Lignocellulose is composed of polysaccharides linked to lignin and other aromatic compounds, making the sugars not readily available to fermentation. This entails that biomass must go through the unit operations of pretreatment and enzyme hydrolysis. Pretreatment opens the structure to allow the enzymes to act on and hydrolyze cellulose and hemicellulose to glucose and/or xylose which in turn are fermented to ethanol. Concomitantly, the enzymes interact with soluble phenols and insoluble solids derived from lignin that inhibit hydrolysis. This leads to high enzyme loadings and higher production costs. Soluble phenols can be eliminated through washing. Insoluble lignin, however, demands another …

In urban centers today, vertical farming is becoming a popular alternative to conventional agriculture in an effort to increase local food production and improve urban food security by growing crops using hydroponic methods in controlled environment spaces. More specifically, one vertical method involves growing crops inside refurbished shipping containers, or a “farm-in-a-box” concept, which offers a flexible, mobile, and scalable means of year-round food production in a variety of climates. Corporations are prioritizing corporate social responsibility (CSR) activities by investing in and actively promoting their social practices. In the U.S. of the modern food supply chain creates a unique challenge for corporations to address concerns about social issues of consumers and non-consumers alike. This study is motivated by the need to better understand individuals’ perceptions of CSR as it pertains to the food supply chain. Shelled corn was collected from two sources - commercial grain elevators and corn harvested from Purdue University farms and then frozen until tested. Using a carbon dioxide (CO2) test kit manufactured by Woods End Laboratories, CO2 evolution measurements were conducted on shelled corn from both sources, after the samples were re-wetted to approximately 21% moisture content, placed in a 473 ml glass jar, and incubated at room temperature for 72 hours. The CO2 test kit uses a gel that changes color in response to a change in the level of CO2 in the surrounding air. …

Pectin is an important polymer used in the food industry as a thickening and gelling agent. Though pectin is ubiquitous in plants, chemical and structural differences among pectin molecules prevent most from being viable for industrial use. Enzymes found naturally in fruit cell walls during the ripening process impair many desirable attributes of fruit pectins, rendering them unsuitable for industrial applications. Pectin methylesterase (PME) is one such enzyme whose expression can be altered during ripening through the use of recombinant genetic engineering. Hermetic storage has been shown to be effective in controlling insect pests and maintaining grain quality of dry grains at 13% moisture and below. However, the feasibility and use of hermetic storage for grains at intermediate mid-moisture levels under the influence of the sub-Saharan African weather conditions is relatively unknown. Hermetic storage experiments were conducted on grade 1 "6297 and 6333 VT RIB" hybrid corn under controlled temperature conditions at 10 degrees centigrade and 25 degrees centigrade at target approximate moisture content levels of 11, 15, 18 and 21% wet basis for a total storage period of 6 months. Production of bio-ethanol from lignocellulose requires the efficient fermentation of glucose and xylose, even in the presence of inhibitors. The desired product, ethanol itself, will inhibit the fermentation. A further understanding of how ethanol affects the organism is critical to overcoming its inhibition.
This thesis evaluated the effect of ethanol on the cofermentation of glucose and xylose in two different cases. The first case had an unstructured model created for Saccharomyces cerevisiae 424A (LNH-ST), a genetically modified strain of yeast capable of cofermenting glucose and xylose. Factors of ion specificity and ionic strength (I~0-100) were studied in the electrostatic complex formation and protein particle formation by thermal treatment for a β-lactoglobulin and pectin system. ζ-potential showed β-lactoglobulin and pectin began to interact near pH 5.50 and interactions were strengthened with decrease in pH. Visible light turbidimetry and light scattering at 90° revealed a trend in critical pH transitions for electrostatic complex formation based on both the ionic strength and the anion of the salt species, while effects of the monovalent cation was insignificant. Estimation of nutrient load is critical for many applications in water quality management; however, infrequent data monitoring and measurement error could raise considerable uncertainty in the load estimations. The objectives of this study were to quantify the overall uncertainty in annual nitrate-N load estimates, and develop a statistical model to predict nitrate loads based on subsurface drainage characteristics in Indiana watersheds. Nitrate was selected as the study object because of the high loads common in Midwestern streams, and its important influence on hypoxia in the Gulf of Mexico.

A flailing knife shredder was designed and tested as an alternative to a hammermill for processing biomass. The machine was comprised of two cylinders with four rungs, each with five free-swinging blades. Each of the two cylinders had adjustable clearance hoods and variable speed drives. Energy usage, output characteristics and device capacities were compared with corn stover processed at moisture contents (MC) of 10%, 35% and 50% wet basis (WB). The hammermill produced a more uniform particle size distribution in comparison to the shredder. Volatile fatty acids (VFA) are among the most abundant organic compounds found in animal manure and act as important intermediates in the production of methane under anaerobic digestion (AD). However, VFA also contribute to odor emissions from manure.
