According to National Breast Cancer Society, one in every eight women in United States is diagnosed with breast cancer in her lifetime. American Cancer Society recommends a semi-annual breast-cancer screening for every woman which can be heavily facilitated by the availability of low-cost, non-invasive diagnostic method with good sensitivity and penetration depth. Ultrasound (US) guided Diffuse Optical Tomography (US-guided DOT) has been explored as a breast-cancer diagnostic and screening tool over the past two decades. Photoacoustic computed tomography (PACT) is an emerging imaging modality that exploitsoptical contrast and ultrasonic detection principles to form images of the photoacousticallyinduced initial pressure distribution within tissue. The PACT reconstruction problemcorresponds to an inverse source problem in which the initial pressure distribution is recoveredfrom measurements of the radiated pressure wavefield. A major challenge in transcranialPACT brain imaging is compensation for aberrations in the measured data due to the presenceof the skull. Ultrasonic waves undergo absorption, scattering and longitudinal-to-shear wavemode conversion as they propagate through the skull. Edge illumination X-ray phase-contrast tomography (EIXPCT) is an emerging X-ray phasecontrast tomography technique for estimating the complex-valued X-ray refractive index distribution of an object with laboratory-based X-ray sources. Conventional image reconstruction approaches for EIXPCT require multiple images to be acquired at each tomographic view angle. This contributes to prolonged data-acquisition times and elevated radiation doses, which can hinder in vivo applications. Ultrasound computed tomography (USCT) and photoacoustic computed tomography (PACT) are two emerging imaging modalities that have a wide range of potential applications from pre-clinical small animal imaging to cancer screening in human subjects. USCT is typically employed to measure acoustic contrasts, including the speed of sound (SOS) distribution, while PACT typically measures optical contrasts or some related quantity such as the initial pressure distribution. Photoacoustic computed tomography(PACT), also known as optoacoustic tomography (OAT), is an emerging imaging technique that has developed rapidly in recent years. The combination of the high optical contrast and the high acoustic resolution of this hybrid imaging technique makes it a promising candidate for human breast imaging, where conventional imaging techniques including X-ray mammography, B-mode ultrasound, and MRI suffer from low contrast, low specificity for certain breast types, and additional risks related to ionizing radiation. Photoacoustic computed tomography (PACT), also known as optoacoustic or thermoacoustic tomography, is a rapidly emerging hybrid imaging modality that combines optical image contrast with ultrasound detection. The majority of currently available PACT image reconstruction algorithms are based on idealized imaging models that assume a lossless and acoustically homogeneous medium. However, in many applications of PACT these assumptions are violated and the induced photoacoustic (PA) wavefields are scattered and absorbed as they propagate to the receiving transducers. Many new promising X-ray-based biomedical imaging technologies have emerged over the last two decades. Five different novel X-ray based imaging technologies are discussed in this dissertation: differential phase-contrast tomography (DPCT), grating-based phase-contrast tomography (GB-PCT), spectral-CT (K-edge imaging), cone-beam computed tomography (CBCT), and in-line X-ray phase contrast (XPC) tomosynthesis. For each imaging modality, one or more specific problems prevent them being effectively or efficiently employed in clinical applications have been discussed.
