Environmental sustainability and human health impact of pulses produced and consumed in the United States was assessed using life cycle assessment (LCA). The study included three objectives 1) to estimate environmental impact of current production and consumption practices in the United States using attributional LCA; 2) to estimate environmental and human health impact of iso-caloric diets containing varying amounts of pulses using Hybrid-LCA and Combined Nutritional and Environmental-LCA (CONE-LCA); and 3) to estimate environmental impact of increased demand for pulses using consequential LCA. Bioassays have been used extensively to assess various toxicity endpoints of drinking water disinfection byproducts (DBPs), but an emphasis on single compounds prevails. In this research, DBP mixtures were assessed using a cytotoxicity test with Chinese Hamster Ovary (CHO) cells and an Ames fluctuation test with Salmonella typhimurium TA-98 and TA-100 with and without S9 rat liver homogenate. Seven whole mixture DBP concentrates were formulated using reconstituted natural organic matter (NOM) extracts from the Upper Mississippi River using scaled disinfectant dosing protocols with free chlorine and monochloramine in the presence and absence of added bromide. Biofiltration without pre-ozonation has the capability to remove natural organic matter (NOM) fractions that serve as precursors of disinfection byproducts (DBPs), which include the four regulated trihalomethanes (THMs) and dichloroacetonitrile (DCAN). Rapid small-scale column tests (RSSCTs) and Pilot Plant filters operated at empty-bed contact times (EBCTs) of 4, 8, and 16 minutes were used to evaluate the performance of nutrient-amended (free ammonia and phosphorus) biofiltration for THM and DCAN precursor removal, as measured using formation potential (FP) tests. In this study, a chemiluminescence-based total N-nitrosamine (TONO) assay was adapted to include a solid-phase extraction (SPE) step to assess the role of biologically derived materials as N-nitrosamine precursors. Methanol was determined to be a suitable solvent for the SPE-TONO assay, with N-nitrosodimethylamine (NDMA) losses estimated to be 30% by SPE alone and an additional 15% from concentration of the methanol extracts by N2-gas blowdown. Three biofilm-derived materials - poly-N-acetylglucosamine (PNAG), Pseudomonas aeruginosa, and tryptophan - were individually chloraminated and dose-response relationships were observed with the SPE-TONO assay, indicating biofilm are potential N-nitrosamine precursors. Microbial communities resistant to common oxidants can cause concerns for water treatment plants (WTPs). If a bacterium is not fully oxidized during disinfection, these species can impede upon filtration processes or seed biofilms in the distribution system. In an effort to minimize disinfection by-products (DBP's) that result from the reaction of chlorine (Cl2) with natural organic matter, water treatment plants have the option to change their primary oxidant to chlorine dioxide (ClO2).
