Assessment Of Morphology And Hemodynamics In Normal And Remodeled Microvasculature With Parameter Sensitivity Analysis, Shilpi Ghosh

The goal of this study was to compare the morphological and hemodynamic characteristics of normal and remodeled vascular networks in the mouse gracilis muscle. To this end, previously developed models to assess flows and vascular branching were used to assess the normal and remodeled microcirculatory networks. The analysis revealed that the average individual vessel flow rates for vessels of similar caliber and total volumetric flow rates in the networks do not change for vessels of the same caliber after remodeling. Connectivity changes and average diameters primarily change in the larger arterioles after remodeling. A few correlations could be made between …

Studies are currently being performed to determine the effects of bisphosphonate treatments on the structure and density of bone tissue. One of the pathways for gaining a better understanding of the effects of this and other treatments involves creating a computer simulation. Theory suggests that bone tissue structure and density are directly related to the manner in which the tissue is loaded. Remodeling is the process in which bone tissue is resorbed in areas of low stress distributions, and generated in areas of high stress distributions. Solutions of reagent grade salt .1, .01, .001, .0001, .00001M were run through a SPR to see if SPR had enough resolution to detect the formation of the electric double layer. We found that NaCl did in fact return a detectable signal. The signal shape seems to depend on concentration. While more study may need to be done to get definite information it looks like SPR can interrogate the double layer.

The application of high frequency ultrasound is the key to higher resolution intravascular ultrasound (IVUS) images. The need to further improve the IVUS spatial resolution may drive the transducer center frequency even higher than the current 40 MHz range. However, increasing the center frequency may be challenging as it leads to stronger scattering echoes from blood. The high level of blood scattering echoes may obscure the arterial lumen and make image interpretation difficult. Blood backscatter levels increase with transmission center frequency at a much greater rate compared to arterial tissue. This thesis successfully integrates laminate microfluidic devices with an analytic Surface Plasmon Resonance (SPR) instrument. Integration was accomplished at low-cost using materials such as polydimethylsiloxane (PDMS), Poly(methyl methacrylate) (PMMA), Tygon tubing, and a 3-way stopcock. The main components of this thesis are the design and fabrication of the low-cost, in-house fluidics that can integrate with upstream microfluidics and the validation of the in-house fluidics using the Biosensing Instruments BI-2000 SPR instrument. The study of blood vessel growth and remodeling is a complex endeavor. Hypoxia, the lack of oxygen in a tissue, is known to stimulate angiogenesis (the growth of new blood vessels), and have little effect on arteriogenesis (the enlargement of existing blood vessels). However, the role of hypoxia in vessel function is unknown, but may be determined using the results and methods developed in this experiment.

The most common injury to the rotator cuff is the partial or full thickness tear of the supraspinatus tendon, which attaches the humerus to the scapula on the superior side, due to an acute or chronic injury. Hip resurfacing arthroplasty has changed the treatment of end stage arthritis without severe deformity for young, active adults. Presently, there are varying clinical approaches to implant design selection and cementation techniques. The purpose of this project is to determine what amount of reamer-femoral component offset allows for the best cement penetration into the femoral head.

Rapid prototyped femoral component models were produced with reamer femoral component offsets of 0.0 mm, 0.5 mm, and 1.0 mm. The purpose of this study was to investigate the relationships between BMP antagonist expression and osteocyte density, lacunar densities and osteocyte viability in cortical bone. This costs an estimated 2.3 billion dollars of medical spending. The best treatment of gunshot wounds relies heavily on experience, but even with experience the unpredictable nature of ballistics can make treatment difficult.

The humerus of a racing horse Thoroughbred is highly susceptible to stress fractures at a characteristic location as a result of cyclic loading. The propensity of a Thoroughbred to exhibit humeral fracture has made equines useful models in the epidemiology of stress fractures. In this study, a racing Thoroughbred humerus was simulated during training using a 3D finite element model in conjunction with a bone remodeling algorithm. Collateral dependent hyperemia is reduced following chronic ischemia, but the contribution of individual vessel reactivity is unknown. Further, it is not known what aspect of the complex ischemic injury response impacts vascular reactivity. To determine the impact of ischemia on vascular reactivity, we measured functional vasodilation in the muscular branch artery following resection of the femoral artery proximal to the muscular branch. On day-14 after surgery the diameter of the muscular branch was measured using side-stream dark field (SDF) imaging intravital microscopy. At moderate intensity skeletal muscle contraction (1mA, 200µs, 8Hz, 90sec), functional vasodilation is reduced compared to the contralateral …
