate oxidase into readily excreted allantoin. We hypothesize that if urate oxidase enzyme activity and dietary purine are both critical in uric acid kidney stone formation, then urate oxidase knockdown in combination with dietary purine supplementation will increase uric acid and induce uric acid kidney …

The Blood Brain Barrier (BBB) functions to control homeostasis within the central nervous system (CNS) via strict control over the passage of molecules into and out of the brain. Dietary Restriction (DR) is a robust intervention that is known to extend lifespan and increase spontaneous activity in multiple species. Whether activity increase plays a causal role in mediating the health protective benefits of DR remains unknown. To investigate this relationship, nutritional manipulations and laboratory selection for lifespan were simultaneously applied. Three physiological outputs were used for the screening and characterization of genes that may mediate the effects of DR: starvation resistance, spontaneous activity levels, and lifespan. Mucopolysaccharidoses is a family of metabolic disorders caused by the absence or irregular functioning of lysosomal enzymes required to degrade glycosaminoglycans. Mucopolysaccharidosis IVA (MPS IVA or Morquio A syndrome) is characterized by the functional loss of the enzyme N-­‐acetylgalactosamine-­‐6-­‐ sulfatase (GALNS). The absence of GALNS leads to the chronic accumulation of the glycosaminoglycans keratan sulfate (KS) and possibly chondroitin-­‐6 sulfate (C6S). KS accumulation occurs in the lysosomes of KS-­‐rich tissues such as cartilage, cornea, and heart valve as well as some specific cell types such as macrophages.
