Traditional diagnostic imaging provides clinicians with anatomical information that guides both diagnosis and treatment planning; however, once a tumor has progressed enough to be visible, it has often reached an advanced stage. Molecular imaging techniques allow for real-time visualization of chemical and biological processes via imaging of specific biomarkers, which can facilitate detection of malignancies before they become visible. One biomarker of interest is blood oxygen saturation (SO2) due to its correlation with hypoxia, which is associated with increased tumor malignancy; some studies have also established SO2 as an independent biomarker of disease progression. Photoacoustic imaging (PAI) is an imaging modality which derives its contrast from differences in optical absorption coefficients of tissue through laser irradiation and acoustic detection. Therefore, PAI derives its signal directly from optical absorbers such as blood; the critical component in a hemorrhage. We discuss operation and processing methods of our Verasonics Vantage system. We then investigate thermal safety of PAI in mouse model for various laser pulse repetitions rates and illumination duration. Visualizing and characterizing atherosclerotic plaques is important in determining the vulnerability of a plaque to rupture. To evaluate rupture risk, several compositional factors should be evaluated, including inflammation, the presence and size of lipid pools, thickness of the fibrous cap, and calcification. Currently, a need exists for an imaging modality that can detect each of these factors in a safe, noninvasive manner with high resolution and contrast at clinically relevant depths. Photoacoustic imaging is a growing field that has the potential to improve plaque diagnosis.
